    Abstract:

    This study was carried out to evaluate the use of Lactobacillus acidophilus as a biocontrol Agent against some common ¢sh pathogenic bacteria (Staphylococcus xylosus, Aeromonas hydrophila gr.2 and Streptococcus agalactiae) in African cat¢sh, Clarias gariepinus. Eight treatments were designed inclusive of 10 C. gariepinus juveniles (mean weight 190 g) per tank, each in triplicate. Four groups of ¢sh were fed a diet supplemented with L. acidophilus, comprising about 3.01 � 10 7 colony-forming units per gram of diet (the Probiotics diet), while the other four groups were fed a diet not supplemented with Probiotics (the non-Probiotics diet). In the ¢rst group, ¢sh were injected with 1mL physiological saline and fed the non-Probiotic diet (non-Probiotic control); in the second, third and fourth groups, ¢sh were injected with 1mL each of S. xylosus, A. hydrophila gr.2 and S. agalactiae, respectively, and were all fed the non-Probiotic diet (designated as non-Probiotic treatments; NPsx, NPah and NPsa respectively). In the ¢fth group, ¢sh were injected with 1mL physiological saline but fed the Probiotic diet (Probiotic control), while ¢sh in the sixth, seventh and eighth groups were each injected with 1mL of S. xylosus, A. hydrophila gr.2 and S. agalactiae, respectively, and were all fed the Probiotic diet (and designated as Probiotic treatments; Psx, Pah and Psa respectively). Blood samples were collected for haematology analysis, while samples of the liver and kidney were examined for pathohistology after 7 and 21 days of infection. The results showed that the haematology parameters, packed cell volume, haemoglobin, erythrocyte sedimentation rate, red blood cell, white blood cell, total serum protein, Mg 21 ,C a 21 , Cl � , glucose, cholesterol and total immunoglobulin concentrations and the pathohistology of the liver and kidney were better in the challenged ¢sh (infected) maintained on the Probiotic diet than those in the groups fed the non-Probiotic diet. It is concluded, based on these results, that L. acidophilus is useful as a Probiotic Agent in C. gariepinus against these pathogenic bacteria (S. xylosus, A. hydrophila gr.2 and S. agalactiae).

    This experiment was carried out to evaluate the effects of the Probiotic, Lactobacillus acidophilus, on the growth performance, haematology parameters and immunoglobulin concentration in African catfish Clarias gariepinus fingerling. Two experimental diets were formulated to contain 35 g kg−1 crude protein and 10 g kg−1 lipids accordingly and fed three times daily for 12 weeks to 25 C. gariepinus fingerlings per fibreglass tank in 12 replicates each. The control diet was prepared with no Probiotic supplementation whereas the second diet was prepared supplemented with a Probiotic, L. acidophilus, containing about 3.01 × 107 colonies/g of diet. The results show that growth performance [specific growth rate (SGR) and relative growth rate (RGR)], nutrient utilization [protein efficiency ratio (PER) and feed conversion ratio (FCR)] and survival were significantly (P 0.05). From the results of this experiment, we conclude that L. acidophilus can be used as a Probiotic Agent in African catfish culture, to enhance fish health, survival and better feed efficiency and growth performance.

    Abstract:

    European foulbrood is a contagious bacterial disease of honey bee larvae. Studies have shown that the intestinal bacteria of insects, including honey bees, act as Probiotic organisms. Microbial flora from the gut of the Japanese honey bee, Apis cerana japonica F. (Hymenoptera: Apidae), were characterized and evaluated for their potential to inhibit the growth of Melissococcus plutonius corrig. (ex White) Bailey and Collins (Lactobacillales: Enterococcaceae), the causative Agent of European foulbrood. Analysis of 16S rRNA gene sequences from 17 bacterial strains isolated by using a culture-dependent method revealed that most isolates belonged to Bacillus, Staphylococcus, and Pantoea. The isolates were screened against the pathogenic bacterium M. plutonius by using an in vitro growth inhibition assay, and one isolate (Acja3) belonging to the genus Bacillus exhibited inhibitory activity against M. plutonius. In addition, in vivo feeding assays revealed that isolate Acja3 decreased the mortality of honey bee larvae infected with M. plutonius, suggesting that this bacterial strain could potentially be used as a Probiotic Agent against European foulbrood.

