optimising protocol for successful development of haploids in Marigold tagetes spp through in vitro androgenesis

Various factors affecting the in vitro androgenesis response in Tagetes spp. were optimised. At first, a correlation was established between floret size/bud size and stage of microspore development in different genotypes of Tagetes erecta and Tagetes patula. The floret size of 3.5–4.0 mm (French Marigold) and 3.0–3.5 mm (African Marigold) contained the highest percent of microspores at early-uninucleate to early-binucleate stage. The frequency of shoot formation from anthers ranged from 0 to 78.6% in T. patula cv. Pusa Arpita and 0 to 63.9% in T. erecta cv. Pusa Basanti Gainda. The French Marigold cultivars exhibited higher androgenic response over the African Marigold cultivars. The effects of basal media, plant growth regulator combinations and sucrose concentration for direct differentiation of shoot buds and haploid induction were studied. Marigold anthers cultured on newly formulated EMS1 basal media (enriched with coconut water, AgNO3, PVP etc.) exhibited a very high androgenic response over the other commercially available media. We have also optimised the cold temperature pre-treatments and culture condition for high frequency androgenesis in Marigold. Ten days cold pre-treated anthers along with 20 days dark incubation of cultures proved highly beneficial for haploid induction. Among a total of 424 anther-regenerants, 56 plants were randomly selected for flow cytometry and cytological analysis. The ploidy analysis revealed 14.3% of anther regenerants as dihaploids, 66.1% as tetraploid (similar to donor mother plant) and 19.6% as polyploid. The determination of ploidy level by counting the number of chloroplast in stomatal guard cells of Marigold was also established for rapid screening of haploids. The identified haploids were successfully diploidised and are being utilised in the hybrid breeding programme at our institute. The developed protocol will facilitate doubled haploids based breeding in Tagetes spp. This is the first report of successful induction of haploids in Tagetes spp. using in vitro androgenesis techniques. We have studied various factors affecting the in vitro androgenesis response in Tagetes spp. A correlation was established between floret size/ bud size and stage of microspore development in different genotypes of Tagetes erecta and Tagetes patula. Basal media and cultural conditions were optimised for high frequency androgenesis in Marigold. PENGARUH PEMBERIAN TEPUNG DAUN Marigold (Tagetes erecta) DALAM RANSUM TERHADAP PERFORMA AYAM BROILER

Marigold mempunyai dua jenis karotenoid yaitu karoten dan xantofil. Selain karotenoid tanamanini juga mengandung flavonoid dan polifenol yang berfungsi sebagai antioksidan alami.Penelitian ini bertujuan untuk mengevaluasi pengaruh pemberian tepung daun Marigold dalam ransum terhadap performa ayam broiler.Penelitian ini dilaksanakan pada bulan April-Mei 2018, berlokasi diZona Peternakan Terpadu (Integrated Livestock Zone) Jurusan Peternakan Fakultas Pertanian Universitas Bengkulu. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) menggunakan seratus dua puluh ekor broiler umur 14 hari didistribusikan kedalam 5 kelompok perlakuan dimana masing-masing kelompok perlakuan terdiri 4 ulangan, dan masing-masing ulangan terdiri 6 ekor broiler. Adapun ke 5 perlakuan sebagai berikut P0 sebagai kontrol, pemberian ransum tanpa mengandung daun Marigold, P1 broiler diberikan ransum yang mengandung 1,25% daun Marigold, P2 broiler yang diberi ransum mengandung 2,5% daun Marigold, P3 broiler yang diberi ransum mengandung 3,75% daun Marigold dan P4 broiler yang diberi ransum mengandung 5% daun Marigold. Variabel yang diamati dalam penelitian ini adalah data konsumsi ransum, berat badan, pertambahan berat badan, dan konversi ransum, yang dianalisis menggunakan Analisis Varian (Anova) dan jika berpengaruh nyata diuji lanjut dengan menggunakan uji Duncan‟s Multiple Range Test (DMRT). Hasil penelitian menunjukkan bahwa pemberian tepung daun Marigold dalam ransum berpengaruh nyata (P>0.05) terhadap konsumsi ransum, berat badan dan pertambahan berat badan, namun berpengaruhtidak nyata terhadap konversi ransum ayam broiler. Pemberian tepung daun Marigold ke dalam ransum dapat diberikan sebanyak 1,25%. Dengan demikian, pemberian tepung daun Marigold (Tagetes erecta) sampai 5% dapat menggantikan feed additive dalam ransum terutama pada konsumsi ransum, berat badan dan pertambahan berat badan ayam broiler

