RSC Advances, 2014Co-Authors: Rui Wang, Li Yang, Jacquelyn Runco, Rui Chen, Zhengtao WangAbstract:A novel comprehensive method using supercritical fluid chromatography-Photodiode Array detector-mass spectrometry (SFC-PDA-MS) was developed for the qualitative and quantitative analyses of a chiral isomer pair in Ban Lan Gen (dried root of Isatis indigotica) and the processed product. A standard goitrin–epigoitrin racemic mixture was used for method development. A total of six chiral stationary phases (CSPs) were screened and the (S,S)-Whelk-O 1 (4.6 × 250 mm, 10 μm) column was chosen as it offers baseline resolution for the enantiomeric pair with separation accomplished in 6 minutes. A single quadrupole MS and a PDA detector were used in line for the detection. The validated method was applied successfully in the analysis of different samples. Results indicated that the developed assay was fast, sensitive and reproducible. SFC should be an integral part of the overall analytical platform for TCM and natural product research, especially in the area of chiral analysis.

International Journal of Molecular Sciences, 2012Co-Authors: Rui Wang, Yiming Li, Shanjun Huang, Zhengtao WangAbstract:A simple and reliable method of ultra-performance liquid chromatography with Photodiode Array detector (UPLC-PDA) was developed to control the quality of Radix Isatidis (dried root of Isatis indigotica) for chemical fingerprint analysis and quantitative analysis of eight bioactive constituents, including R,S-goitrin, progoitrin, epiprogoitrin, gluconapin, adenosine, uridine, guanosine, and hypoxanthine. In quantitative analysis, the eight components showed good regression (R > 0.9997) within test ranges, and the recovery method ranged from 99.5% to 103.0%. The UPLC fingerprints of the Radix Isatidis samples were compared by performing chemometric procedures, including similarity analysis, hierarchical clustering analysis, and principal component analysis. The chemometric procedures classified Radix Isatidis and its finished products such that all samples could be successfully grouped according to crude herbs, prepared slices, and adulterant Baphicacanthis cusiae Rhizoma et Radix. The combination of quantitative and chromatographic fingerprint analysis can be used for the quality assessment of Radix Isatidis and its finished products.

In total 14 monacolin compounds such as monacolin K (mevinolin), J, L, M, X, and their hydroxy acid form, as well as dehydromonacolin K, dihydromonacolin L, compactin, 3α-hydroxy-3,5-dihydromonacolin L, etc. were identified in red yeast rice, using high-performance liquid chromatography with Photodiode Array detector and tandem mass spectrometry. A chemical fingerprint profiling method to display bioactive monacolins in red yeast rice was established and could be used for the quality control of the target material and its related products. Ten finish products labeled as red yeast rice from different manufacturers in marketing were traced using the chromatographic chemical profiling method, and the results show that only two of them were similar while the other eight were significantly different from the reference red yeast rice. All of these materials including raw material powder and finished products available were quantified and the contents of monacolins were calculated with reference of monacolin K (mevinolin) as the standard.

The separation was achieved on a C(18) column with a gradient solvent system of 0.08% aqueous formic acid solution-acetonitrile. The compounds were identified with MS-spectra and authentic standards. The contents of jujuboside A, jujuboside B, spinosin and 6'''-feruloyl spinosin were quantified by using DAD in different extract of zizyphi spinosi semen.

Linearity of around three orders in the magnitude of concentration was generally obtained and limits of detection for these compounds were in the range of 30–90 pg.

Caffeic acid derivatives and alkamides produced very abundant peaks in the total ion current chromatogram during negative and positive cone voltage switching. Cichoric acid and the isomer pair, dodeca-2E,4E,8Z,10E/Z-tetraenoic acid isobutylamide, were used as a standard for quantification of caffeic acid derivatives and alkamides in E. purpurea. This novel method surpasses previously published ones in product quality control and providing the HPLC chromatographic fingerprints of biological active components in E. purpurea.

Fifteen flavonoids were identified by high-performance liquid chromatography with Photodiode Array detection/mass spectrometry. Among them, two anthocyanins and nine flavonols were discovered in lotus petals for the first time. This work is valuable for both the hybrid breeding on lotus oriented to flower color and the utilization of lotus petals as functional food materials.
