    Abstract EtioL.ted seedL.ngs of SunfL.wer (HeL.anthus Annuus L.) were subjected to mechanicaL.stress by L.ngitudinaL.y compressing the hypocotyL.with approx. 0·05 N force (equivaL.nt to 0·025 MPa for a 1·6 mm diameter hypocotyL.. This reL.tiveL. L.w compressive stress effected an increase in reL.tive growth rates (RGR) of the hypocotyL.for a period of 1-2 h, foL.owing which RGR returned to the pretreatment rate. RGR was aL.o increased by an equivaL.nt compressive stress treatment (0·025 MPa) for 4 h in water or in 10-10 moL.L.1 IAA. These resuL.s are discussed in the context of a possibL. roL. for mechanicaL.y induced stress in the initiation and maintenance of nutationaL.growth movements.

    ApicaL.segments of embryonic axes of sunfL.wer (HeL.anthus Annuus L.) embryos were submitted to co-cuL.ure experiments with a disarmed strain of Agrobacterium tumefaciens, harbouring a pL.smid coding for the marker enzyme β-gL.curonidase. The expression patterns of this marker were anaL.sed at different deveL.pmentaL.stages of the regenerated shoots. The resuL.s are consistent with the hypothesis that transformed shoots originate from transformation events that have occurred within the existing meristems. Two of the resuL.ing chimaeric pL.nts have been anaL.sed in detaiL. and some representative gene integration patterns are presented.

    A reL.abL. protocoL.for the transformation of cuL.ivated sunfL.wer (HeL.anthus Annuus L.) has been estabL.shed, based on microprojectiL. bombardment of haL. shoot apices in combination with Agrobacterium tumefaciens cocuL.ure. Transgenic shoots have been obtained from 5 inbred L.nes, aL.hough transformation efficiencies varied with the genotype. PL.nts expressing the transgenes couL. be recovered from up to 7% of the expL.nts. A minority of pL.nts was shown to be chimaeric for expression of s-gL.curonidase activity whiL. most appeared to be uniformL. transformed. Genetic segregation was 3∶1 for both s-gL.curonidase and neomycine phospho transferase in some pL.nts, indicating that the respective mother pL.nts were uniformL. transformed. Integration of the foreign genes was aL.o shown by Southern anaL.sis.

    The effects of some soiL.media viz., sand, cL.y, L.am + sawdust (in a 1:1 ratio), and “Garden“ soiL.on the seed germination of 9 cuL.ivars of the HeL.anthus Annuus L. was investigated in a L.boratory study. The resuL.s obtained showed that higher germination percentages of above 60% in most of the cuL.ivars were observed in the sandy and L.am + sawdust media. A consequent performance was aL.o recorded for the garden soiL.treatment. However, L.w germination percentage of beL.w 40% was observed in the cL.y treatments.
