PSU-F11, whereas five known compounds: 4-hydroxybenzoic acid ( 4 ), aplysiopsene D ( 5 ), 3-isochromanone ( 6 ), (−)-drimenin ( 7 ) and diketopiperazine derivative ( 8 ), were obtained from the fungus Nigrospora sp. PSU-F12. Their structures were established by spectroscopic evidence. We also tested their cytotoxic (on African green monkey kidney fibroblast and breast cancer cells), antioxidant (in the DPPH assay), and antibacterial (against the standard Staphylococcus aureus ATCC 25923 and methicillinresistant S. aureus ) activities.

CBMAI 1328 and Arthopyrenia sp. CBMAI 1330 isolated from marine sponge to synthesise laccases (Lcc) in the presence of the inducer copper (1–10 μM) was assessed. In a liquid culture medium supplemented with 5 μM of copper sulphate after 5 days of incubation, Nigrospora sp. presented the highest Lcc activity (25.2 U·L−1). The effect of copper on Lcc gene expression was evaluated by reverse transcriptase polymerase chain reaction. Nigrospora sp. showed the highest gene expression of Lcc under the same conditions of Lcc synthesis. The highest Lcc expression by the Arthopyrenia sp. was detected at 96 h of incubation in absence of copper. Molecular approaches allowed the detection of Lcc isozymes and suggest the presence of at least two undescribed putative genes. Additionally, Lcc sequences from the both fungal strains clustered with other Lcc sequences from other fungi that inhabit marine environments.

METHODS AND RESULTS While screening for bioactive secondary metabolites from endophytic fungi, we found that A. Nigrospora JS-1675 showed strong in vitro antibacterial activity against Ralstonia solanacearum. One compound (1) was isolated and identified as (4S, 5S, 6S)-5,6-epoxy-4-hydroxy-3-methoxy-5-methyl-cyclohex-2-en-1-one. Growth of most of the tested phytopathogenic bacteria was inhibited by compound 1 and the ethyl acetate (EtOAc) layer except Pseudomonas syringae pv. lachrymans. Compound 1 also inhibited the mycelial growth of several plant pathogenic fungi. Both compound 1 and the EtOAc layer reduced bacterial leaf spot disease in detached peach leaves. They also suppressed the development of bacterial wilt on tomato seedlings quite effectively. CONCLUSIONS Amphirosellinia Nigrospora JS-1675 showed antimicrobial activity against plant pathogenic bacteria and fungi by producing compound 1. SIGNIFICANCE AND IMPACT OF THE STUDY This is the first report on the occurrence of compound 1 in A. Nigrospora JS-1675 and its efficacy against plant pathogenic bacteria and fungi. Their strong disease control efficacy against tomato bacterial wilt suggests that this fungus can be used as a microbial bactericide.

Its inhibitory activities against sporangium and zoospore germination of P infestans were similar to those against Phytophthora capsici. In vivo, at 100 and 500 mg litre-1, it reduced the development of tomato late blight caused by P infestans.

molecular characterization of a novel partitivirus and a fusarivirus coinfecting the fungus Nigrospora sphaerica
Archives of Virology, 2021Co-Authors: Jie Zhong, Ze Zhong Yang, Xin Yang, Zhao Jiang Guo, Wen Xie, Youjun ZhangAbstract:In this report, we describe the molecular characterization of two novel mycoviruses coinfecting the plant pathogenic fungus Nigrospora sphaerica, which were designated "Nigrospora sphaerica fusarivirus 1" (NsFV1) and "Nigrospora sphaerica partitivirus 1" (NsPV1). NsFV1 has an undivided genome measuring 6,147 nt, excluding the polyA tail, and was predicted to contain two nonoverlapping open reading frames (ORF1 and 2). The larger ORF1 encodes a polyprotein containing a conserved RNA-dependent RNA polymerase (RdRp) and a helicase domain that has functions related to RNA replication, and the smaller ORF2 encodes a putative protein with an unknown function. NsPV1 consists of two genome segments, which measure 1,796 bp and 1,455 bp in length. Each of the two dsRNAs has a single ORF, and they are predicted to encode proteins with homology to viral RdRps and coat proteins of members of the family Partitiviridae. Phylogenetic analysis indicated that NsFV1 is a member of the recently proposed family "Fusariviridae", while NsPV1 was determined to belong to the genus Gammapartitivirus in the family Partitiviridae. To the best of our knowledge, this report is the first to describe mycoviruses infecting N. sphaerica.

