    Abstract:

    Objective To optimize the condition for opsonophagocytic assay(OPA)for evaluation of potency of Pneumococcus Polysaccharide conjugate vaccine.Methods The experimental condition for OPA was optimized according to the protocol for opsonophagocytic assay against streptococcus pneumoniae(version A.02,January 2007)published by WHO reference laboratory in US University of Alabama.The opsonophagocytic titers of drug-resistant and drug-sensitive Pneumococcus strains against the sera of mice immunized with Pneumococcus Polysaccharide conjugates of the corresponding types were determined and compared with the corresponding ELISA titers,between which the relationship was analyzed.Results After HL-60 cells were differentiated for 4 d under induction of 0.8% DMF,both the expression of cell surface markers CD11b and CD35 were not less than 55%,and that of CD71 was not more than 15%,and the proportion of live cells was not less than 65%.The recovery rate of working seed lot of Pneumococcus of various types were not more than 80%,while the non-specific killing rate of complement was 18%.All the serotypes showed high bactericidal activity,and similar correlation coefficients and slopes were observed between OPA and ELISA titers.Conclusions The optimized condition met the requirements for OPA and might be used for the evaluation of potency of Pneumococcus Polysaccharide conjugate vaccine.

    Abstract:

    Streptococcus pneumoniae (Pneumococcus) Polysaccharide serotype-specific antibodies that have opsonophagocytic activity are considered a primary mechanism of host defense against pneumococcal disease. In vitro opsonophagocytic assays (OPAs) with antibody and complement to mediate opsonophagocytic killing of bacteria have been designed and developed as an adjunct to the standardized serum immunoglobulin G antipneumococcal capsular Polysaccharide enzyme immunoassay to assess the effectiveness of pneumococcal vaccines. OPA presents challenges for assay standardization and assay precision due to the multiple biologically active and labile components involved in the assay, including human polymorphonuclear leukocytes or cultured effector cells, bacteria, and complement. Control of these biologically labile components is critical for consistent assay performance. An approach to validating the performance of the assay in accordance with International Conference for Harmonization guidelines, including its specificity, intermediate precision, accuracy, linearity, and robustness, is presented. Furthermore, we established parameters for universal reagents and standardization of the use of these reagents to ensure the interlaboratory reproducibility and validation of new methodologies.

    Abstract:

    Objective To explore the relationship between Polysaccharide chain length and immunogenicity of Pneumococcus Polysaccharide-protein conjugate vaccine types 1 and 14 so as to solve the problem that Polysaccharide conjugate is difficult to be puri- fied.Methods Hydrolyze the Polysaccharides of Pneumococcus types 1 and 14 into oligosaccharides with acid and conjugate to tetanus toxoid(Tr)by the same method as that for preparation of Polysaccharide-TT conjugate.Immunize mice with the prepared oligosaccha- ride-TT(OS-TT)and Polysaccharide-TT(PS-TT)conjugates respectively and determine the serum antibody level.Results High antibody levels were induced in the mice immunized with both OS-Tr and PS-TT,and immunopotentiatin was observed.OS-TT showed no significant difference with PS-TT in immunogenicity.Conclusion The acid hydrolysis of polysaecharide of Pneumococcus types 1 and 14 showed no significant influence on the immunogenicity of Polysaccharide-protein conjugate vaccine.
