    Background: Cypress Pollen allergy is a major cause of rhinoconjunctivitis and asthma in the Mediterranean area. The nonstandardized cypress allergen Extracts currently available for the diagnosis of cypress allergy have a low level of activity. The search for an active material has led to the selection of Juniperus ashei (Ja) Pollen because of its very high cross-reactivity with cypress Extracts and its superior allergenic activity. The aim of this study was to characterize in vitro and calibrate in vivo an in-house reference Extract (IHRS) of J. ashei Pollen and determine the specificity and sensitivity of a standardized Ja Extract for the prick test diagnosis of cypress allergy. Methods:Juniperus ashei Pollen Extract was analysed by 2-D electrophoresis. The IHRS Ja Extract was calibrated by skin prick testing in 28 cypress-allergic patients. The sensitivity and specificity of cypress allergy diagnosis using a standardized Ja Extract was studied by skin prick test in 42 cypress-allergic patients and 53 nonallergic patients. Jun a 1 content of the IHRS was determined by a monoclonal antibody-based electrophoretic technique. Results: The Jun a 1 content of the 100 IR/ml Ja IHRS Extract was 180 μg/ml. For in vivo diagnosis of cypress allergy, Ja Pollen Extract demonstrated a sensitivity of 95%, a specificity of 100%, a negative predictive value of 96%, and a positive predictive value of 100%. Conclusion: Standardized Ja Pollen Extract is therefore a very appropriate tool for the in vivo diagnosis of cypress Pollen allergy and good candidate for specific immunotherapy.

    Background Calcium phosphate-adsorbed allergen Extracts are used for subcutaneous immunotherapy to avoid the use of aluminium adjuvants. Objectives A double-blind, placebo-controlled study was performed in order to confirm the safety and assess the efficacy of a standardized five-grass-Pollen Extract adsorbed onto calcium phosphate for specific immunotherapy (IT). Methods Twenty-nine patients with seasonal rhinoconjunctivitis were randomized to receive either the active preparation (16 patients) or placebo (13 patients), in a 1-year study. During the increasing dose phase, an Extract ranging from 0.1 IR per ml to 50 IR per ml was administered at a rate of one subcutaneous injection per week until a maintenance dose was reached. The patients were assessed by symptom diary and rescue medications during seasonal exposure and specific nasal and skin reactivity before and after IT. Immunological parameters (specific IgE and IgG 4 antibodies) were assessed before, during and after IT. Results The overall symptoms score (mean AUC) was not significantly different between the IT group and the placebo group during grass-Pollen exposure (49.6 vs. 56, respectively). The total medication score (mean AUC) was significantly lower in the IT group than in the placebo group (11 vs. 41, P < 0.01, Mann-Whitney U-test). The cumulative symptom/medication score was significantly lower in the IT group than in the placebo group (64.5 VS. 102.3, P < 0.05, U-test). A significant increase in nasal reactivity threshold was observed after IT in the IT group (21. 4 IR/mL before IT vs. 63.4 IR/mL after IT, P < 0.01, Wilcoxon), whereas no significant changes were observed in the placebo group (31.0 IR/mL before IT vs. 37.7 IR/mL after IT). IT induced a significant reduction in grass Pollen cutaneous reactivity in the actively treated group (P < 0.001). A significant increase in serum-specific IgG 4 antibody response was observed in the IT group (3.1% before IT VS. 10.1% after IT, P < 0.001). Nine patients in the IT group developed moderate immediate systemic reactions vs. two patients in the placebo group. Conclusion Specific immunotherapy with calcium phosphate-adsorbed standardized grass Pollen Extract was safe and effective for the treatment of patients with seasonal allergic rhinoconjunctivitis.

    Sublingual-swallow immunotherapy (SLIT) using high doses of standardized allergen Extracts has been found to be effective in reducing allergic symptoms and medication needs. A double-blind, placebo-controlled study was carried out in a large number of patients to determine whether medication needs can be reduced by SLIT. Some 136 patients with grass-Pollen rhinitis with or without mild asthma were studied. Patients received either placebo or SLIT with a standardized grass-Pollen Extract administered daily with increasing doses up to 300 IR (index of reactivity) from January to the end of July 1994. During the grass-Pollen season, patients were instructed to use medications as required and to visit their doctors in case of asthma. Symptom-medications scores were assessed during the Pollen season, and serum-specific IgG4 was measured before and at the end of SLIT. In the SLIT group, drug consumption dropped significantly throughout the Pollen season (P<0.02). Moreover, at the peak of the Pollen season, betamethasone consumption was significantly reduced in the SLIT group (P<0.02). Only one patient in the SLIT group had an asthma attack compared to eight patients in the placebo group (P<0.02). IgG4 levels increased significantly in the SLIT group (P<0.001) but without correlation with symptoms. Side-effects were comparable in both groups. This study indicates that SLIT in grass-Pollen rhinitis is well tolerated, improves overall clinical symptoms, and reduces drug consumption and the need for oral corticosteroids.

    Fifty-eight patients with well-documented history of seasonal rhinoconjunctivitis caused by grass Pollens were allocated randomly on a double-blind basis to receive either sublingual therapy with a solution of purified, standardized allergen preparation (Stallergenes) or a matched placebo for 17 weeks. The assessment of the effect of oral immunotherapy, done with drops of five-grass allergen Extract, was on the clinical symptoms and on the medication score of the authorized rescue treatments. The actively treated patients had significantly (P<0.05 to P<0.01) fewer symptoms of rhinitis (sneezing and rhinorrhea) and of conjunctivitis (redness and tears) during the Pollen season than the placebo group. Consumption of nasal solution of sodium cromoglycate and of betamethasone and dexchlorpheniramine was significantly less in the desensitized group (P<0.01). Side-effects were negligible. This study concludes that perlingual immunotherapy with grass Pollen Extract in grass-Pollen-sensitive seasonal hay fever and conjunctivitis patients is effective, easy to perform, inexpensive, and safe.

