  • Refrigeration or anti-theft? Food-caching behavior of Wolverines (Gulo Gulo) in Scandinavia

    Abstract:

    Food-caching animals can gain nutritional advantages by buffering seasonality in food availability, especially during times of scarcity. The wolverine (Gulo Gulo) is a facultative predator that occupies environments of low productivity. As an adaptation to fluctuating food availability, wolverines cache perishable food in snow, boulders, and bogs for short- and long-term storage. We studied caching behavior of 38 GPS-collared wolverines in four study areas in Scandinavia. By investigating clusters of GPS locations, we identified a total of 303 food caches from 17 male and 21 female wolverines. Wolverines cached food all year around, from both scavenging and predation events, and spaced their caches widely within their home range. Wolverines cached food items on average 1.1 km from the food source and made between 1 and 6 caches per source. Wolverines cached closer to the source when scavenging carcasses killed by other large carnivores; this might be a strategy to optimize food gain when under pressure of interspecific competition. When caching, wolverines selected for steep and rugged terrain in unproductive habitat types or in forest, indicating a preference for less-exposed sites that can provide cold storage and/or protection against pilferage. The observed year-round investment in caching by wolverines underlines the importance of food predictability for survival and reproductive success in this species. Increasing temperatures as a consequence of climate change may provide new challenges for wolverines by negatively affecting the preservation of cached food and by increasing competition from pilferers that benefit from a warmer climate. It is however still not fully understood which consequences this may have for the demography and behavior of the wolverine. Food caching is a behavioral strategy used by a wide range of animals to store food for future use. Choosing appropriate caching sites appears important for slowing down decomposition rates and minimizes competition. In this study, we demonstrate that the wolverine, an opportunistic predator and scavenger, utilizes available carrion to create caches all year around. By following wolverines with GPS collars, we registered that they carried food far away to cache it in secluded and cold places, which are often located on steep slopes or in forest. However, when scavenging other carnivores’ prey, they move food in shorter distances, possibly to be able to quickly return for more. The observed efficiency in wolverine caching behavior is likely vital for their survival and reproductive success in the harsh and highly seasonal environment in which they live.

    Capture and anesthesia with medetomidine-ketamine were evaluated in free-ranging wolverines (Gulo Gulo) immobilized for marking with radiocollars or intraperitoneal radio- transmitters in Norrbotten, Sweden, during early June 2004 and 2005. Twelve juvenile wolverines were captured by hand and injected with 0.1460.03 mg/kg (mean6SD) medetomidine and 7.562.0 mg/kg ketamine. Twelve adult wolverines were darted from a helicopter or the ground, or captured by hand. Adults received 0.3760.06 mg/kg medetomidine and 9.461.4 mg/kg ketamine. Arterial blood samples were collected between 15 min and 30 min and between 45 min and 60 min after drug administration and immediately analyzed for selected hematologic and plasma variables. Hyperthermia was recorded initially in one juvenile wolverine and 11 adults. Rectal temperature, heart rate, and lactate decreased significantly during anesthesia, whereas hemoglobin oxygen saturation, pH, partial pressure of arterial carbon dioxide, and base excess increased. Adult wolverines darted from a helicopter had a significantly higher rectal temperature, higher glucose and hematocrit values, and a lower heart rate than juveniles captured by hand. Impaired arterial oxygenation was evident in all wolverines. This study provides baseline data on physiologic variables in adult and juvenile wolverines captured with different methods and anesthetized with medetomidine-ketamine.

    Knowledge of the wolverine Gulo Gulo mating system is limited. In this study, we use 20 microsatellite loci for paternity testing in 145 wolverine offspring with known mothers. Samples were collected during > 10 years in two Scandinavian populations, mainly in connection with radio-telemetry studies and as part of long-term population monitoring. In total, 51% of the offspring were assigned a father. Our results demonstrate that the wolverine exhibits a polygamous mating system as some males were shown to produce offspring with more than one female in a single year. Females often reproduced with the same male in subsequent breeding years, but sometimes changed their partner, potentially as a consequence of a change in the territory-holding male in the area. In the majority of litters, siblings were unambiguously assigned the same father, indicating that multiple paternity is rare. Of 23 breeding pairs, for which telemetry data were available, 20 had overlapping home ranges, suggesting that pair formation generally is consistent with the territories held by wolverine males and females.

