    Abstract:

    Tight junctions (TJs) are the most apical junctional complexes and restrict the fluid flux through the paracellular pathway. In the Mammary glands, the tightness of TJs occurs shortly after parturition to prevent the leakage of milk components from the lumen and the loosening of TJs is induced immediately after weaning. Claudins are transmembrane proteins, and their composition at the apical-most regions determines the permeability of TJs. In this study, we investigated the localization and expression patterns of claudin-3 and -4 in the Mammary glands around the lactation period because it is unclear how claudins construct Mammary TJs in the apical-most regions. Our results showed that claudin-3 and -4 change not only their level of expression but also their localization in the processes of parturition, lactation, and weaning. Claudin-3 was concentrated in the apical-most regions during lactation, whereas claudin-4 gradually decreased at the beginning of lactation and increased drastically immediately after weaning. The qualitative change of claudin-3 was also identified by western blotting analysis as an additional band around the lactation period. In addition, parts of the Mammary epithelial cells showed intensive positive reactions to claudin-4 in the lateral membrane and cytoplasm after weaning, concurrently with the involution Mammary glands. These results indicate that claudin-3 in the apical-most regions maintains the impermeable TJs during lactation, and claudin-4 contributes to the permeability changes of TJs immediately after parturition and weaning.

  • Mastectomia radical e unilateral no tratamento de mastite gangrenosa em cabras

    Abstract:

    Com o objetivo de avaliar a mastectomia radical e unilateral como procedimento cirúrgico de escolha no tratamento da mastite gangrenosa foram utilizadas quatro cabras, na faixa etária de dois a três anos de idade, sem raça definida. Todos os animais passaram por exame clínico, foram considerados portadores de mastite gangrenosa de curso agudo variando de três a quatro dias pós-partos. Submetidas à anestesia epidural baixa e mastectomia radical e unilateral. Na cultura bacteriana das amostras do leite ordenhado no período pré-operatório da glândula mamária não acometida pela mastite gangrenosa obteve-se a frequência de 25% (1/4) de cada microrganismo isolado, sendo respectivamente Staphylococcus sp. associado a Corynebacterium sp., Streptococcus sp., Corynebcterium sp.. Na cultura bacteriana da secreção da parte central do parênquima da mama gangrenosa de cada animal foram identificados nas amostras Corynebcterium sp., Staphylococcus coagulase negativa, Enterococcus faecalis eStaphylococcus sp. associado a Corynebacterium sp. com a frequência de 25% (1/4). Ao exame histológico do parênquima da glândula mamária os cortes revelaram presença de necrose de coagulação, proliferação fibroblástica, colágeno e infiltrado inflamatório mononuclear. Formação de placas hialinas sob o endotélio vascular e dilatação dos ductos excretores dos alvéolos mamários. Alguns “clusters” bacterianos limitados perifericamente por tecido conjuntivo. Hemorragia e hemosiderose no alvéolo mamário. Material caseoso na luz de alguns ductos excretores. Em outros, observou-se rosetas. O tempo de observação pós-operatória foi de 18 meses. Com o procedimento empregado mostrou-se a recuperação vital dos animais operados. Concluiu-se que a mastectomia, radical e unilateral, se presta para o tratamento da mastite gangrenosa. É essencial a adoção de medidas práticas de baixo custo que facilitem a vida dos produtores diminuindo seus gastos. Têm-se a perspectiva de que este trabalho sirva de subsídiopara a preservação no rebanho de animais de alta linhagem genética.With the objective of to evaluate the radical and unilateral mastectomy as surgical procedure of choice in the treatment of the gangrenous mastitis four goats were used in the age from two to three years old, without defined race.     Abstract:

    This study was aimed to know the effects of oyster mushroom supplement to milk secretion and Mammary Alveolus diameter of mice. The research was carried out under laboratory condition at the laboratory of Reproduction Biology, Faculty of Mathematic and Sciences, Airlangga University by using Complete Random Method. The study was designed by using totally 32 mice post partum (PP) and devided into four groups of treatment which consisted 8 female mice post partum of each group. The groups are: (a) the control groups, the groups that were given 0.2 ml aquadest, (b) the treatments of oyster mushroom solution with 3 variation of mushroom concentration, there are 2%, 4% and 6%. The oyster mushroom solution was given by gavage. The treatments were started on 3th to 12th day of lactation period. The increasing of milk secretion were showed by the increasing of body weight of offsprings. The data were collected from the difference result of the balance of weight body of offspring and the data were observed on 4th, 6th, 8th, 10th, and 12th of lactation periode and then on 13th day, the mice (five mice for each group) were killed to make histological preparat of Mammary glands. The datas were analyzed by ANOVA and LSD test (a = 0.05). The results of this research showed that oyster mushroom supplement can increase milk secretion and Mammary Alveolus diameter of mice. Oyster mushroom with 6% concentration gave the best effects And there was a positive corelation between the weight of oyster mushroom that was used with the increasing of the oyster mushroom effects
