    Abstract:

    Recombinant antibody phage display technology has been used to mimic many aspects of the processes that govern the generation and selection of high-affinity natural human antibodies in the human immune system, especially for infectious disease prophylaxis. An anti-rabies virus immunized phage-display Fab library was constructed from peripheral blood lymphocytes from vaccinated volunteers. The immunized antibody library, with a diversity of 6.7610 8, was used to select and produce antibodies that bound to rabies virus glycoprotein. After five rounds of immobilized fixed rabies virion panning, four unique DNA sequences were found in the higher binding clones, and only one, Fab094, showed neutralization activity. Fab094 components were analyzed by ELISA, immunoprecipitation and immunofluorescent staining. ELISA and immunofluorescence showed that Fab094 bound specifically to rabies virions. Immunoprecipitation and mass spectrometry showed that Fab094 reacted with rabies virus glycoprotein. To improve the penetration power of Fab094 antibodies, we developed Fab094 calcium phosphate nanoparticles (Fab094-CPNPs) and tested their efficacy. The rapid fluorescent focus inhibition test indicated that the neutralizing antibody titers of Fab094 and Fab094-CPNPs were reached at 200.17 IU/Kg and 246.12 IU/Kg, respectively. These findings were confirmed in vivo in a Kunming Mouse challenge model. Our results demonstrate that human Fab094 and Fab094-CPNPs are efficacious candidate drugs to replace rabie

    Abstract:

    The spleen lymphocytes of Kunming Mouse and BALB/c Mouse were isolated and then cultivated in PRMI-1640 culture containing 10 mg/mL Con-A for activation.The total cellular RNA was extracted and the invariant chain(P31,P41) were obtained by RT-PCR,then the sequences were analyzed using DNAStar software.Then the BALB/c Mouse P31/P41 molecules were cloned into the multiple cloning site of pGEX-KG to construct the prokaryotic expression vectors.The GST-P31/P41 proteins were expressed by IPTG induction and purified with Glutathione Sepharose TM 4B affinity columns according to the manufacture's instructions,the purified GST-P31/P41 proteins were emulsified with Freund' s adjuvant to prepare the water-in-oil vaccine for rabbits subcutaneous immunization.The sequencing results showed that the P31 and P41 were 648 bp and 840 bp in length encoding 215 and 279 amino acids respectively.Sequences alignment showed that there were multiple nucleotide and/or amino acid differences in both of P31 and P41 of this two strain Mouse when compared with the sequences that published in GenBank.Furthermore,the nucleotide homology and amino acid homology between this two cloned P31 were 98.8% and 98.6%,respectively,and the nucleotide homology and amino acid homology between this two cloned P41 were all 99.3%.Then the sequences comparisons of Mouse P31/P41 with human and chicken corresponding Ii molecules were analyzed,the nucleotide homology and amino acid homology between Mouse and human were 79.0% and 73.0% or more respectively,while the nucleotide homology and amino acid homology between Mouse and chicken were only 55.0% and 41% or more respectively.The rabbit anti-Mouse P31/P41 polyclonal serum antibodies were collected and tested by ELISA and Western-blotting.These results indicated that the sequences of Ii between Kunming Mouse and BALB/c Mouse were not completely consistent,the rabbit anti-Mouse Ii polyclonal antibodies were obtained and could be used to develop the targeting epitope vaccine.