    L-Gulono-c-lactone oxidase (Gulo) catalyzes the last step of ascorbic acid biosynthesis, which occurs in the kidney of elasmobranchs. This study aimed to clone and sequence Gulonolactone oxidase (Gulo) from the kidney of the euryhaline freshwater stingray, Himantura signifer, and to determine the effects of acclimation from freshwater to brackish water (salinity 20) on its renal Gulo mRNA expression and Gulo activity. We also examined the effects of brackish water acclimation on concentrations of ascorbate, dehydroascorbate and ascorbate + dehydroascorbate in the kidney, brain and gill. The complete cDNA coding sequence of Gulo from the kidney of H. signifer contained 1323 bp coding for 440 amino acids. The expression of Gulo was kidney-specific, and renal Gulo expression decreased significantly by 67 % and 50 % in fish acclimated to brackish water for 1 day and 6 days, respectively. There was also a significant decrease in renal Gulo activity after 6 days of acclimation to brackish water. Hence, brackish water acclimation led to a decrease in the ascorbic acid synthetic capacity in the kidney of H. signifer. However, there were significant increases in concentrations of ascorbate and ascorbate + dehydroascorbate in the gills (after 1 or 6 days), and a significant increase in the concentration of ascorbate and a significant decrease in the concentration of dehydroascorbate in the brain (after 1 day) of fish acclimated to brackish water.     Gulo Puan adalah makanan tradisional dari kecamatan Pampangan yang keberadaannya mulai ditanyakan. Kesulitan untuk mengetahui keberadaan produk, menyebabkan terputusnya informasi kepada generasi Z. Kecamatan Pampangan sebagai sentra pembuatan Gulo Puan yang masih berjalan dan berkembang, belum memiliki penjenamaan visual yang mampu merepresentasikan keberadaan Gulo Puan di kecamatan Pampangan. Tidak adanya penjenamaan visual, mengakibatkan masyarakat kurang mengetahui keberadaan destinasi ini. Maka dari itu, sangat penting melakukan penjenamaan visual destinasi wisata gastronomi “Guwan” Gulo Puan di kecamatan Pampangan sebagai upaya untuk mengenalkan keberadaan Gulo Puan pada destinasi wisata gastronomi Gulo Puan di kecamatan Pampangan. Penelitian ini menggunakan metode kualitatif dengan pendekatan design thinking yang melalui lima tahapan, yaitu emphaty, define, ideate, prototype, dan test. Proses pengumpulan data dilakukan dengan wawancara, observasi, literasi, dokumentasi, dan kuesioner. Data yang diperoleh kemudian dianalisis hingga mendapatkan sintesis yang menjadi dasar melakukan perancangan. Solusi permasalahan yang ditemukan dari sintesis, yaitu dengan melakukan penjenamaan visual destinasi wisata gastronomi Gulo Puan di kecamatan Pampangan, dengan gagasan berupa “Jelajah Pampangan” yang mencitrakan sebagai destinasi petualangan edukatif untuk target sasaran. Kebaruan perancangan ini adalah konsep penjenamaan visual yang merepresentasikan destinasi wisata gastronomi di kecamatan Pampangan. Terdapat beberapa teori yang digunakan, diantaranya yaitu teori penjenamaan untuk strategi merancang penjenamaan visual, teori sosiologi desain untuk menentukan kebutuhan target sasaran, teori destinasi wisata untuk merancang destinasi wisata yang unik dan kreatif, dan teori desain komunikasi visual digunakan untuk merancang media komunikasi visual yang disampaikan kepada target sasaran. Perancangan ini menyumbangkan pengetahuan bagi ilmu desain komunikasi visual berupa, konsep perancangan yang mengkolaborasikan penjenamaan visual dengan destinasi wisata gastronomi, yang kemudian wajib untuk dilanjutkan sebagai upaya mengenalkan produk kebudayaan, khususnya Gulo Puan. Solusi permasalahan yang ditemukan dari sintesis, yaitu dengan melakukan penjenamaan visual destinasi wisata gastronomi Gulo Puan di kecamatan Pampangan, dengan gagasan berupa “Jelajah Pampangan” yang mencitrakan sebagai destinasi petualangan edukatif untuk target sasaran. Kebaruan perancangan ini adalah konsep penjenamaan visual yang merepresentasikan destinasi wisata gastronomi di kecamatan Pampangan. Terdapat beberapa teori yang digunakan, diantaranya yaitu teori penjenamaan untuk strategi merancang penjenamaan visual, teori sosiologi desain untuk menentukan kebutuhan target sasaran, teori destinasi wisata untuk merancang destinasi wisata yang unik dan kreatif, dan teori desain komunikasi visual digunakan untuk merancang media komunikasi visual yang disampaikan kepada target sasaran. Perancangan ini menyumbangkan pengetahuan bagi ilmu desain komunikasi visual berupa, konsep perancangan yang mengkolaborasikan penjenamaan visual dengan destinasi wisata gastronomi, yang kemudian wajib untuk dilanjutkan sebagai upaya mengenalkan produk kebudayaan, khususnya Gulo Puan

