Marine Pollution Bulletin, 2002Co-Authors: Rueyan Doong, Chinkai Peng, Peilin LiaoAbstract:The contamination of Organochlorine Pesticides (OCPs) in sediments from the Wu-Shi River estuary was investigated to evaluate the pollution potentials and distribution of OCPs in central Taiwan. A total of 19 sediment samples were collected at five sampling stations along the River estuary. The concentrations of OCPs were in the range of 0.99–14.5 ng/g-dry weight (dw) for ΣHCH (α-, β-, γ-, δ-HCH), 0.46–13.4 ng/g-dw for Σcyclodiene and 0.53–11.4 ng/g-dw for ΣDDT (p,p′-DDD, p,p′-DDE, p,p′-DDT). The mean concentrations of ΣHCH, Σcyclodiene and ΣDDT were 3.79, 4.87 and 2.51 ng/g-dw, respectively. The total concentrations of OCPs correspond to 1.73–71.9 μg/g-OC when normalized to TOC contents. Among the Organochlorine Pesticides, endosulfan sulfate, β-HCH, and p,p′-DDD were the most dominant compounds in the sediments with the average concentrations of 1.97, 3.43 and 2.08 ng/g, respectively. Also, different contamination patterns among sampling seasons were observed. The measured concentrations of OCPs collected in spring were higher than those in autumn and winter. A linear relationship between sediment characteristics and OCP residues was also demonstrated. The results obtained in this study show that there still exist a variety of Organochlorine Pesticide residues in the sediments from the near shore of central Taiwan.

Dietary intake and residues of Organochlorine Pesticides in foods from Hsinchu, Taiwan.
Journal of AOAC International, 1999Co-Authors: Rueyan DoongAbstract:: The levels of contamination with various Organochlorine Pesticides (such as total HCH, heptachlor, heptachlor epoxide, aldrin, dieldrin, endrin, endosulfan, and total DDT) of different foods from 3 traditional markets were determined to estimate Taiwanese daily intake of Organochlorine Pesticides. Of the 18 Organochlorine Pesticides investigated, alpha-HCH, beta-HCH, lindane, delta-HCH, heptachlor, heptachlor epoxide, dieldrin, endrin, alpha-endosulfan, p,p'-DDE, and p,p'-DDT were detected at concentrations ranging from 0.26 to 10.2 ng/g wet weight. Contamination with Organochlorine Pesticides followed the order heptachlor > dieldrin > alpha-endosulfan > HCH isomers > heptachlor epoxide > DDT. Frequencies of detection of Organochlorine Pesticide residues ranged from 2.0 to 52.3%. alpha-Endosulfan was the most frequently detected Organochlorine Pesticide in the foods analyzed, followed by heptachlor epoxide (47.6%) and alpha-HCH (38.9%). Estimated daily intakes (EDIs) of Organochlorine Pesticides from foods were 1.137 micrograms for total HCH, 2.147 micrograms for heptachlor, 0.702 microgram for heptachlor epoxide, 0.624 microgram for endosulfan, 0.098 microgram for cyclodiene, and 0.541 microgram for total DDT. These EDIs were only 0.075% of the acceptable daily intake (ADI) for lindane, 47.5% of ADI for heptachlor and heptachlor epoxide, 0.045% of ADI for total DDT, and 1.01% of ADI for aldrin and dieldrin. Therefore, consumption of the foods analyzed does not pose a risk to consumer health.

Analyst, 1999Co-Authors: Rueyan DoongAbstract:Fourteen Organochlorine Pesticide residues in fatty foods were determined using a simple and rapid procedure based on solid-phase extraction (SPE) clean-up cartridges with octadecyl (C18)-bonded porous silica, a tandem C18 and Florisil column, Alumina-N and Florisil. A Florisil cartridge eluted with 12 ml petroleum ether–ethyl ether (95 + 5) was the most efficient clean-up procedure capable of eliminating the matrix interference and satisfying the agreed acceptable recovery for the large numbers of Organochlorine Pesticides in nine kinds of foods having different fat contents. Average recoveries of Organochlorine Pesticides in shellfish, fish and meats ranged from 77 to 105%, 84 to 98% and 85 to 107%, respectively. In addition, analysis of a certified Standard Reference Material (SRM 1945) verified the satisfactory performance of Florisil clean-up cartridge. This SPE method not only yielded comparable results for nonfatty foods, but also provided a reliable separation and quantification of Organochlorine Pesticides for analyzing a large number of foods with a wide range of fat content.