PENGARUH PEMBERIAN TEPUNG DAUN DAN TEPUNG BUNGA Marigold (Tagetes erecta) DALAM RANSUM TERHADAP ORGANOLEPTIK AYAM BROILER
2019Co-Authors: Saputra Memo, Urip Santoso, Yosi FenitaAbstract:Penelitian ini bertujuan untuk mengevaluasi pengaruh pemberian tepung daun Marigold dan tepung bunga Marigold (Tagetes erecta) terhadap karakteristik organoleptik ayam broiler. Industri broiler dihadapkan pada adanya tuntutan konsumen yang menginginkan rasa yang enak, bau daging yang kurang amis lebih digemari, dan warna daging yang lebih cerah tidak pucat lebih menarik minat konsumen. Marigold (Tagetes erecta) mempunyai senyawa aktif antara lain berfungsi sebagai antibakteri, antimikroba, antioksidan, dan nematoda karena kandungan terpenoid dan flavonoidnya. Penelitian ini dilaksanakan 8 Februari -15 Maret 2019 bertempat dikandang Jurusan Peternakan Fakultas Pertanian Universitas Bengkulu. Rancangan yang digunakan adalah Rancangan Acak Lengkap (RAL), 6 perlakuan dan 4 ulangan. Masing-masing ulangan terdapat 7 ekor ayam. Adapun 6 perlakuan sebagai berikut: P0: ransum kontrol, tidak mengandung tepung daun Marigold (TDM) dan tepung bunga Marigold (TBM), P1: ransum mengandung 5% TDM dan 0% TBM , P2: ransum mengandung 3,75% TDM dan 1,25% TBM, P3: ransum mengandung 2,50% TDM dan 2,50% TBM, P4: ransum mengandung 1,25% TDM dan 3,75 TBM, dan P5 : ransum mengandung 0% TDM dan 5% TBM. Variabel yang diamati dalam penelitian ini adalah uji organoleptik warna daging (sebelum dimasak), bau daging, rasa daging serta tekstur daging. Hasil penelitian menunjukkan bahwa, skor warna daging P0 (2,88) lebih rendah dibanding skor warna daging dari P1-P5 dengan kisaran (3,00-3,63). Skor bau daging yang paling tinggi adalah pada P5 (30,21%). Skor rasa daging yang paling tinggi adalah P3 (46,88%). Skor nilai tekstur daging yang paling tinggi adalah P1 (37,50%). Berdasarkan hasil penelitian dapat disimpulkan bahwa pemberian tepung daun Marigold dan tepung bunga Marigold. Pada level pemberian 5% TDM dalam ransum lebih meningkatkan warna daging, pemberian 5,% TBM dalam ransum lebih meningkatkan bau daging, pemberian 2,50% TDM dan 2,50% TBM dalam ransum lebih meningkatkan rasa daging, dan pemberian 5% TDM dalam ransum lebih meningkatkan tekstur daging. Dengan demikian pemberian TDM dan TBM dapat diaplikasikan tanpa menurunkan sifat organoleptiknya

The objective of the study was to extract the oleoresin from Marigold with supercritical CO2 (SC-CO2) at different pressures and temperatures, detect the fatty acid composition by gas chromatography-mass spectrometry and investigate the antioxidative components in the extracts by combined online high-performance liquid chromatography-2,2-azinobis-(3-ethylbenzothiazolin-6-sulfonic acid (HPLC-ABTS·+) post-column assay and HPLC-tandem mass spectrometry. For the pressure range (20-40 MPa) and temperature range (30-70 °C), 30 MPa/70 °C gave the highest yield of oleoresin (58.9 g kg–1). The dominant fatty acids of Marigold flower oleoresin were linoleic acid (>26.41%), palmitic acid (>24.22%) and oleinic acid (>20.12%). Significant effects of the extraction pressure and temperature on the antioxidant activity were observed (P < 0.05). Lutein esters, α-tocopherol, β-tocopherol, γ-tocopherol and δ-tocopherol were the dominant antioxidant compounds in the extracts. The study has shown that the yield and total antioxidant activity of the Marigold extracts were affected by the pressure and temperature of SC-CO2, and that online HPLC technique could be used as an efficient and rapid method for separation and identification of bioactive compounds from a complex mixture.