    L-Gulono-c-lactone oxidase (Gulo) catalyzes the last step of ascorbic acid biosynthesis, which occurs in the kidney of elasmobranchs. This study aimed to clone and sequence Gulonolactone oxidase (Gulo) from the kidney of the euryhaline freshwater stingray, Himantura signifer, and to determine the effects of acclimation from freshwater to brackish water (salinity 20) on its renal Gulo mRNA expression and Gulo activity. We also examined the effects of brackish water acclimation on concentrations of ascorbate, dehydroascorbate and ascorbate + dehydroascorbate in the kidney, brain and gill. The complete cDNA coding sequence of Gulo from the kidney of H. signifer contained 1323 bp coding for 440 amino acids. The expression of Gulo was kidney-specific, and renal Gulo expression decreased significantly by 67 % and 50 % in fish acclimated to brackish water for 1 day and 6 days, respectively. There was also a significant decrease in renal Gulo activity after 6 days of acclimation to brackish water. Hence, brackish water acclimation led to a decrease in the ascorbic acid synthetic capacity in the kidney of H. signifer. However, there were significant increases in concentrations of ascorbate and ascorbate + dehydroascorbate in the gills (after 1 or 6 days), and a significant increase in the concentration of ascorbate and a significant decrease in the concentration of dehydroascorbate in the brain (after 1 day) of fish acclimated to brackish water. Taken together, our results indicate that H. signifer might experience greater salinity-induced oxidative stress in freshwater than in brackish water, possibly related to its short history of freshwater invasion.     This study aimed to test the hypothesis that the brain of Protopterus annectens expressed l-Gulono-γ-lactone oxidase (Gulo/Gulo), the enzyme catalyzing the last step of ascorbate biosynthesis, and could maintain high concentrations of ascorbate during estivation. We cloned and sequenced Gulo from the kidney of P. annectens and performed quantitative PCR to determine its mRNA expression in kidney and brain. Gulo activity was assayed and its protein abundance was determined by Western blot using custom-made anti-Gulo antibody. Effects of estivation on concentrations of ascorbate and dehydroascorbate in the kidney and brain were also determined. Both brain and kidney, but not liver, of P. annectens expressed Gulo/Gulo. Desiccation induced P. annectens to estivate, and 6 mo of estivation led to drastic decreases in Gulo/Gulo expression and ascorbate concentration in the kidney.
