    Abstract:

    Two types of infection with myxosporean parasites, which were different in cyst size, were found from the Gill of Rhinogobius sp. OR collected from the Nagara River, Gifu Prefecture, Japan. A myxosporean forming large-type cysts in the Gill filament was morphologically identified as Henneguya rhinogobii. Another myxosporean forming small-type cysts was found to be parasitic in the Gill Lamella. Average spore sizes of the latter myxosporean were 15.8 (range 14.2–17.8) μm in length, 5.3 (4.7–5.8) μm in width, 6.5 (5.9–7.6) μm in polar capsule length and 34.9 (25.3–42.9) μm in caudal appendage length, which were almost identical to those of H. rhinogobii. However, the two were distinguishable by the location in the Gill (intrafilamental or intraLamellar) and by the difference in the sequence of SSU rDNA (about 96% similarity). We propose the myxosporean forming intraLamellar small-type cysts as Henneguya pseudorhinogobii n. sp. and redescribe H. rhinogobii by morphological and molecular studies.

  • Diet-tissue isotopic fractionation of the Pacific oyster Crassostrea gigas

    Abstract:

    Juveniles of the Pacific oyster Crassostrea gigas were reared on a monospecific microalgal diet in order to quantify their 13C and 15N diet–tissue fractionations. The weights of these juveniles increased by up to 18-fold within 33 d. The juveniles reached isotopic equilibria with the diet, enabling calculations of fractionation values. The 13C fractionation for tissues containing lipids ranked as 0.6‰ (Gill Lamella) >0.3‰ (adductor muscle) >–0.2‰ (mantle lobe) >–0.9‰ (whole soft body) >–2.2‰ (midgut gland), while the 15N fractionation ranked as 8.7‰ (adductor muscle) >6.5‰ (mantle lobe) >5.4‰ (whole soft body) >5.2‰ (Gill Lamella) >2.3‰ (midgut gland). Removal of lipids shifted the diet-equilibrated δ13C and δ15N values in all tissues except the adductor muscle, with resultant increases in the 13C and 15N fractionation values. The expected annual mean δ13C value for the diet of the oyster in the field is –17.0‰, which is an intermediate value among the δ13C of coastal phytoplankton (–20.2‰), epilithon (–20.0‰), epipelon (–14.8‰), and seaweeds (–14.9‰), suggesting that the oyster feeds on a mixture of these micro- and macroalgae. The expected δ15N diet value is 3.1‰, which is more depleted than values for micro- and macroalgae, suggesting that the 15N fractionation in the field is smaller than that obtained from the feeding experiment.

    Abstract:

    Effect of PCBs on the skin and Gill development of olive flounder, Paralichthys olivaceus were investigated by histological methodology and morphometric data. The olive flounder were exposed to limit concentration of effulent of PCBs (3.0 ) for 60 days. Skin development can be classified into four stages: SSEL (simple squamous epithelial layer), MCA (mucous cell appearance) & CCA (club cell appearance) DLA (dermal layer appearance), and SEL (startification of epidermal layer) stages. Gill development had five stages: GFA (Gill filament appearance), IGFE (identification of early Gill filament epithelial cell) MCA (mucous cell appearance), PCA (pillar cell appearance), and FGL (formation of Gill Lamella) stages. The periods of structural completion of skin and Gill were 22-30 days and 23-30 days after hatching in the exposure group respectively. The process of development of skin and Gill of the exposure group was very similar to that of the control group. Therefore, PCBs (3 ) have no influence on the development of skin and Gill in the larval stage of olive flounder.
