Observational studies suggest that agents that lower testosterone are an effective treatment for paraphilia. Aim We report a case of hormonal treatment of paraphilia that was associated with side effects. Method A 35-year-old man with a paraphilia was treated with long-acting Gonadorelin. Results The desired result was reduced preoccupation with sexuality, but there were various side effects including a serious amount of bone loss. Conclusion We believe that more attention should be given to the adverse effects of long-term treatment with triptorelin. In our view the drug regime needs to be revised.     Avaliou-se a eficácia da dose de 0,10 ou de 0,25mg de Gonadorelina nas taxas de ovulação e de sincronização no protocolo Ovsynch e de 150mcg de D-cloprostenol ou 0,530mg de cloprostenol sódico na regressão do corpo lúteo (CL) de sete dias e de todos os CL. Foram utilizadas 136 vacas lactantes da raça Holandesa, com média de produção de leite de 23,75,8kg/dia, com 138,4±72,0 dias em lactação. As vacas foram distribuídas aleatoriamente em quatro grupos, de acordo com a dose de Gonadorelina e o tipo da prostaglandina. As taxas de ovulação e de sincronização foram de 52,9% e 80,9% para 0,10mg de Gonadorelina e de 57,4% e 80,9% para 0,25mg de Gonadorelina, respectivamente. A taxa de regressão do CL de sete dias foi de 97,1% para o D-cloprostenol e de 97,5% para o cloprostenol sódico. A taxa de prenhez não foi influenciada pelos tratamentos, mas foi influenciada pela taxa de ovulação à primeira aplicação de Gonadorelina, 16,0% vs. 6,6% para as vacas que ovularam e não ovularam, respectivamente. Conclui-se que 0,10mg de Gonadorelina foi eficiente e ambas prostaglandinas podem ser usadas em protocolos de sincronização da ovulação.The efficacy of 150mcg D-cloprostenol or 0.530mg cloprostenol sodium on the 7- day corpus luteum (CL) regression rate, and of two doses of Gonadorelin, 0.10mg vs. 0.25mg, on the ovulation and synchronization rates during the Ovsynch protocol were evaluated. Lactating Holstein cows (n=136), producing 23.75.8kg milk/d and 138.4±72.0 days in milk were randomly assigned to four groups, according to dose of Gonadorelin and prostaglandin type. No effect of the products and doses tested on conception rate were observed. The ovulation and synchronization rates using 0.10mg or 0.25mg of Gonadorelin were 52.9% and 80.9% vs. 57.4% and 80.9%, respectively. The CL regression rates in cows ovulating after the first GnRH using D-cloprostenol or cloprostenol sodium were 97.1% and 97.5%, respectively. The pregnancy rate was not affected by treatment, but by ovulation rate after the first injection of GnRH. The pregnancy rates were 16.0% and 6.6% for ovulated and non ovulated cows, respectively. The results suggest that 0.10mg dose of Gonadorelin was efficient and both prostaglandins can be used in synchronization of ovulation protocols

    Lactating Holstein cows (n=158), at 213±112 days in milking and averaging 26±9kg of milk per day, were randomly assigned to one of three treatment groups: control (CG, n=52, saline), GnRH (GG, n=55, 100g Gonadorelin), and hCG (HG, n=51, 2500IU) given five days after artificial insemination (AI). Rectal temperature was taken at the moment of AI and blood samples were collected five, seven, and 12 days after AI. Pregnancy was determined between 42 and 49 days after AI. Concentration of progesterone (P4) in serum (ng/ml, mean±SE) for CG, GG, and HG were, respectively, 2.7±0.4, 2.5±0.4, and 3.2±0.5 on day 5; 4.8±0.4, 4.2±0.4, and 5.7±0.5 on day 7; and 5.2±0.4, 6.9±0.4, and 8.5±0.5 on day 12 after AI. P4 concentration had proportional increase in serum between days 5 and 7 after AI (CG: 178%, GG: 168%, and HG: 178%), suggesting that the treatments did not induce a luteotropic effect on the existing corpus luteum (CL).     Optimized fixed-time embryo transfer (FTET) protocols for synchronization of recipients have the potential to improve the overall efficiency and profitability of embryo transfer (ET) programs. The objective of the present study was to evaluate the effect of dose of gonadotrophin-releasing hormone (GnRH) at initiation of a 5-day synchronization protocol for FTET. Holstein heifers (n = 2689) at two locations were synchronized using a 5-day CO-Synch protocol as follows: Day 0: CIDR inserted, Day 5: CIDR removed, prostaglandin (PG)F2α treatment (500 μg cloprostenol), Day 6: PGF2α treatment, Day 8: GnRH (100 μg of Gonadorelin). On Day 0, at the time of CIDR insertion, heifers were assigned in a completely randomised design to the