Ultra-sonografia bidimensional em tempo real do bulbo ocular de cães (Canis familiaris, Linnaeus, 1758) com opacificação de meios transparentes. Emprego do transdutor mecânico setorial de 7,5 MHz com almofada de recuo

Empregou-se transdutor mecânico setorial de 7,5 MHz com almofada de recuo, em técnica de contato corneano. Para a realização dos exames, os animais foram contidos mecanicamente e as córneas receberam anestesia tópica. A blefarostasia deu-se manualmente. Os bulbos foram avaliados com o transdutor aplicado nas posições vertical e horizontal. As características ultra-sonográficas encontradas em olhos normais o foram a similitude da literatura especializada. Relativamente aos bulbos oculares com opacificação de meios transparentes, encontraram-se alterações junto às estruturas e compartimentos. Destacaram-se as cataratas, as luxações e subluxações do cristalino, as degenerações do vítreo, as hemorragias e inflamações em câmaras anterior e vítrea e os descolamentos de retina. A base dos achados e a fidedignidade das imagens colhidas permitem admitir que a ultra-sonografia bidimensional em tempo real, utilizando o transdutor mecânico setorial de 7,5MHz com almofada de recuo, adequa-se ao exame das estruturas intra-oculares, mormente na Medicina Veterinária em que os recursos são parcos à aplicação de procedimentos mais sofisticados ou de equipamentos para ultra-sonografia próprios à especialidade.The two-dimensional real-time ultrasonography was used to evaluate 20 normal eyes of 10 dogs and 55 eyes o f 30 dogs with ocular opacity. Open Access Primary Lens Luxation in Australian Tenterfield and Miniature Bull Terriers is Due to An Old ADAMTS17 Mutation and is an Additive Trait
2016Co-Authors: Puya Gharahkhani, David Duffy, Michael BernaysAbstract:Abstract: Primary Lens Luxation (PLL) is an inherited disease common in Terrier breeds. A truncating mutation in the ADAMTS17 orthologue on CFA03 is reported to cause PLL in 17 breeds, mostly Terriers. This study investigated the mode of inheritance and penetrance of PLL, age of the ADAMTS17 mutation, and other possible causes of PLL in Australian Tenterfield Terriers and Miniature Bull Terriers. The ADAMTS17 mutation and 30 nearby microsatellites on CFA03 were genotyped in 66 Australian Tenterfield Terriers, and 74 Miniature Bull Terriers. Lifetime risk of developing clinical PLL in homozygote and heterozygous animals, penetrance of PLL in general population, and recurrence risk of the disease for offspring and siblings of an index case was estimated. The effect of the ADAMTS17 mutation on the risk of PLL was dose and age dependent, and best fitted an additive model. Primary Lens Luxation was fully penetrant in homozygotes over 6 years and incompletely penetrant with a significant risk of PLL in heterozygote animals. Microsatellite haplotype testing estimated the mutation to be an old mutation, consistent with its appearance on several different background haplotypes and suggesting it entered the tested Tenterfield Terrier population at least three times, and the tested Miniature Bull Terrier population once. Finite polygenic model and survival analyses suggested that the ADAMTS17 mutation is the main cause of PLL in these breeds, and if the PLL phenotype is controlled by factors other than this mutation, such as a second locus or an environmental effect, the effect is much less than that of this diseas

Primary Lens Luxation (PLL) in dogs is an inherited disease in which the Lens is displaced from its normal position. A truncating mutation in the ADAMTS17 orthologue on CFA03 is reported to cause PLL in several breeds, mostly terriers. However, the complex inheritance pattern of PLL in miniature bull terriers (MBTs) suggests that other loci may have a modifying effect on the ADAMTS17 mutation. This study aimed to detect such loci increasing risk of PLL in Australian MBTs. METHODS. More than 170,000 single-nucleotide polymorphisms (SNP) across the canine genome were genotyped in 23 PLL-affected and 73 normal Australian MBT, and association between the PLL phenotype and the genetic markers was investigated by using general mixed effects Cox model survival analysis. RESULTS. The highest association peaks, other than that associated with the ADAMTS17 mutation (P = 2.2e-05), were SNP BICF2G630420272 located at 62.2 Mb on chromosome 15 (P = 7.8e-05) and the region between 30 Mb and 32.5 Mb on chromosome 1 (P = 9.3e-05). Joint analysis showed that the PLL-associated allele of the BICF2G630420272 SNP increased risk of PLL in the presence of the ADAMTS17 mutation (P = 8.117e-04). Candidate genes in the two regions of interest included CPE on chromosome 15 and CTGF on chromosome 1. The ADAMTS17 mutation was also associated with abnormal foot and nail shapes, pedal hyperkeratosis, and persistent pupillary membranes. CONCLUSIONS. Two loci with potentially enhancing effects on the ADAMTS17 mutation were associated with PLL in Australian MBTs. Association of the ADAMTS17 mutation with possible pedal skeletal abnormalities in MBTs supports PLL in this breed and Weill-Marchesani syndrome-like disease in humans as being homologous diseases.