Viruses, 2019Co-Authors: Chang Xin Li, Qian Zhou, Hui Wang, Jie ZhongAbstract:Three dsRNAs, in sizes of approximately 2.5–5 kbp, were detected in the plant pathogenic fungus Nigrospora oryzae strain CS-7.5-4. Genomic analysis showed that the 5.0 kb dsRNA was a victorivirus named as Nigrospora oryzae victorivirus 2 (NoRV2). The genome of NoRV2 was 5166 bp in length containing two overlapping open reading frames (ORFs), ORF1 and ORF2. ORF1 was deduced to encode a coat protein (CP) showing homology to the CPs of viruses belonging to the Totiviridae family. The stop codon of ORF1 and the start codon of ORF2 were overlapped by the tetranucleotide sequence AUGA. ORF2 was predicted to encode an RNA-dependent RNA polymerase (RdRp), which was highly similar to the RdRps of victoriviruses. Virus-like particle examination demonstrated that the genome of NoRV2 was solely encapsidated by viral particles with a diameter of approximately 35 nm. The other two dsRNAs that were less than 3.0 kb were predicted to be the genomes of two mitoviruses, named as Nigrospora oryzae mitovirus 1 (NoMV1) and Nigrospora oryzae mitovirus 2 (NoMV2). Both NoMV1 and NoMV2 were A-U rich and with lengths of 2865 and 2507 bp, respectively. Mitochondrial codon usage inferred that each of the two mitoviruses contains a major large ORF encoding a mitoviral RdRp. Horizontal transfer experiments showed that the NoMV1 and NoMV2 could be cotransmitted horizontally via hyphal contact to other virus-free N. oryzae strains and causes phenotypic change to the recipient, such as an increase in growth rate. This is the first report of mitoviruses in N. oryzae.

The genome of NoPV1 contains two dsRNA segments (dsRNA1 and 2), 1875 bp and 1601 bp in length, respectively. dsRNA1 and -2 both have a single open reading frame encoding the RNA-dependent RNA polymerase and coat protein, respectively. NoPV1 has a high degree of sequence identity to members of genus Gammapartitivirus in the family Partitiviridae. This is the first report of a mycovirus in the family Partitiviridae that infects N. oryzae.

Excluding a poly (A) tail, the genome of the virus is 7004 nucleotide (nt) long containing three putative nonoverlapping open reading frames (ORF1, ORF2, and ORF3). The large ORF1 encodes a polypeptide with a conserved RNA-dependent RNA polymerase (RdRp) domain and a helicase domain that functions for RNA replication. Each of the smaller ORF2 and the smallest ORF3 encodes a putative protein with an unknown function. Amino acid (aa) sequence similarities between the NoFV1-ORF1- and ORF2-encoded proteins and the homologous sequences from other mycoviruses were found. Phylogenetic analysis on the basis of the RdRp and helicase domains showed that NoFV1 is phylogenetically related to viruses in the newly proposed family Fusariviridae. Thus, we suggest that NoFV1 might be a novel member of family Fusariviridae.

In this study, complete sequence of a novel mycovirus from N. oryzae was reported. The viral genome is 5100 base pairs in length and possesses two overlapping open reading frames (ORFs). The two ORFs potentially encode proteins that showed significant similarity to the capsid protein and RNA-dependent RNA polymerase, in the family Totiviridae, respectively. Phylogenetic tree showed that this novel mycovirus is a new member of the genus Victorivirus in the family Totiviridae. We here designated the virus as Nigrospora oryzae victorivirus 1 (NoRV1), the first putative victorivirus identified in N. oryzae.