following groups: Single (a single dose of GnRH; 100 μg of Gonadorelin), Double (200 μg of Gonadorelin) or No GnRH (control). All heifers received an Estrotect patch placed on Day 5 and evaluated for signs of oestrus on Day 8. At location A, heifers were evaluated by ultrasonography 5 days after GnRH to determine presence and size of corpus luteum (CL), whereas at location B presence and location of CL were determined by transrectal palpation at the time of transfer. Heifers with a CL received an embryo 7 ± 1 days after GnRH administration, and pregnancy was determined by ultrasonography 41 and 63 days after GnRH. Data were analysed by generalized linear mixed models. Oestrus expression was greater in heifers that received Single and Double GnRH than in the No GnRH group (P = 0.001). Similarly, utilisation rate (number transferred per number treated) was greater for heifers in the Single and Double GnRH group than for those in the No GnRH group (P = 0.02). Pregnancy data were analysed for a subset of recipients using data from Day 41 (n = 2267) and Day 63 (n = 2042). The analysis of fertility outcomes included as covariates the type of embryo (in vitro fresh or frozen and in vivo fresh or frozen), embryo stage, embryo quality, interval from GnRH to transfer, and oestrus expression. Pregnancies per embryo transfer (P/ET) at Days 41 and 63 were not different between treatment groups (P = 0.86), and there was no interaction between type of embryo and treatment (P > 0.15). Pregnancy loss between Days 41 and 63 was not different (P = 0.49) between treatments groups. In conclusion, the removal of the initial GnRH from a 5-day FTET protocol resulted in a slight but significant reduction in the utilisation rate and the percentage of heifers showing oestrus. However, there was no detrimental effect on fertility. As a result, the overall cost of the FTET program can be reduced by eliminating the need for the initial GnRH treatment without compromising fertility.     Previous research has shown that induction of an accessory corpus luteum (CL) by administration of gonadotrophin-releasing hormone (GnRH) on Day 5 increases circulating progesterone and reduces pregnancy loss between Days 33 and 60 in heifers receiving in vitro-produced (IVP) embryos. Therefore, the objective of the present study was to determine whether timing of induction of an accessory CL influenced pregnancy loss in IVP recipients. Holstein heifers (n = 1,658) were synchronized using a modified 5-day CIDR CO-Synch protocol. Briefly, heifers received an intravaginal progesterone device (CIDR) on Day −8, Day −3: CIDR removed and first prostaglandin F2α (PGF2α) treatment, Day −2: second PGF2α, and Day 0: GnRH (G1, 100 µg of Gonadorelin acetate). Oestrus expression was evaluated on Day 0 with the use of an oestrus detection device. Transfer of fresh IVP embryos was performed on Day 7 ± 1 after G1. At the time of transfer, heifers were randomly assigned, in a 2 × 2 factorial design, to receive GnRH (200 µg of Gonadorelin acetate) or remain as Control (untreated) at two different times: Day 7 (time of embryo transfer (ET)) or Day 21. Thus, the following groups were formed: Control-Control (n = 410); Control-GnRH (n = 409); GnRH-Control (n = 419); and GnRH-GnRH (n = 420). All heifers were evaluated by transrectal ultrasonography to determine number, size, and location of CL on Days 5, 14, 21, 28, 32, and 60 and pregnancy status on Days 28, 32, and 60. Data collected from each heifer included embryo stage and quality, oestrus expression, body condition score, number of transfers, and technician. Pregnancies per embryo transfer (P/ET) and pregnancy loss data were analysed by logistic regression (SAS 9.4). Ovulation to Day 7 treatment was greater (P   0.68) or Day 21 treatment (P > 0.18), nor a Day 7 × Day 21 treatment interaction (P > 0.48) on P/ET at Day 32 or 60 (Table 1). Treatment with GnRH on Day 7 or 21 did not alter pregnancy loss between Days 32 and 60 (P > 0.10). Heifers with an accessory CL present at Day 32 (11.7%; 41 of 350) had similar (P = 0.55) pregnancy loss compared with heifers with no accessory CL (14.4%; 54 of 375). The number of CL present on Day 32 did not affect (P = 0.23) pregnancy loss; however, heifers with 3 CL (4.9%; 3 of 61) tended (P < 0.10) to have reduced pregnancy loss compared with heifers with 1 CL (14.4%; 54 of 375) or 2 CL (13.2%; 38 of 289). In conclusion, treatment with GnRH on Day 7 or 21 induced an accessory CL; however, it did not affect P/ET on Days 32 or 60 and pregnancy loss.     