To identify the genetic cause of isolated canine ectopia lentis, a well-characterized veterinary disease commonly referred to as primary Lens Luxation (PLL) and to compare the canine disease with a newly described human Weill-Marchesani syndrome (WMS)-like disease of similar genetic etiology. METHODS. Genomewide association analysis and fine mapping by homozygosity were used to identify the chromosomal segment harboring the PLL locus. The resequencing of a regional candidate gene was used to discover a mutation in a splice donor site predicted to cause exon skipping. Exon skipping was confirmed by reverse transcription-polymerase chain reaction amplification of RNA isolated from PLL-affected eyes and from skin fibroblast cultures from PLL-affected dogs. An allelic discrimination assay was used to genotype individual dogs at the splice donor site mutation. RESULTS. The PLL locus was mapped to a 664-kb region of canine chromosome 3 containing regional candidate gene ADAMTS17. Resequencing ADAMTS17 revealed a GT→AT splice-donor-site mutation at the 5' end of intron 10. The predicted exon 10 skipping and resultant frame shift were confirmed with RNA derived from PLL-affected dogs. The ADAMTS17 mutation was significantly associated with clinical PLL in three different dog breeds. CONCLUSIONS. A truncating mutation in canine ADAMTS17 causes PLL, a well-characterized veterinary disease, which can now be compared to a recently described rare WMS-like disease caused by truncating mutations of the human ADAMTS17 ortholog.

Several changes were found in the structures and chambers of the eye which presented opacities of the media, such as cataracts, Lens Luxation and sub-Luxation, vitreous degeneration, hemorrhage and inflammation in the vitreal chamber, besides retinal detachments. On the basis of the studies carried out and the obtained results, it was possible to conclude that the two-dimensional real-time ultrasonography, with the use of a 7.5MHz mechanical sector fluid offset transducer shows a satisfactory adaptability to the evaluation of the intra-ocular structures. It is indicated specially in Veterinary Medicine in which resources are scanty to sophisticated procedures or specific eye ultrasound equipment

Ultra-sonografia bidimensional em tempo real do bulbo ocular de cães (Canis familiaris, Linnaeus, 1758) com opacificação de meios transparentes. Emprego do transdutor mecânico setorial de 7,5 MHz com almofada de recuo

Empregou-se transdutor mecânico setorial de 7,5 MHz com almofada de recuo, em técnica de contato corneano. Para a realização dos exames, os animais foram contidos mecanicamente e as córneas receberam anestesia tópica. A blefarostasia deu-se manualmente. Os bulbos foram avaliados com o transdutor aplicado nas posições vertical e horizontal. As características ultra-sonográficas encontradas em olhos normais o foram a similitude da literatura especializada. Relativamente aos bulbos oculares com opacificação de meios transparentes, encontraram-se alterações junto às estruturas e compartimentos. Destacaram-se as cataratas, as luxações e subluxações do cristalino, as degenerações do vítreo, as hemorragias e inflamações em câmaras anterior e vítrea e os descolamentos de retina. A base dos achados e a fidedignidade das imagens colhidas permitem admitir que a ultra-sonografia bidimensional em tempo real, utilizando o transdutor mecânico setorial de 7,5MHz com almofada de recuo, adequa-se ao exame das estruturas intra-oculares, mormente na Medicina Veterinária em que os recursos são parcos à aplicação de procedimentos mais sofisticados ou de equipamentos para ultra-sonografia próprios à especialidade.The two-dimensional real-time ultrasonography was used to evaluate 20 normal eyes of 10 dogs and 55 eyes o f 30 dogs with ocular opacity. The ultraso- nographic characteristics found in the normal eye structures were compatible with the experiences related by several dijferent authors. To identify the genetic cause of isolated canine ectopia lentis, a well-characterized veterinary disease commonly referred to as primary Lens Luxation (PLL) and to compare the canine disease with a newly described human Weill-Marchesani syndrome (WMS)-like disease of similar genetic etiology. METHODS. Genomewide association analysis and fine mapping by homozygosity were used to identify the chromosomal segment harboring the PLL locus. The resequencing of a regional candidate gene was used to discover a mutation in a splice donor site predicted to cause exon skipping. Exon skipping was confirmed by reverse transcription-polymerase chain reaction amplification of RNA isolated from PLL-affected eyes and from skin fibroblast cultures from PLL-affected dogs. An allelic discrimination assay was used to genotype individual dogs at the splice donor site mutation. RESULTS. The PLL locus was mapped to a 664-kb region of canine chromosome 3 containing regional candidate gene ADAMTS17. Resequencing ADAMTS17 revealed a GT→AT splice-donor-site mutation at the 5' end of intron 10. The predicted exon 10 skipping and resultant frame shift were confirmed with RNA derived from PLL-affected dogs. The ADAMTS17 mutation was significantly associated with clinical PLL in three different dog breeds. CONCLUSIONS. A truncating mutation in canine ADAMTS17 causes PLL, a well-characterized veterinary disease, which can now be compared to a recently described rare WMS-like disease caused by truncating mutations of the human ADAMTS17 ortholog.
