    The effects of the inoculation of a mixed starter culture and vacuum packaging (during Resting Stage) on odour, appearance, texture and flavour of dry-cured ham were studied. After salting, half of the 36 processed hams were inoculated with a commercial starter culture containing lactic-acid bacteria, Gram-positive catalase-positive cocci and yeasts. Nine hams per group (inoculated and non-inoculated) remained vacuum-packaged during Resting. External odour during the process, as well as appearance of the cut surface, texture and flavour on semimembranosus and biceps femoris of the final product were assessed. Vacuum packaging during Resting caused an increase in white film and feedstuff flavour, as well as a decrease in aged flavour, hardness, fibrousness and overall liking. The use of the starter culture brought about an increase in feedstuff flavour, a decrease in sweetness, aged flavour, nutty flavour and overall liking and, only in vacuum-packaged hams, the development of a floral flavour, but had no significant effect on texture descriptors. The starter culture studied is considered inappropriate for the production of traditional Spanish dry-cured ham regardless of the type of Resting used.

    Abstract The effect of the inoculation of a starter culture and vacuum packaging (during the Resting Stage) on dry-cured ham appearance, microbiological and physicochemical parameters was studied. Half of the 36 processed hams were inoculated, after salting, with a commercial starter culture containing lactic-acid bacteria, Gram-positive catalase-positive cocci and yeasts. 18 hams per group (inoculated and non-inoculated) remained vacuum packaged during Resting. Microbiological analyses were carried out on the lean surface during processing, on subcutaneous fat tissue at the drying Stage and on lean tissue in the finished product. Appearance was evaluated during processing. Physicochemical analyses (NaCl, H 2 O, proteolysis index, a w ) were done on Semimembranosus and Biceps femoris in the final product. Inoculation caused a reduction of mould growth and oil drip. Vacuum packaging induced increased proteolysis and increases in all microbial counts and a reduction of oil drip, mould growth and weight loss during processing.

We combined different approaches to evaluate the importance of Resting Stages in Singapore coastal waters. We used molecular approaches to improve the knowledge on Singapore biodiversity, we sampled and extracted cysts from sediments to evaluate the density of Resting Stages in Johor Strait, and we compared systematically information on Singapore planktonic biodiversity to existing published information on Resting Stages from these reported organisms. This is the first study evaluating the importance of Resting Stages in Singapore waters. Above 120 species reported in Singapore are known to produce Resting Stages though no previous work has ever been done to evaluate the importance of these strategies in these waters. The results from the Resting Stage survey confirmed 0.66 to 5.34 cyst g^−1 dry weight sediment were present in the Johor Strait suggesting that cysts may be flushed by tidal currents into and out of the strait regularly. This also suggest that the blooms occurring in Singapore are likely due to secondary growth of Harmful Algae Bloom species in the water rather than from direct germination of cysts from sediment. Finally, we discuss the importance of these Resting eggs for three main national industries in Singapore (shipping, marine aquaculture and provision of drinking water through seawater desalination). We argue that this study will serve as a baseline for some of the future management of Singapore waters.

    Abstract The distribution of Resting Stage cells of diatoms in the surface sediments and water samples collected in the Chukchi Sea was investigated using the most probable number (MPN) analysis to understand the mechanism underlying diatom blooms in the Arctic region. High densities (3.5 × 105 to 6.8 × 106 MPN cells cm−3 wet sediment) of viable Resting Stage cells of typical Arctic diatom species were found in the surface sediments of the Chukchi Sea. Resting Stage cells of the sea-ice–related diatom genus Fragilariopsis were more abundant (2.6 × 106 MPN cells cm−3 wet sediment) than Chaetoceros (2.18 × 106 MPN cells cm−3 wet sediment) or Thalassiosira (1.76 × 106 MPN cells cm−3 wet sediment) in the southern Chukchi Sea. This reflected a previous bloom of Fragilariopsis ahead of typical planktonic blooms of Chaetoceros and Thalassiosira and suggests that sea-ice–related blooms are a substantial factor for primary production in this area.     Abstract We devised a simple and rapid technique for establishing axenic cultures of centric diatoms by sieving and washing Resting Stage cells. This technique is an effective means for establishing the axenic cultures of species that have endogenous Resting spores. The primary advantage of this method is in the isolation of cells when the target species is absent from the water column.

