Immunodiffusion test for diagnosis and monitoring of human pythiosis insidiosi

A soluble antigen was prepared from a human isolate of Pythium insidiosum, an aquatic, thermotolerant oomycete that causes infections in cattle, dogs, horses, and humans. Sera from seven proven cases of disseminated human pythiosis insidiosi were tested in an Immunodiffusion test along with appropriate control sera from patients with a variety of actinomycotic, bacterial, and mycotic diseases as well as sera from uninfected individuals. Titers ranged from 1:1 to 1:32 in the seven serum samples from the disseminated cases of pythiosis insidiosi of varying severity. The heterologous sera gave negative reactions. The rapidity and specificity of the Immunodiffusion test makes it a useful diagnostic tool for the serodiagnosis of P. insidiosum infections.

Complement fixation tests were positive for both sera using a polysaccharide-rich antigen. This study reports the results of a more extensive serological investigation of these two sera. Both a somatic antigen and a saline extract from the fungus yielded positive results in the double Immunodiffusion. However, the immunodominant 43 kDa glycoprotein antigen showed negative results, although it was recognized by both sera in the Western blot assay. The value of the double Immunodiffusion as a single serological test in paracoccidioidomycosis diagnosis is discussed.

Journal of Virological Methods, 2002Co-Authors: Yeon K Choi, Gertrude C BuehringAbstract:Abstract Three serologic methods for the detection of antibodies to bovine leukemia virus (BLV) were compared using the sera of 140 dairy cows. A widely used commercial agarose Immunodiffusion screening assay and a commercial antibody capture enzyme immunosorbent assay were compared for sensitivity and specificity with immunoblotting as the standard. The immunoblot utilized the same antigen preparations that were provided in the commercial kits. The agarose Immunodiffusion and the enzyme immunosorbent assay were comparable in the number of positive animals detected. However, the commercial screening kits failed to detect 39% (agarose Immunodiffusion) and 35% (immunosorbent assay), respectively, of the animals determined serologically positive by immunoblot. These findings corroborate those of some other groups and emphasize the need for more sensitive tests to identify BLV positive cattle for culling or separation in order to create BLV-free herds.
