A novel organophosphate hydrolase gene designated mpd was selected from its Genomic Library prepared by shotgun cloning. The nucleotide sequence of the mpd gene was determined. The gene could be effectively expressed in Esherichia coli.

Standard methods were follwed for culture and preparation of the cosmids and P1 clones. Restriction mapping was performed by hybridization of T3 and T7 oligonucleotides to partial EcoRI/total NotI digests. A total of 43 cosmids and 4 P1 clones covering 420 kb in three contigs were isolated.

Conservation Genetics, 2008Co-Authors: Jinju Zhang, Qigang Ye, Hongwen HuangAbstract:Fourteen polymorphic microsatellite loci were isolated and characterized from an AC-enriched Genomic Library of Liriodendron chinense (Magnoliaceae). The average allele number of these microsatellites was 5.2 per locus, ranging from three to eight. The observed and expected heterozygosity at population level were 0.07–1.00 and 0.10–0.83, respectively. These sets of microsatellites will be useful for studies of population genetic structure of L. chinense and L. tulipifera as well as to estimate fine-scale gene flow rates.

Molecular Ecology Notes, 2006Co-Authors: Lei Chen, Donald H Les, Xiaohong Yao, Ming Kang, Hongwen HuangAbstract:Twenty-six microsatellites were isolated and characterized from an AAG-enriched Genomic Library of Vallisneria spinulosa , a dominant submerged macrophyte in the middlelower reaches of the Yangtze River. Microsatellite polymorphism was evaluated in a random population and 11 microsatellite loci were found polymorphic with 2–18 alleles per locus and observed heterozygosity ranging from 0.281 to 1.000. These polymorphic markers were expected to be valuable for population genetic and demographic analyses of V. spinulosa .