Abstract This study evaluated effects of equine chorionic gonadotropin (eCG) on fertility of 679 crossbred ( Bos taurus x Bos indicus ) lactating grazing cows synchronized for fixed-time AI (FTAI). At a random day of the estrous cycle cows received an intravaginal progesterone (P4) implant, 2 mg estradiol benzoate (EB) and 100 μg Gonadorelin (D0-AM). On D7-AM, cows received 0.5 mg sodium cloprostenol and were randomly assigned into two treatments: eCG (n = 340; 400 IU eCG on D7), or Control (n = 339; no eCG). On D8-PM, P4 implants were removed and cows received 0.5 mg sodium cloprostenol and 1 mg EB. Insemination was performed on D10-AM. Pregnancy was diagnosed 30 and 60 d after AI. Treatment with eCG tended to increase pregnancy per AI (P/AI) compared to Control at 30 (37.8 vs. 30.2%; P = 0.06) and 60 (31.9 vs. 25.1%; P = 0.08) d.     Bu çalışmada, tohumlama ile birlikte, kas içi uygulanan GnRH'nın kan serumu progesteron seviyesine ve gebe kalma oranı üzerine etkilerini araştırmak amaçlanmıştır. Çalışmanın materyalini, yaşları 3-11 arasında değişen toplam 23 inek oluşturdu. Östrüsleri PGF2 alfa ile senkronize edilen hayvanlardan, östrüs günü başlanarak, siklus boyunca haftada 3 defa kan örnekleri alındı. Takip eden östrüste tüm hayvanlar tabii olarak tohumlandı ve 2 gruba ayrıldı. Tohumlamadan hemen sonra birinci gruba (uygulama grubu n=12) 100 mcg Gonadorelin, 2. gruba (kontrol grubu n=11) 2 ml izotonik NaCl solüsyonu kas içi olarak uygulandı. Uygulamalar sonrasında östrüs siklusu süresi kadar (21-22 gün) bir zaman süresince, haftada 3 defa kan örnekleri alınmasına devam edildi. Kan örneklerinin serumları çıkarıldı ve progesteron değerleri RIA ile tayin edildi. \ud Tohumlama sonrası östrüs siklusu süresinin, uygulama ve kontrol grubunda gebe kalmayan hayvanlarla benzerlik gösterdiği tespit edildi. Uygulama ve kontrol grubunda gebe kalan hayvanların tohumlama sonrası progesteron seviyelerinin karşılaştırılmasında, 10. gün progesteron seviyeleri arasındaki farkın önemli olduğu belirlendi. Uygulama ve kontrol grubunda gebe kalmayan hayvanların tohumlama sonrası progesteron seviyelerinin karşılaştırılmasında ise, 17 ve son gün progesteron değerleri arasındaki farkın önemli olduğu tespit edildi. Uygulama grubunu oluşturan 9 (%77.7) inekten 7'si ve 11 kontrol inekten 5'i (%45.4) gebe kaldı. \ud Sonuç olarak, tohumlama ile birlikte uygulanan 100 mcg Gonadorelin, gebe kalan ineklerde progesteron seviyelerini önemli derecede etkilememekle beraber, gebe kalan ineklerin sayısını artırdı. Bu sebeple, tohumlamayı takiben kas içi olarak 100 mcg Gonadorelin uygulamasının, mekanizması tam olarak ortaya konamamakla birlikte ineklerin gebe kalma oranının yükseltilmesi amacıyla kullanılmasının faydalı olabileceği sonucuna varıldı

Uygulamaların sonrasında östrüs siklusu süresi kadar (21-22 gün) bir zaman süresince, haftada 3 defa kan örnekleri alınmasına devam edildi. Kan örneklerinin soğutmalı santrifüjde serumları çıkarıldı ve daha sonra progesteron hormonu tayini yapılıncaya kadar derin dondurucuda muhafaza edildi. Kan serumundaki progesteron değerleri RIA ile tayin edildi.\ud Gonadorelin’den 100 mcg uygulanan inekler ile kontrol grubunu oluşturan ineklerin tohumlama öncesi ve sonrası progesteron konsantrasyonlarının karşılaştırılmasında, istatistiki olarak farkın önemli olmadığı tespit edildi. Tohumlama sonrası östrüs siklusu süresinin, uygulama ve kontrol grubunda gebe kalmayan hayvanlarla benzerlik gösterdiği tespit edildi. Uygulama grubunda, gebe kalmayan hayvanların tohumlama öncesi ve sonrası 17. gün ve ortalama progesteron seviyeleri arasında önemli bir farkın olduğu tespit edilirken, kontrol grubunda gebe kalmayan hayvanlarda, istatistiki olarak bir fark tespit edilemedi. Bu iki grubun tohumlama sonrası progesteron seviyelerinin karşılaştırılmasında ise, 17 ve son gün progesteron değerleri arasındaki farkın önemli olduğu belirlendi. Uygulama grubunda, gebe kalan hayvanların, tohumlama öncesi ve sonrası dönemde sadece son gün, kontrol grubunda, gebe kalan hayvanlarda ise, 19. gün progesteron değerlerindeki farkın önemli olduğu tespit edildi. Bu iki grubun tohumlama sonrası progesteron seviyelerinin karşılaştırılmasında ise, 10. gün ve ortalama progesteron seviyeleri arasındaki farkın önemli olduğu belirlendi. Uygulama grubunu oluşturan 9 inekter 7’si ve 11 kontrol inekten 5’i gebe kaldı.\ud Sonuç olarak, tohumlama ile birlikte uygulanan 100 mcg Gonadorelin, gebe kalan ineklerde progesteron seviyelerini önemli derecede etkilememekle beraber, gebe kalan ineklerin sayısını artırdı.