The purpose of this research was to investigate the use of vermicompost at different points in the production cycle of tomato, Marigold, pepper, and cornflower. The incorporation of vermicompost of pig manure origin into germination media up to 20% v/v enhanced shoot and root weight, leaf area, and shoot:root ratios of both tomato and French Marigold seedlings; however amendment with vermicompost had little influence on pepper and cornflower seedling growth. Moreover there was no effect on the germination of seed of any species. When seedlings of tomato, French Marigold, and cornflower were transplanted into 6-cell packs there was greater plant growth in media amended with vermicompost compared to the control media, and the greatest growth when vermicompost was amended into both the germination and transplant media. This effect was increased when seedlings in the transplant media were irrigated with water containing fertilizer.

Bioresource Technology, 2002Co-Authors: Rola M Atiyeh, Norman Q Arancon, Clive A Edwards, James D MetzgerAbstract:The effects of additions of earthworm-processed pig manure (vermicompost) on the growth and productivity of French Marigold (Tagetes patula) plants were evaluated under glasshouse conditions. Marigolds were germinated and grown in a standard commercial greenhouse container medium (Metro-Mix 360), substituted with 10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90%, and 100% (by volume) pig manure vermicompost. The control consisted of Metro-Mix 360 alone without vermicompost. Plants were supplied regularly with a complete mineral nutrient solution for 121 days. The greatest vegetative growth resulted from substitution of Metro-Mix 360 with 30% and 40% pig manure vermicompost, and the lowest growth was in the potting mixtures containing 90% and 100% vermicompost. Most flower buds occurred in the potting mixtures containing 40% pig manure vermicompost (19.4 buds), and fewest in the potting mixtures containing 100% vermicompost. Marigolds grown in Metro-Mix 360 substituted with 90% and 100% pig manure vermicompost had the fewest and smallest flowers. After substitution of Metro-Mix 360 with 20%, 30%, 40%, 50%, 60%, 70%, 80%, and 90% vermicompost, the Marigold roots were larger than those of plants grown in the Metro-Mix 360 controls. Substitution of Metro-Mix 360 with any concentration of pig manure vermicompost, with all needed nutrients provided, increased the overall nitrate-nitrogen concentrations of the Marigold leaf tissues at flowering stage. Some of the Marigold growth and productivity enhancement, resulting from substitution of Metro-Mix 360 with pig manure vermicompost, may be explained by nutritional factors; However, other, factors, such as plant-growth regulators and humates, might have also been involved since all plants were supplied regularly with all required nutrients.

Half of the treatments were watered with liquid inorganic fertilizer while the other half received only water. Germination rates of tomato, pepper, lettuce, and Marigold seeds in the coir/perlite mixture did not differ significantly from that in Metro-Mix 360. However, the germination rate of tomato, pepper and lettuce seedlings was very low in the peat/perlite mixture. Substituting some of the peat/perlite mixtures with equal amounts of vermicomposts, particularly pig manure vermicompost, enhanced germination rates greatly, making it comparable to that in the commercial medium (Metro-Mix 360). Marigold as a carotenoid source on pigmentation and growth of red swordtail xiphophorus helleri

In captive conditions the fish show fade colouration and fail to get attracted by the buyers. To overcome the problem we have planned an experiment with the cheap pigmenting source namely the Marigold petal meal. The results are encouraging so that Marigold petal meal could be used as a pigmenting source at a concentration of 15 g / 100 g feed of dry weight. Leaching of pigmenting source is also a problem. Hence, experiment was conducted with Marigold petal enriched gelatine based diet. From the present investigation, it is considered that a gelatine based diet enriched with both Marigold petal powder and protein ingredients might give a better result of growth and pigmentation.
