    Abstract:

    Homoveratric acid (HVA) degradation was observed in cultures of Pleurotus eryngii lacking lignin peroxidase (LiP) activity. Extracellular enzymes seemed responsible for this transformation, and the lack of activity after ultrafiltration of the culture liquid suggests that the presence of some low-molecular-size compounds is required. This hypothesis is supported by rapid HVA transformation after addition of the synthetic laccase substrate 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) to the ultrafiltered liquid. HVA transformation by the extracellular enzymes from P. eryngii takes place via C_α-C_β breakdown and formation of veratryl alcohol, which is further transformed into veratraldehyde. The same major compounds were found during HVA transformation by LiP from Phanerochaete chrysosporium , but this reaction was not stimulated by ABTS. Although the involvement of other enzymes cannot be ruled out, purified laccase from Pleurotus eryngii caused the same HVA transformation pattern in presence of ABTS. Moreover, veratryl alcohol oxidation by P. eryngii laccase was demonstrated in the presence of ABTS. These results suggest that enzymatic systems lacking LiP could be responsible for natural degradation of lignin.

    Considering the hysteresis of switch timing control system used in the industrial farming of Pleurotus eryngii, respiration control experiments were carried out with given temperature, humidity and light in a factory farm. Planting environment was monitored by a remote monitor system with a radio frequency module. A CO 2 concentration prediction model is developed based on BP neural network and a cycle control strategy for sporocarp of Pleurotus eryngii. This model provides a reference for the precise control of factory-farming Pleurotus eryngii, so as to improve the quality and increase the production of Pleurotus eryngii.

    Aiming at the defects of the existing technology for Pleurotus eryngii cultivation environment monitoring system in Ningxia Pengyang County, such as: transmission of the data is not stable, small monitoring range and the sensors cannot move conveniently, only the environmental data in a fixed position within each mushroom shed can be collected, need to lay a lot of cables between the collector and the sensors, wiring difficulty, high cost etc., this system is designed a low-cost remote monitoring of digital information platform to automatically, accurately, and remote wirelessly monitor the environmental temperature, humidity, light intensity and carbon dioxide concentration in eight Pleurotus greenhouse simultaneously, based on single-chip, the integrated application of multi sensor fusion technology, Bluetooth wireless transmission technology, GPRS wireless remote transmission technology, combined with the wireless RF module NRF905 set up a tree network structure, the PC monitor centre was developed based on the software LabVIEW. The environmental control equipment heater, refrigerator, fan and shower facilities in each Pleurotus shed can be manually and automatically controlled remotely. The real-time environmental data can also be stored and managed as EXCEL files. This study provides effective technical support for establishing the growth model of Pleurotus eryngii and forming the control strategy of saving energy, so as to improve the quality and yield of Pleurotus eryngii.

    Pleurotus eryngii was transformed via restriction enzyme-mediated integration. In order to construct the transformation plasmid, the enhanced cyan fluorescent protein (ECFP) gene was ligated next to the gpd promoter of the plasmid pAN7-1. Transformation was facilitated via the heat treatment of a transformation mixture containing 1 μg of the HindIII-digested plasmid DNA and 106 mushroom protoplasts in 40% polyethyleneglycol solution, resulting in 10–40 hygromycin-resistant transformants. Successful transformation was evidenced by PCR, Southern blot, and confocal fluorescence microscopic analyses on the selected transformants. To date, this is the first report on the transformation of P. eryngii by REMI technique.

    Abstract Polysaccharides were extracted from freeze-dried-, oven-dried- and boiling-treated Pleurotus eryngii (FDPEPS, ODPEPS and BTPEPS). Yield of FDPEPS and its total carbohydrates, total polysaccharides, reducing sugars, phenolics and protein content were higher than ODPEPS and BTPEPS. High performance liquid chromatography (HPLC) identified that FDPEPS, ODPEPS, and BTPEPS were composed of mannose (5.75%, 5.52%, 6.97%), glucose (88.90%, 89.31%, 87.68%) and galactose (5.34%, 5.17%, 5.35%). All three polysaccharides showed typical saccharic absorption bands in FT-IR. The FDPEPS showed the highest antioxidant activities in ferric reducing antioxidant power (FRAP), ABTS, superoxide anion and hydroxyl radical scavenging tests. Denser aggregates and larger serum pores were observed in confocal micrographs of soymilk added with ODPEPS.
