    Abstract:

    Peptides from Sumbawa Horse Milk are active as antibacterial and antioxidant. Horse Milk has a higher content of vitamin C than cow or goat's Milk, and cream products that contain Milk can overcome dry skin. The Gel is one of the topical dosage forms suitable for acne medications and oily skin types. This study aimed to determine the effect of variations in Horse Milk content formulated in gel preparations with carbomer 1% base as gelling agents on physical characteristics (organoleptic, viscosity, pH) and antibacterial activity in Propionibacterium acnes. In this study, gel preparations were made with six different formulas on Horse Milk content, namely FI (2.5%), FII (5%), and FIII (10%), FIV (15%), FV (20%), and FVI (25%) with 1% carbomer base on each formula and well method were used for the antibacterial test. From the organoleptic test, it was found that in all of the gel formula had soft textured, thick white color, and distinctive aroma like Horse Milk. The viscosity test showed that was no significant difference, but the pH test showed a significant difference for each formula. In this study showed that all of the gel formulae did not have an inhibitory zone, which means that at each level there was no evidence of antibacterial activity in the P. acnes. So, it could be concluded that all gel formula had good gel characteristics but there was no inhibitory activity against the P. acnes.

    Abstract:

    Background: Acne is a disease that reduces the confidence of sufferers which is caused by bacteria Propionibacterium acnes. Sumbawa Horse Milk has the best antimicrobial activity due to the galactoferin compounds the Milk has. Objective: This research aimed to determine the effect of Horse Milk with concentration at 15%, 20%, and 25% in 1% carbomer gelling agent toward the physical characteristics of the gel preparation, and to determine the antibacterial activity against Propionibacterium acnes. Method: The method applied in this research was the experimental to compare 3 formulas with different Horse Milk concentration. Then researcher analyzes the physical and chemical characteristics and antibacterial activity. Results and Conclusions: All formulas have the same characteristics, which were thick white, soft texture, and typical Horse Milk scent. The viscosity of all formulas is 12700cPs ± 2861.82, 11000cPs ± 9050.41, and 9066cPs ± 9625.66. The results of the spreadability are 0.014 g /cm, 0.017 g /cm, and 0.034 g /cm. The pH results for all formulas are 5.55, 6.24, and 6.46. The results of the antibacterial activity test of Horse Milk gel do not have antibacterial activity against the Propionibacterium acnes bacteria which is indicated by the absence of inhibition zones around the wells.

Results and Conclusions: Based on the evaluation test, the result showed that all formulations had same organoleptic result, such as: soft texture, typical smell of Horse Milk, and Milky white color. In addition, evaluation testwere showing a result thatviscosity and pH had no significant difference in the formulation. Besides, the dispersive power test was resulting a significant difference. Lastly,in the antibacterial test, the result showed that there was no inhibition zone resulting in the formulation test.

    Background: Sumbawa Horse Milk contains a compound called galactoferrin which has a very good antimicrobial activity. Acne is a skin disease that often occurs at a young or old age. Although not a serious skin disease, but if not treated will cause depression and a crisis of confidence. One of the bacteria that causes acne is Propionibacterium acnes. Objective: This research conducted to determine the effect of Horse Milk (15%, 20%, and 25% concentration) on a 2% carbomer basis preparations gel toward the physical characteristics (pH, spreadability, viscosity) and on the activity of Propionibacterium acne bacteria. Methods: This research was an experimental research to compare 3 formulas with difference Horse Milk levels then analyze the physical and chemical characteristics of antibacterial activity. Results and Conclusions: Based on the physical properties test showed it has the same physical characteristics which are soft textured, has the smells typical of Horse Milk, Milky white. Based on the results of the test using the One-way Anova statistic test, the viscosity and spreadability did not have a significant difference in each formula with the value (sig> 0.05).     Background : Acne is inflammatory disease due to blockage in pilosebasea and triggered by bacteria propionibacterium acnes. Sumbawa Horse Milk is the best antibacterial activity good as it is tested with some bacteria test by comparing cow’s Milk. Objective : This research is aimed to investigate the usage comparison of Sumbawa Horse Milk in the level of 2,5%, 5% and 10% with a carbomer gel base 2% towards characteristic of physical, chemical, and antibacterial activity using Propionibacterium acnes bacteria (a bacteria causing acne). Method : The method applied in this research was the experimental to compare 3 formulas with different Horse Milk concentration then analyzes the physical and chemical characteristics and antibacterial activity. Result and Conclusion : All formulas have the same characteristics are white colour, soft texture, and typical Horse Milk scent. pH all formula respectively are 6,67, 6,41, and 6,31. Result of viscosity are 12666 cPs, 13366 cPs, and 14133 cPs. While the result of the spreadabillity are 0,013 g/cm, 0,011 g/cm, 0,008 g/cm. The results of the antibacterial activity test of Horse Milk gel do not have antibacterial activity against the Propionibacterium acnes bacteria which is indicated by the absence of inhibition zones around the wells.