    Abstract Information on diatom Resting Stages is fundamentally important to understanding the population dynamics of diatoms including bloom formation. The distribution of viable diatom Resting Stage cells in bottom sediments of the eastern Bering Sea in July 2009 was investigated by the most probable number (MPN) method. The abundances of diatom Resting Stage cells ranged from 1.7×10 3 to 1.2×10 6  MPN cells cm −3 wet sediment, comparable to those in shallow eutrophic areas where diatom blooms frequently occur. Common species during the spring phytoplankton bloom in the eastern Bering Sea were also dominant in sediments as Resting Stage cells. It should be noted that relatively high numbers of ice algae species, especially ribbon-shaped chain forming pennate diatoms, were found in the sediments. The life cycle strategy using Resting Stage cells allows planktonic and ice algal species to survive unfavorable environmental conditions such as the dark winter season, and potentially contribute to form blooms of several types (subsurface of ice, ice edge, plankton) through vertical mixing.

    Resting Stage cells of six diatom species, Actinoptychus senarius, Biddulphia alternans, Lithodesmium variabile, Odontella longicruris, O. mobiliensis, and Detonula pumila were newly identified from bottom sediments of Ago Bay, central Japan. The Resting Stage cells of A. senarius, B. alternans and L. variabile are very similar in morphology to their vegetative cells; hence, these Resting Stage cells areResting cells". The Resting Stage cells of O. longicruris and O. mobiliensis closely resemble the vegetative cells in morphology, therefore, they are regarded as Resting cells. However, they have some morphological differences compared with the vegetative cells, indicating a possibility of them being Resting spores. The Resting Stage cells of D. pumila are quite different in morphology from the vegetative cells; therefore, these areResting spores". The morphology of the Resting spore of D. pumila is circular in valve view. In girdle view, each valve shows the same shape, slightly vaulted, and has a distinct mantle. As a com- mon feature of the Resting Stage cells of the six species, the plastids show a darker color than those of vegetative cells, and are positioned unevenly in the cells.

    Abstract:

    The abundance and species composition of viable Resting Stage cells of diatoms were investigated in deep waters (200, 500 and 1,000 m depths) collected at neighboring stations in Kumano-Nada, central part of Japan, in April, August and October 2006. Viable Resting Stage cells were enumerated by the modified extinction dilution method (most probable number (MPN) method) based on incubation. Resting Stage cells were detected from all samples, except at 500 m depth in August, in a range of abundances between 200-4,415 cells L � 1 . The Resting Stage cells of diatoms observed belonged to four genera of the Centrales, Chaetoceros spp. (C. curvisetus, C. socialis and Chaetoceros sp.), Leptocylindrus danicus, Skeletonema spp. and Thalassiosira spp., and two of the Pen- nales, Cylidrotheca closterium and Navicula sp., along with one centric and three pennate unidentified diatoms, in total. It is indicated, in this study, that the abundances enumerated by the MPN method could represent those of disassembled Resting Stage cells originally aggregated in marine snow, of which the sinking rate is reportedly great (e.g. 68 m day � 1 ). This, eventually, suggests that the abun- dance and species composition of Resting Stage cells in the deep waters reflected the states of blooms occurring just before our sam- plings in the surface water of the Kumano-Nada region. In this region, coastal upwelling occasionally occurs, implying that such rest- ing Stage cells sinking from the euphotic layer to deeper depths might have some chance to return to the surface and be able to act as a "seeding population" for further blooms.
