Optimization of Factors Related to High Frequency Plant Regeneration of Pueraria lobata
Subtropical Plant Science, 2008Co-Authors: Shao Xing-huaAbstract:The effects of several factors on dedifferentiation and redifferentiation of Pueraria lobata earthnut were studied.The results showed that the best disinfection way of Pueraria lobata stems with a bud was firstly treated with 70% ethyl alcohol for 30 seconds then treated with 0.1% HgCl2 for 15 minutes,and the optimum regeneration culture medium was MS+NAA 0.5mg/L + 6-BA 5.0mg/L.Liquid culture with sucrose 30g/L,pH 6.0,cultured in light at 25℃ gave a best result for high frequency plant regeneration of Pueraria lobata.

A Study on Cryopreservation Technique for Pueraria lobata Germplasm
2007Co-Authors: Shao Xing-huaAbstract:This paper deals with the cryopreservation technique for Pueraria lobata germplasm.The procedure of cryopresevation was as follows: the stem with a bud was used as materials;At first,the plantlets of Pueraria lobata cultured in vitro for 60 days were treated at 4 ℃for 5 days;the stems of Pueraria lobata about 1~1.5 cm in length were cut and precultured for 1 day at 4 ℃ in MS(Medium supplemented) with 50 g/L DMSO+50 g/L Sucrose,then were dehydrated with the combinative cryoprotectant(100 g/L DMSO+50 g/L Mannitol+50 g/L PVP) for 60 minutes at 0 ℃,-20 ℃ and-80 ℃ respectively;At last,the stems were immersed immediately into liquid nitrogen directly and conserved for 24 hours.After rapidly thawing in a water bath at 40 ℃ for 90 seconds,the stems were washed 2 times with MS medium supplemented with 1.2 mol/L sucrose,then transferred and re-cultured on the MS medium supplemented with BA 2 mg/L and NAA 1 mg/L for 7 days in dark and then in light.The survival rate was up to 45%~47%.The regenerated plantlets of Pueraria lobata were the same as the normal plantlets in morphology.

Cryopreservation of Pueraria lobata Shoot-tip in vitro by Vitrification and Plant Regeneration
Subtropical Plant Science, 2007Co-Authors: Shao Xing-huaAbstract:The cryopreservation technique of Pueraria lobata shoot-tip was studied in this paper.The results showed that the better procedure of cryopreservation by vitrification was as follows.The aseptic plantlets of H# Pueraria lobata subcultured in vitro for 30 days were treated at 4 ℃ for 5 days.The shoot-tips 1.5~2.5mm in length with one or two leaf primordium of H# Pueraria lobata were cut and precultured at 4℃ for 1 day in MS supplemented with 5%DMSO+5%sucrose.They were dehydrated with 60% and 100% PVS2(PVS2: 30%glycerol+15%glycol+15% dimethyl sulfoxide + 13.7% sucrose) at 0℃ for 30 minutes,respectively.The shoot-tips were immersed immediately into liquid nitrogen directly and conserved for 24 hours.After rapidly thawing in a water bath at 40℃ for 90 seconds,the shoot-tips were washed two times with MS supplemented with 41.1% sucrose(10 minutes each time),then transferred and re-cultured on the MS medium supplemented with BA 2 mg/L and NAA 1mg/L in dark for 7 days and then in light.The survival rate was up to 60%.The regenerated plantlets of Pueraria lobat were the same as the normal plantlets in morphology.

The Effect of PP_(333) on Conservation in vitro of Pueraria lobata
Journal of Shangrao Normal College, 2007Co-Authors: Hong SenAbstract:ObjectTo study the effects of PP_(333) on preservation in vitro of Pueraria lobata.MethodsThe effect of PP_(333) on preservation in vitro of Pueraria lobata were explored by room temperature and 4℃ low temperature culture methods.Results At room and low temperature,PP_(333) could improve revival rates of Pueraria lobata plantlets.At room temperature,the treatment effect of PP_(333) with 2 mg/L 6-BA and 1 mg/L NAA was better than that of PP_(333) alone.The best PP_(333) concentrations of Pueraria lobata were 8 mg/L(treated by PP_(333) with 2 mg/L 6-BA and 1 mg/L NAA)and 5 mg/L(treated by PP_(333) alone) respectively,its revival rate was up to 80% after 180 days;At 4℃ low temperature,its treatment effect of PP_(333) was better than that of PP_(333) at room temperature.The best PP_(333) concentrations of Pueraria lobata was 5 mg/L(treated by PP_(333) with 2 mg/L 6-BA and 1 mg/L NAA),its revival rate was up to 98% after 180 days.The plantlets were cultured at room temperature that transferred to subculture medium after conserved in vitro,Pueraria lobata regenerated plantlets were as same as the normal subculture plantlets in morphology.Conclusion The suitable concentration of PP_(333) with 4℃ low temperature can improve evidently revival rates of Pueraria lobata plantlets.

Effect of CaCl_2 on Physiological and Biochemical Indices of Pueraria lobata Earthnuts at Low Temperature after Harvest