    Lactoferrin is a multifunctional defense protein, eg. as immunomodulator , which is known as glycoprotein which has 80 kDa molecullar weight. The aim of this research is to characterize of Horse Milk lactoferrin as immunomodulator in humoral immune response. Lactoferrin in defiled Horse Milk, was isolated and purified on Ion Exchange (CM Sephadex C-50) and Gel Filtration (Sephadex G-75) column. Horse Milk lactoferrin is administered orally into a group of 6-8 weeks old female Balb/c mice. The dose of lactoferrin were : 0 nig (control); 0,1 mg; 0,5 mg, and 1,0 mg/mice/day in assay period of 28 day. On the r; 4 216, and 286 day, specific IgA and IgG serum of mice were detected with enzyme-linked immunosrbent assay (ELISA). The results showed that IgG and Ig,4 responses of mice that were administered with 0,5 mg or 1,0 mg Horse Milk lactoferrin, were higher than the control and 0,1 mg lactoferrin (P

    Lactoferin is an iron-binding glycoprotein, has a very broad range of biological functions relating to the host defence system, e.g as immunomodulator. The aim of this research were to study of Horse Milk lactoferrin glycan type, and immune responses (phagocytosis activity and IgA producing cells) of mice fed Horse Milk lactoferin. Horse Milk lactoferin was purified on cation exchange chromatography and gel filtration, and glycan type was analyzed with spesific lectin. Female Balb/c mice 6-8 weeks old, fed 0.0 (control); 0.1; 0.5; 1.0 mg/mice/day of Horse Milk lactoferin orally, in assay period of 28 day. In vitro phagocytosis activity were assayed with latex beads, and IgA producing cells was analyzed with IHC (immunohistochemical) methods. The result showed that molecular weight of Horse Milk lactoferin was 79.18 kDa, and its glycan type reacted with WGA (wheat germ agglutinine), but not with Con-A (concanavalin A) lectin. The phagocyte cells and IgA producing cells of mice fed Horse Milk lactoferin were higher than the control mice. In conclusion, Horse Milk lactoferin had complex type glycan and could stimulate the mice immune responses through the increasing phagocytosis activity of periteal macrophage and IgA producing cells in the intestinal tissue Key Words: Horse Milk Lactoferin, Glycan, Phagocytosis, IgA Producing Cells

    Abstract:

    Many kinds of Milk protein components have different chemical and physical characteristics that determine its bioactive properties, including its immune function. The aim of this research were to isolate and characterize of protein in pasteurized and fermented Horse Milk as immunomodulator in mice humoral (serum antibody) and cellular immune responses (lymphocyte proliferation and phagocytosis activity). Protein of pasteurized Horse Milk was fractionated on Sephadex G-75 column, and obtained two protein fractions (fraction 1 and fraction 2). Female mice BALB/c 6 – 8 weeks old were administered orally with protein of pasteurized and fermented Horse Milk up to 28 day of experiment. On the 7th, 14th, 21th, and 28th day of experiment, the serum IgG and IgA were analized with enzyme-linked immunosorbent assay (ELISA). Whereas phagocytosis activity of peritoneal macrophage and lymphocyte proliferation were analyzed on the 14th and 28th day of experiment. The pasteurized and fermented Horse Milk protein were characterize with sodium dodecyl sulphate polyacrylamide gel electrrophoresis (SDS-PAGE). The result of this research showed that protein of pasteurized and fermented Horse Milk can take part as immunomodulator in humoral immune response, stimulate in phagocytosis activity, but there were no effect on lymphocyte proliferation.
