    Abstract:

    Summary Inonotus Obliquus has been used as a folk remedy for a long time. An improved method using fine grinding by ball mill was investigated, and the main chemical components and physicochemical properties of Inonotus Obliquus powders were analysed. The Inonotus Obliquus fine grinding powders were prepared by pulverising the coarse powder with planetary ball mill for different time. The results showed that the yields of polysaccharides increased from 41.08 mg g−1 (coarse powder) to 319.62 mg g−1, and the yields of polyphenols were also increased fourfold in ball-milled powders as compared to the untreated one. Ball-milled powders showed higher values in the bulk density, angles of repose and slide, water solubility index and swelling capacity (P < 0.05), but lower values in the water-holding capacity and oil-holding capacity. The shape and surface morphology of the powders were changed after ball mill treatment. The antioxidant properties of the extracts from ball-milled powders were significantly improved (P < 0.05). The results will be helpful for the application of Inonotus Obliquus in food and pharmaceutical industries.

    Abstract:

    Abstract Inonotus Obliquus is a kind of mushroom which has long been used as a folk remedy for curing various diseases such as cancers, heart disease and diabetes in Russian and Eastern Europe. Polysaccharides are one of the main bioactive constituents of Inonotus Obliquus with health functions. Three drying methods, freeze drying, hot air drying and vacuum drying methods were comparatively studied on the physicochemical and antioxidant properties of Inonotus Obliquus polysaccharide (IOPS) with chemical methods: gas chromatography, size exclusion chromatography, scanning electron micrograph, 1,1′-diphenyl-2-picrylhydrazyl (DPPH) assay, ferric reducing power assay and lipid peroxidation inhibition assay, respectively. Results showed that physicochemical and antioxidant properties of IOPS differed from each other after the treatment of the three drying methods. Compared with hot air drying and vacuum drying methods, freeze drying method resulted in the properties of IOPS with lower molecular weight distribution, a hyperbranched conformation with triple helix, higher antioxidant abilities on DPPH radical scavenging, ferric-reducing power and lipid peroxidation inhibition activity. Freeze drying was a good choice for the preparation of polysaccharides from Inonotus Obliquus and could be used to produce antioxidants for food industry.

    Physical modification of polysaccharides exerted better biological properties because of the change of physicochemical properties. Polysaccharides from Inonotus Obliquus (IOPS) were modified by acid, alkali hydrolysis, thermal and ultrasonic treatment in this study. The physicochemical and antioxidant properties of IOPS and its physical modified products were comparatively investigated by chemical methods, gas chromatography, size exclusion chromatography, scanning electron micrograph, circular dichroism spectra, and ferric reducing power assay and lipid peroxidation inhibition assay, respectively. Results showed that physicochemical and antioxidant properties of IOPS were changed after the physical modification of acid, alkali hydrolysis, thermal and ultrasonic treatment. Thermal treated polysaccharide (Th-IOPS) and ultrasonic treated polysaccharide (Ul-IOPS) showed the properties of lower molecular weight distribution, lower intrinsic viscosity, a hyperbranched conformation, and higher antioxidant abilities on ferric-reducing power and lipid peroxidation inhibition activity compared with the native polysaccharide IOPS. Th-IOPS and Ul-IOPS might be explored as a novel potential antioxidant for food industry.

Among the three derivatives, acetylationed polysaccharide (Ac-IOPS) resulted in lower molecular weight distribution, lower intrinsic viscosity, a hyperbranched conformation, higher antioxidant abilities on ferric-reducing power and lipid peroxidation inhibition activity compared with the native polysaccharide IOPS. Ac-IOPS might be explored as a novel potential antioxidant for human consumption.

    Abstract:

    Abstract The medicinal mushroom Inonotus Obliquus has been used as a folk remedy for a long time in Russia and East-European countries to treat gastrointestinal cancer, cardiovascular disease and diabetes. In our study, a water-soluble polysaccharide (ISP2a) was successfully purified from I. Obliquus by DEAE-Sepharose CL-6B and Sepharose CL-6B column chromatography. In vivo ISP2a had not only shown antitumor activity, but also could significantly enhance the immune response of tumor-bearing mice. In addition, ISP2a significantly enhanced the lymphocyte proliferation and increased the production of TNF-α. Results of these studies demonstrated that ISP2a had a potential application as natural antitumor agent with immunomodulatory activity.

    Abstract Five polysaccharides (IOP1b, IOP2a, IOP2c, IOP3a and IOP4) were isolated and purified from Inonotus Obliquus by DEAE-Sepharose fast flow and SepharoseCL-6B column chromatography. Their chemical and physical characteristics were determined and antioxidant activities were investigated on the basis of hydroxyl radical assay, superoxide radical assay and ferric-reducing antioxidant power assay. The results showed that five polysaccharides exhibited antioxidant activities, and the higher content of uronic acid and proteinous substances, the stronger antioxidant activities of polysaccharides. Besides, molecular weights of polysaccharides also influence their antioxidant activities. IOP3a and IOP4 showed higher antioxidant properties than IOP1b, IOP2a and IOP2c.