    Bifidobacteria were isolated from the intestinal tract of the Japanese honeybee, Apis cerana japonica, and investigated for potential application as a Probiotic Agent against Melissococcus plutonius, the causal Agent of European foulbrood (EFB), based on the findings of in vitro inhibition assays. A total of 11 bifidobacteria strains (designated as AcjBF1-AcjBF11) were isolated using a culture-dependent method and their 16S rRNA gene sequences were analyzed. The AcjBF isolates belonged to three distinct bifidobacterial phylotypes that were similar to those found in the European honeybee, Apis mellifera. Although the Japanese and European honeybees are distinct species with different traits and habits, the observation that they share highly similar bifidobacterial phylotypes suggests that bifidobacteria are conserved among honeybee species. Despite having extremely high 16S rRNA gene sequence similarities, the AcjBF isolates had markedly different carbohydrate fermentation profiles. In addition, in vitro growth inhibition assays revealed that the cell-free supernatants of all AcjBF isolates exhibited antagonistic effects on M. plutonius growth. These results indicate that the bifidobacteria isolated from the gut of Japanese honeybee could potentially be employed as a new biological Agent to control EFB.

    Abstract:

    Antibiotic associated diarrhea due to human intestinal microbiota abnormalities is a side effect of H. pylori eradication therapy. We examined intestinal microbiota changes during H. pylori eradication therapy and the preventive effect of CBM588 as a Probiotic Agent. Nineteen patients with gastro-duodenal ulcer were randomly divided into three groups: group A (without Probiotics), group B (with regular doses of CBM588) and group C (with double doses of CBM588). The incidence of diarrhea and soft stools during H. pylori eradication therapy was 43% in group A and 14% in group B, while none of the patients in group C reported diarrhea or soft stools. Both bacterial counts and detection rates of bifidobacteria and/or obligate anaerobe were decreased by eradication therapy. However, bacterial counts of obligate anaerobes in group C were significantly higher than in group A (P < 0.05). Additionally, during eradication therapy C. difficile toxin A was detected in both group A and group B but not in group C. In conclusion, these results indicate that H. pylori eradication therapy induces antibiotic associated diarrhea due to abnormalities in intestinal microbiota and/or C. difficile. However, these side effects might be prevented by Probiotics.

    Abstract:

    Enterohemorrhagic Escherichia coli (EHEC) O157:H7 has been considered as an Agent responsible for outbreak of hemorrhagic colitis and the hemolytic uremic syndrome. We examined the effect of the Probiotic Agent Clostridium butyricum MIYAIRI strain 588 on EHEC O157:H7 infections in vitro and in vivo using gnotobiotic mice. The growth of EHEC O157:H7 and the production of Shiga-like toxins in broth cultures were inhibited by co-incubation with C. butyricum. The antibacterial effects of butyric and lactic acid were demonstrated in a dose-dependent manner. In addition, the inhibitory effect of butyric acid on the viability of EHEC was demonstrated not only at low pH, but also at neutral pH adjusted to 7.0. Flowcytometric analysis showed that pre-incubation of Caco-2 cells with C. butyricum and E. coli K12 inhibited the adhesion of EHEC O157:H7. However, the effect of C. butyricum on adhesion of EHEC to Caco-2 cells was more inhibitory than that of E. coli K12. Gnotobiotic mice mono-associated with EHEC O157:H7 died within 4–7 days after the infection. On the other hand, all gnotobiotic mice prophylactically pre-treated with C. butyricum survived exposure to EHEC O157:H7 and of the gnotobiotic mice therapeutically post-treated with C. butyricum, 50% survived. Both counts of EHEC O157:H7 and the amounts of shiga-like toxins (Stx1 and Stx2) in fecal contents of gnotobiotic mice di-associated with EHEC O157:H7 and C. butyricum were less than those of gnotobiotic mice mono-associated with EHEC O157:H7. These results indicated that the Probiotic bacterium C. butyricum MIYAIRI strain 588 has preventive and therapeutic effects on EHEC O157:H7 infection in gnotobiotic mice.

    The possible role of Saccharomyces boulardii, a nonpathogenic yeast with beneficial effects on the human intestine, in the maintenance treatment of Crohn's disease has been evaluated. Thirty-two patients with Crohn's disease in clinical remission (CDAI < 150) were randomly treated for six months with either mesalamine 1 g three times a day or mesalamine 1 g two times a day plus a preparation of Saccharomyces boulardii 1 g daily. Clinical relapses as assessed by CDAI values were observed in 37.5% of patients receiving mesalamine alone and in 6.25% of patients in the group treated with mesalamine plus the Probiotic Agent. Our results suggest that Saccharomyces boulardii may represent a useful tool in the maintenance treatment of Crohn's disease. However, in view of the product's cost, further controlled studies are needed to confirm these preliminary data.
