Herein the roLE of immunoglobulin G (IgG) anti-dsDNA autoantibodies in nucLEo-phagocytosis by polymorpho-nucLEar Cells (PMN) has been analyzed. Necrotaxis, phagocytosis and PMN with engulfed nucLEi can be imaged by vital microscopy. Vital microscopy reveaLEd dead neutrophils with persistence of nucLEi or ingested nucLEi in 8/10 patients with lupus erythematosus (LE) and in 4/20 healthy controls (P < 0.0006). The phagocytic cLEarance of dead Cells/nucLEi did not correspond to any of the analyzed apoptosis markers (annexin V, CD95, and active caspase 3). IgG staining of isolated PBMC nucLEi as detected by flow cytometry was significantly higher in paralLEl to NUC+ (P < 0.02). In vitro phagocytosis of substrates pre-treated with anti-dsDNA containing serum induced an increased phagocytic capacity as compared to normal serum. These data suggest that anti-dsDNA antibodies seem to inhibit enzymatic cLEavage of nucLEi from damaged Cells, and improve their phagocytic uptake by PMN, which finally results in LE Cell formation.

Since the lupus erythematosus (LE) Cell phenomenon is a special form of necro-phagocytosis in the presence of anti-dsDNA antibodies, dead substrate Cells or chicken erythrocytes nucLEi (CEN) served as targets that were labeLEd with propidiumiodide (PI). Phagocytes (PMN) were stained by anti-CD45 mAb FITC. After co-incubation phagocytosis was measured by flow cytometry. Flow cytometric analysis enabLEd the discrimination between PI+/CD45- targets, PI-/CD45+ phagocytes, and PI+/CD45+ phagocytes with engulfed targets. Maintaining the sampLEs on ice significantly reduced the phagocytic uptake as compared to sampLEs co-cultivated at 37 degrees C (p < 0.0002). The phagocytic up-take was lowest after substrate pre-treatment in normal serum as compared to sampLEs with either no serum exposure or pre-treatment in LE-serum with anti-dsDNA antibodies (p < 0.05). Taken together, these data suggest the phagocytosis-based flow cytometry assay is suitabLE for analyzing the LE Cell phenomenon. This method provides an interesting, simpLE and rapid new tool, and will possibly alLEviate further studies on the LE Cell phenomenon with modified Cell models and/or conditions.

Due to their differentiation capabilities, ASCs became Cells of considerabLE interest for regenerative medicine and are of high interest for type II diabetes diagnosis. Known to migrate and circulate in lymph, the hypothesis of their presence in blood is not excluded but no method exists to prove it. The aim of this study is to develop a lab-on-chip abLE to isolate ASCs from compLEx biological sampLEs by using passive and label-free microfluidic sorting methods. These methods involve intrinsic properties of fluids and objects. Yet, ASCs do not have specific physical characteristic. We have demonstrated that their diameter is comprised between 10 and 25 µm: they cannot be distinguished from most of other blood Cells. In addition, they do not present specific antigen on their membrane. In order to compLEtely isolate ASCs from other Cell types, we propose an original approach combining two compLEmentary steps. The first step aims at pre-treating the sampLE by removing, via hydrodynamic filtration, all the Cells with a diameter below 10 µm. With this device, red blood Cells, which represent more than 99% of blood Cells, plateLEts and some LEukocytes, have to be removed. This study has demonstrated that the device is abLE to effectively pre-treat pure blood sampLE (either from human or from mouse) as it removes more than 99.9% of red blood Cells. It has also been demonstrated that filtration does not LEad to Cell lysis, which is a promising result for Cell viability and the reuse of Cells after filtration. The obtained sampLE contains Cells of interest and some remaining hematopoietic Cells. The second step aims at refining ASCs isolation by separating them from remaining hematopoietic Cells. The method used, calLEd immunological exclusion by Cell rolling, is based on antigen-antibody specific reaction. As ASCs do not have specific antigen, LEukocytes antigens have been involved. The objective is so to depLEte the sampLE of the remaining LEukocytes. This study LEd to the elaboration of an optimised surface functionalization protocol. eLEctrochemical characterizations on mno2 supercapacitors with potassium polyacrylate and potassium polyacrylate co polyacrylamide gel polymer eLEctrolytes
Electrochimica Acta, 2009Co-Authors: Kuangtsin Lee, Jyhfu LeeAbstract:Abstract MnO2·nH2O supercapacitors with potassium polyacrylate (PAAK) and potassium polyacrylate-co-polyacrylamide (PAAK-co-PAAM) gel polymer eLEctrolytes (GPEs) having the weight compositions of polymer:KCl:H2O = 9%:6.7%:84.3% have been characterized for their eLEctrochemical performance. Compared with the liquid eLEctrolyte (LE) counterpart, the GPE Cells exhibit remarkabLE (∼50–130%) enhancement in specific capacitance of the oxide eLEctrode, and the extent of the enhancement increases with increasing amount of the carboxylate groups in the polymers as well as with increasing oxide/eLEctrolyte interfacial area. In situ X-ray absorption near-edge structure (XANES) analysis indicates that the oxide eLEctrodes of the GPE Cells possess higher Mn-ion vaLEnces and are subjected to greater extent of vaLEnce variation than that of the LE Cell upon charging/discharging over the same potential range. Copolymerization of PAAK with PAAM greatly improves the cycling stability of the MnO2·nH2O eLEctrode, and the improvement is attributabLE to the alkaline nature of the amino groups. Both GPEs exhibit ionic conductivities greater than 1.0 × 10−1 S cm−1 and are promising for high-rate applications.
