    Abstract:

    To study the expressive characteristics of SKP2 protein in Lung Carcinoma and its implication for prognosis. The expression of SKP2 protein was detected in 89 non small cell Lung Carcinoma, 13 small cell Lung Carcinoma, 10 Lung benign lesion tissues by Tissue Chip and Immunohistochemistry technology. The positive rate of SKP2 protein staining was (23.52±13.57)% in non small cell Lung Carcinoma and (53.85±12.26)% in small cell Lung Carcinoma, which were significantly higher than (2.91±1.27)% in Lung benign lesion tissues. It was highest in small cell Lung Carcinoma and lowest in Lung benign lesion tissues, with a significant difference between them (P=0.000). The expressive level of SKP2 protein in Lung Carcinoma tissues was closely related to cell differentiation, lymph node metastasis and pathological types, but not to age, sex, smoking history, tumor site and size, and TNM staging. The survival analysis revealed that the 5-year survival rate of Lung Carcinoma patients was lower in SKP2 protein positive expression group than that in negative expression group (P 1=0.003/0.002; r=−0.275, P 2=0.005). The positive expression of SKP2 protein is higher in Lung Carcinoma than in Lung benign lesion tissues, in particular, much higher in small cell Lung Carcinoma. In Lung Carcinoma, its expressive level was closely related to cell differentiation, lymph node metastasis and pathological types. Moreover, it may be an independent factor to prognosis of patients with Lung Carcinoma.

  • Expression of Hsp70 Grp94 and IgG in human Lung Carcinoma

    Abstract:

    AIM To explore the expression and significance of heat shock protein 70 (Hsp70), glucose regulated protein 94 (Grp94) and immunoglobulin G (IgG) in human Lung Carcinoma. METHODS The expression of Hsp70, Grp94 and IgG in 40 human Lung Carcinomas was studied using immunohistochemical technique and image analysis. The localization among Hsp70, Grp94 and IgG was analyzed by double labeling immunofluorescent staining and laser scanning confocal microscopy. RESULTS Hsp70, Grp94 and IgG in Human Lung Carcinomas showed high expression. The positive rate of Hsp70, Grp94 and IgG was 65% (26/40), 45% (18/40), and 82.5% (33/40), respectively. The average value of optical density was 5.10 +/- 0.32, 3.52 +/- 0.35, and 8.12 +/- 0.31, respectively. Hsp70 was localized in nucleus and cellular cytoplasm while Grp94 and IgG were mainly localized in cellular cytoplasm. Ten cases showed Hsp70 was co-localized with IgG and Eighteen cases showed Grp94 was co-localized with IgG. CONCLUSION High expression of Hsp70, Grp94 and IgG in Human Lung Carcinomas suggested that Hsp70, Grp94 and IgG might play an important role in the development of human Lung Carcinoma. IgG is co-localized with Hsp70 or Grp94. The study will lay a theorical basis for further research on anti-tumor immunotherapy.

    Abstract:

    Paraffin sections of 247 primary and metastatic non-small cell Lung Carcinomas, the corresponding non-neoplastic Lungs, and 75 other specimens were examined by immunohistochemical procedures using a panel of antibodies against the specific products of peripheral airway cells: the major surfactant-associated protein and 10-kD Clara cell protein. Non-small cell Lung Carcinoma tumors most frequently positive for either peripheral airway cell marker were adenoCarcinomas (41%), especially those with papillolepidic growth pattern (56%), followed by large cell Carcinomas (25%), other adenoCarcinomas (22%), and squamous cell Carcinomas (16%). Immunoreactivity was mainly focal and the expression of the two peripheral airway cell markers was discordant. The incidence of marker expression was similar in metastatic and primary non-small cell Lung Carcinoma. Other organs and their tumors were negative, with few exceptions. Non-small cell Lung Carcinomas positive for peripheral airway cell markers were associated with younger age and less-intense smoking, and surfactant-associated protein reactivity was more common in women than in men. Peripheral airway cell markers were independent prognostic factors for survival and delayed development of metastases in patients with less-advanced disease. It is concluded that surfactant-associated protein and 10-kD Clara cell protein are specific markers for non-small cell Lung Carcinoma and peripheral airway cell differentiation and provide useful tools to study the pathogenesis, biology, and prognosis of non-small cell Lung Carcinoma.