Previous work showed that developmental dieldrin exposure increased neuronal susceptibility to MPTP toxicity in male C57BL/6 mice, possibly via changes in dopamine (DA) packaging and turnover. However, the relevance of the MPTP model to PD pathophysiology has been questioned. We therefore studied dieldrin-induced neurotoxicity in the α-synuclein (α-syn)-preformed fibril (PFF) model, which better reflects the α-syn pathology and toxicity observed in PD pathogenesis. Specifically, we used a “two-hit” model to determine whether developmental dieldrin exposure increases susceptibility to α-syn PFF-induced synucleinopathy. Dams were fed either dieldrin (0.3 mg/kg, every 3–4 days) or vehicle corn oil starting 1 month prior to breeding and continuing through weaning of pups at postnatal day 22. At 12 weeks of age, male and female offspring received intrastriatal α-syn PFF or control saline injections. Consistent with the male-specific increased susceptibility to MPTP, our results demonstrate that developmental dieldrin exposure exacerbates PFF-induced toxicity in male mice only. Specifically, in male offspring, dieldrin exacerbated PFF-induced motor deficits on the challenging beam and increased DA turnover in the striatum 6 months after PFF injection. However, male offspring showed neither exacerbation of phosphorylated α-syn aggregation (pSyn) in the substantia nigra (SN) at 1 or 2 months post-PFF injection, nor exacerbation of PFF-induced TH and NeuN loss in the SN 6 months post-PFF injection. Collectively, these data indicate that developmental dieldrin exposure produces a male-specific exacerbation of synucleinopathy-induced behavioral and biochemical deficits. This sex-specific result is consistent with both previous work in the MPTP model, our previously reported sex-specific effects of this exposure paradigm on the male and female epigenome, and the higher prevalence and more severe course of PD in males. The novel two-hit environmental toxicant/PFF exposure paradigm established in this project can be used to explore the mechanisms by which other PD-related exposures alter neuronal vulnerability to synucleinopathy in sporadic PD.

Levels and distribution pattern of Organochlorine Pesticide residues in eggs of 22 terrestrial birds from Tamil Nadu, India

Bird eggs have several advantages than other environmental matrixes, which have been used extensively to portray the accumulation and distribution of OCPs. The present study investigated the Organochlorine Pesticide (OCP) residues in eggs of 22 species of terrestrial birds collected from Tamil Nadu, India. Eggs found abandoned were collected during nest monitoring between 2001 and 2008 and analyzed for the presence of Organochlorine Pesticide residues. The results showed that the mean concentrations of total hexachlorohexane (∑HCHs), total dichlorodiphenyltrichloroethane (∑DDTs), heptachlor epoxide, and dieldrin ranged from non-detectable (nd) to 2800 ng/g, nd to1000 ng/g, nd to 700 ng/g, and nd to 240 ng/g on a wet mass (wm) basis, respectively. The variation in magnitude of contamination among the species and feeding guilds were not significantly different ( p > 0.05). Among the OCPs analyzed, the residues of β- HCH and p,p ′-DDE were found to be the abundant in concentration. Similarly, among various bird species studied, the highest concentrations of ∑OCPs (> 5000 ng/g wm) were recorded in the eggs of gray junglefowl, scaly-breasted munia, and red-whiskered bulbul. This may be due to their widespread occurrence of their habitat at proximity to the agricultural fields, where Organochlorines were in use until recently. Among the various contaminants analyzed, concentrations of p,p ′-DDE and heptachlor epoxide exceeded the threshold levels of toxicity for wild birds in > 5% of the egg samples. Hence, this study indicates the need for continued monitoring and further systematic ecotoxicological investigation of these compounds not only in eggs but also in other environmental media.

Science of The Total Environment, 2003Co-Authors: Susana Cruz, C M Lino, Maria Irene Noronha Da SilveiraAbstract:Organochlorine Pesticide residues were measured in human serum from an urban and two rural populations in Portugal, in an attempt to evaluate the contamination level of Portuguese population. Serum levels of 12 residues were determined using a validated methodology that included gas chromatography-electron-capture detection. It was made an attempt to point out the differences of contamination between rural and urban populations; and among these, if it could be established a relation with sex and with age of individuals. p,p′DDE, α-hexachlorocyclohexane (HCH), p,p′DDD, and β-HCH were the most frequently identified residues. p,p′DDE concentrations ranged from undetected to 390.5 μg/l in urban samples, and from undetected to 43.5 μg/l and to 171.2 μg/l in both rural samples. Maximum α-HCH concentration level was 114.4 μg/l in urban samples, 261.3 and 45.5 μg/l in both rural samples. Mean total DDT levels were always higher than mean total HCH levels. About p,p′DDE, in all three populations, the majority of the results above the limit of quantification were found among female sex. The analysis of different aged groups showed that younger groups continue to reveal contamination. Comparing obtained results with others from Europe, Asia and America, it was observed that Portugal is between the highest levels of contamination.