    :  An ultrasonic technique was employed to extract polysaccharides from Inonotus Obliquus (UPS). The effects of ultrasonic conditions on the recovery and 1, 1-diphenyl -2-picrylhydrazyl (DPPH) radical scavenging activity of UPS were evaluated. The physicochemical properties of UPS were analyzed by differential scanning calorimetry (DSC), scanning electron microscopy (SEM), and UV-visible spectrophotometry. Molecular weights of UPS were determined by gel permeation chromatography (GPC). The optimal ultrasonic conditions to obtain the highest recovery of polysaccharides were 80 min, 75 °C, and 100 w. While the optimal ultrasonic conditions to obtain the strongest DPPH scavenging activity of polysaccharide were 80 min, 95 °C, and 160 w. The differences were found by DSC, SEM, GPC analysis and no apparent differences were found from the UV-vis spectra of the polysaccharides after the ultrasonic treatment, which indicated that ultrasonic treatment might cause the physicochemical changes in UPS conformation and degradation of UPS chain but not the main groups. Ultrasonic technique was an effective method to extract bioactive polysaccharides from I. Obliquus (UPS). Practical Application: Polysaccharides from Inonotus Obliquus exhibited many biological activities including antitumor, antioxidant, hypoglycemic, immune-stimulating effects, and so on. Ultrasonic treatment is now an usual proceeding technique for the extraction of some bioactive constituents. But there was no information about the effects of ultrasonic treatment on the physicochemical properties and DPPH radical scavenging activity of polysaccharides from I. Obliquus until now. Ultrasonic technique was an effective method to extract bioactive polysaccharides from I. Obliquus (UPS) according to this study.

    Abstract:

    A water-soluble polysaccharide (IOPS) was isolated from the Inonotus Obliquus (Fr.) Pilat, which was a medicinal mushroom with many health-benefiting functions. The chemical composition and acute toxicity of IOPS was investigated in order to determine the possibility of using them for human consumption. Chemical analysis of IOPS showed that it was a nonstarch protein bounded acidic polysaccharide. The protein, neutral sugar, uronic acid contents of the polysaccharide IOPS were 42.50%, 18.50% and 6.10%, respectively. The neutral sugars of IOPS were composed of Rha (D-rhamnose), Xyl (D-xylan), Gal (D-galactose) and Glc (D-glucose). The contents of thirteen mineral elements in IOPS were measured by inductivity coupled plasma-optical emission spectrometry (ICP-OES). Results showed that the concentrations of the elements in IOPS were much higher than that of Inonotus Obliquus powder. IOPS, when orally administrated to mice at the single dose of 5000 mg/kg body weight, was found no toxicity to the liver, kidney, heart, thymus or spleen of the mice and none of the mice died throughout the period of the experiment. Based on the study, IOPS could be classified either as a very low toxicity substance, i.e. GHS Category 5 (globally harmonized system) or unclassified when orally administrated to mice and it might be a candidate of dietary supplements.

    ABSTRACT A water-soluble polysaccharide from Inonotus Obliquus (IOPS) was isolated from the mushroom Inonotus Obliquus (Fr.) Pilat. The chemical compositions, molecular weight and inhibitory activities on glycosidase and antioxidant properties of IOPS were investigated. The results indicated that IOPS was an acid protein-bound polysaccharide, with a molecular weight of 1.7 × 104 Da and the contents of neutral sugar, protein and uronic acids being 42.5, 18.5 and 6.1%, respectively. IOPS exhibited an inhibitory activity against α-glucosidase with the IC50 value of 93.3 µg/mL, whereas it had no effective inhibition on α-amylase. Results of antioxidant activity assays revealed that IOPS had inhibitory activity on the concentration-dependent quenching of 1,1-Diphenyl-2-picrylhydrazyl and hydroxyl radicals. Furthermore, IOPS inhibited the formation of thiobarbituric acid-reactive substances in Fe2+/ascorbate-induced lipid peroxidation in rat liver tissue. These results clearly demonstrated that IOPS was one of the main bioactive components of I. Obliquus that contributed to hypoglycemic activity and antioxidant activity. PRACTICAL APPLICATIONS Diabetes mellitus is one of the primary threats to human health because of its increasing prevalence, chronic course and disabling complications. Postprandial hyperglycemia plays an important role in the development of type 2 diabetes mellitus and complications associated with the disease. One therapeutic approach to decrease postprandial hyperglycemia is to retard the absorption of glucose through inhibition of carbohydrate-hydrolyzing enzymes in the digestive organs. In this study, a polysaccharide isolated from the mushroom Inonotus Obliquus (IOPS) was shown to have notable glycosidase inhibitory effects and antioxidant activities. This research will benefit for the investigation of effective and safe α-glucosidase inhibitors from natural materials. IOPS could be a good candidate for application in food and medicinal fields. It might be developed for functional food or lead compounds for use in antidiabetes.

    Abstract A simple and rapid synthetic methodology for gold nanoparticles (AuNPs) was developed using the extracts of Inonotus Obliquus (Chaga mushroom) at room temperature without the use of deleterious agents. The AuNPs were spherical, triangle, hexagonal, and rod shaped with a mean diameter of 23 nm. The eco-friendly synthesized AuNPs showed excellent antibacterial, antioxidant, and cytotoxicity against the MCF-1 human breast cancer cell line and NCI-N87 human stomach cancer cell line.

    Abstract In the present study, silver nanoparticles (AgNPs) were rapidly synthesized from silver nitrate solution at room temperature using Inonotus Obliquus extract. The mycogenic synthesized AgNPs were characterized by UV–Visible absorption spectroscopy, Fourier transform infrared (FTIR), scanning electron microscopy (SEM) with energy dispersive spectroscopy (EDS), transmission electron microscopy (TEM) and atomic force microscopy (AFM). SEM revealed mostly spherical nanoparticles ranging from 14.7 to 35.2 nm in size. All AgNPs concentrations showed good ABT radical scavenging activity. Further, AgNPs showed effective antibacterial activity against both gram negative and gram positive bacteria and antiproliferative activity toward A549 human lung cancer (CCL-185) and MCF-7 human breast cancer (HTB-22) cell lines. The samples demonstrated considerably high antibacterial, and antiproliferative activities against bacterial strains and cell lines.