    Oxidative stress plays an important role in the pathogenesis of myocardial infarction (MI). Schisandra chinensis bee Pollen Extract (SCBPE) possesses powerful antioxidant capacity. This study aimed to further explore the antioxidative and cardioprotective effects of SCBPE on acute MI induced by isoprenaline (ISO) in rats. The rats were intragastrically administrated with SCBPE (600, 1200, or 1800 mg/kg/day) and Compound Danshen dropping pills (270 mg/kg/day) for 30 days, then subcutaneously injected with ISO (65 mg/kg/day) on the 29th and 30th day. Compared with the model group, pretreatment with middle and high doses of SCBPE significantly reduced serum aspartate transaminase, lactate dehydrogenase, and creatine kinase activities and increased myocardial superoxide dismutase, glutathione peroxidase, and catalase activities. The histopathologic aspects showed that pathological heart change was found in the model group and reduced to varying degrees in the SCBPE groups. Moreover, the protein expression of nuclear factor-erythroid 2-related factor 2 (Nrf-2), heme oxygenase-1 (HO-1), and Bcl2 in the heart increased in the SCBPE groups, while that of Bax decreased compared to the model group. Besides this, uridine was isolated from S. chinensis bee Pollen for the first time. This study could provide a scientific basis for using Schisandra chinensis bee Pollen as a functional food for the prevention of MI.

    Abstract Cisplatin (CP) has been used to cure numerous forms of cancers effectively in clinics, however, it could induce some toxic effects. Bee Pollen is a natural compound, produced by honey bees. It is obtained from collected flower Pollen and nectar, mixed with bee saliva. Bee Pollen produced from Schisandra chinensis plants is described to exert potent antioxidant effects and to be a free radical scavenger. The purpose of this study was to investigate the effects of therapeutic treatment with Schisandra chinensis bee Pollen Extract (SCBPE) on liver and kidney injury induced by CP. The rats were intragastrically administrated with different doses of SCBPE (400 mg/kg/day, 800 mg/kg/day, 1200 mg/kg/day) and vitamin C (400 mg/kg/day, positive control group) for 12 days, and the liver and kidney injury models were established by single intraperitoneal injection of CP (8 mg/kg) at seventh day. The effect of SCBPE on CP toxicity was evaluated by measuring markers of liver and kidney injury in serum, levels of lipid peroxidation and antioxidants in liver and kidney, observing pathological changes of tissue, and quantified expression of NFκB, IL-1β, IL-6, cytochrome C, caspase3, caspase9, p53 and Bax in liver and kidney. Compared with the model group, the activities of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and the content of blood urea nitrogen (BUN), creatinine (Cr) in serum all decreased in SCBPE high dose group. Meanwhile, the activities of superoxide dismutase (SOD), catalase (CAT) and the content of reduced glutathione (GSH) in liver and kidney increased, and the content of malondialdehyde (MDA) and inducible nitric oxide synthase (iNOS) decreased. In addition, the histopathologic aspects showed that the pathological changes of liver and kidney were found in the model group, and SCBPE group reduced to varying degrees. Moreover, the expression of NFκB, IL-1β, IL-6, cytochrome C, caspase3, caspase9, p53 and Bax in liver and kidney decreased. Therefore, SCBPE could reduce the damage of liver and kidney caused by CP by reducing the level of oxidative stress, and improving the antioxidant, anti-inflammatory and anti-apoptotic capacity of the body.

    Background Conventional allergen immunotherapy is clinically effective in reducing the symptoms of allergic rhinitis and asthma. It differs from other pharmacotherapies in that it can induce long-term clinical remission of these diseases. However, it requires years of treatment and is associated with serious allergic reactions. Objective To evaluate the safety, clinical efficacy, and immunologic mechanisms of immunotherapy with an oral, microencapsulated form of timothy grass allergen. Methods In this double-blind, placebo-controlled study, 24 patients aged 19 to 55 years with grass Pollen allergy were randomized to receive either microencapsulated timothy grass Pollen Extract or placebo once a day for 10 weeks. The dose of study drug was doubled weekly. Safety was evaluated through weekly visits, daily symptom diaries, and routine laboratory tests. Efficacy was evaluated by comparing medication use and symptoms scores during peak grass Pollen season before and after treatment. Allergen-specific T-cell responses, cytokine production, and IgG, IgE, and skin reactivity were measured to evaluate immunologic mechanisms. Results Eleven of 12 patients in the active treatment group had a decrease in the combined medication and symptom score, but only 4 of 10 patients in the placebo group had a decrease in scores. The proliferative response to timothy grass was reduced by at least 30% in 9 of the 12 grass-treated patients, but only 3 of 11 placebo patients had a proliferative response reduction. Timothy grass-induced interleukin-5 messenger RNA was reduced in the active group, but not in the placebo group. There were no significant changes in either group in IgG, IgE, and skin reactivity. Conclusions Oral immunotherapy with microencapsulated allergen induces a form of immunologic tolerance to the allergen and is a safe, efficient, and effective method of allergen immunotherapy.
