The work reported here aimed to identify optimum dose levels of these plants to decrease methane production without causing adverse changes in ruminal fermentation. Increasing doses of both RH and FR, used as a dried powder, were tested in batch cultures in vitro . Amounts added of these plants replaced 48, 91, 135 and 178 mg/g of the substrate incubated (520 mg in the control), respectively, corresponding to 0.50, 0.95, 1.40 and 1.85 g/l in the medium of 50 ml, respectively. Substrate incubated was a mixture of alfalfa hay (500 g/kg), grass hay (200 g/kg) and barley grain (300 g/kg). Both plants caused similar changes in rumen fermentation pattern, resulting in a dose-dependent linear decrease in methane production and in the acetate to propionate ratio. These changes were similar to those observed with monensin, included in the assay as a positive control. At the higher doses ( i.e. , 1.40 and 1.85 g/l), the plant additives seemed to have an adverse effect on rumen fermentation, causing a decrease in extent of substrate degradation accompanied by lower production of total volatile fatty acids (VFA). However, this inhibition did not occur with the lower doses of addition ( i.e. , 0.50 and 0.95 g/l). Based on effects of these plants on butyrate and branched-chain VFA produced, and on the diminished calculated hydrogen recovery, our suggestion is that these plants may have a direct effect on methanogens. In this in vitro system, doses up to 1 g/l of either RH or FR caused a decrease in methane production accompanied by changes in the VFA molar proportions, whereas other fermentation parameters were not affected, suggesting that this would be the optimum rate of addition of these plant additives when the goal is to decrease methane emissions from ruminal fermentation.

M. rosenbergii were randomly divided into five groups: a control group was fed with basal diet, and four treated groups fed with basal diet supplemented with 0.05%, 0.1%, 0.2%, and 0.4% anthraquinone extract for 8 weeks, respectively. The growth, the variations of haemolymph lysozyme, alkaline phosphatase, nitrogen monoxide, total anti-oxidative capacity, glutathione peroxidase, and malondialdehyde content and the relative level of hepatic HSP70 mRNA were investigated. The growth gain rate, length gain rate, haemolymph alkaline phosphatase activity and the relative level of hepatic HSP70 mRNA of M. rosenbergii fed with 0.1% anthraquinone extract for 6 or 8 weeks were higher than those of the control. Weight gain rate and haemolymph alkaline phosphatase activity of M. rosenbergii fed with 0.2% anthraquinone extract for 8 weeks were higher than those of the control. However feed conversion ratios of M. rosenbergii fed with 0.1% and 0.2% anthraquinone extracts for 8 weeks were lower compared to those of the control (P < 0.05). The haemolymph lysozyme activity and total anti-oxidative capacity in the group fed with 0.05% anthraquinone extract for 8 weeks were significantly higher than those of the control. However, there were no significant differences in haemolymph nitrogen monoxide concentrations and glutathione peroxidase activities between all treatment groups and the control. The present study suggested that the ingestion of 0.1%–0.2% anthraquinone extracts supplemented with a basal diet had the potential to stimulate immunity, increase the gene expression of HSP70, and promote the growth of prawns.

Then, freshwater prawns were exposed to high temperature stress at 35 °C for 48 h. The growth, changes in haemolymph total protein, aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), lysozyme, nitrogen monoxide (NO) and hepatic catalase (CAT), superoxide dismutase (SOD) and malondialdehyde (MDA) were investigated. The results showed that compared the control group, the specific growth rates, feed conversion efficiency, haemolymph ALP and lysozyme activities, total protein contents, hepatic CAT and SOD activities increased while haemolymph AST, ALT and hepatic MDA contents decreased in treatment groups before the stress, but their levels did not correlate with the doses of anthraquinone extracts. The specific growth rate (SGR), feed conversion efficiency and haemolymph lysozyme activity significantly increased but haemolymph AST activity decreased in 0.1% dose group; whereas haemolymph ALP activity and feed conversion efficiency increased but ALT activity and hepatic MDA contents significantly decreased in 0.2% dose group before the stress compared with the control. After high temperature stress, 0.1–0.2% anthraquinone extract also could improve the haemolymph total proteins, lysozyme and ALP activities, hepatic catalase, and superoxide dismutase, and reduce haemolymph ALT and AST activities, hepatic malondialdehyde contents. The cumulative mortality in the control was about 100% at 48 h after high temperature stress while the cumulative mortality in the treatment groups supplemented with 0.1–0.2% anthraquinone extract were about 48–65%. The artificial infection with Vibrio anguillarum also showed the cumulative mortality in the control was about 100% while the cumulative mortality in the treatment groups supplemented with 0.1–0.2% anthraquinone extracts were about 57–80%. The present study suggested that ingestion of a basal diet supplemented with 0.1–0.20% anthraquinone extracts could prevent high temperature stress and promote the growth of prawns.

A 12-week rearing experiment was conducted with 0, 125, 250, 500, and 1000 mg kg-1 anthraquinone extract from Rheum Officinale Bail-supplemented diets (AE0, AE125, AE250, AE500, and AE1000), and followed a 48-h thermal stress with 32℃ incubation. Results indicate AE250 and AE500 significantly improved the growth performance and feed utilization, the optimum level was estimated to be 251.22 mg kg-1 based on the regression analysis of special growth ratio (SGR). Meanwhile, AE250 and AE500 improved antioxidant enzyme activity and immune-related protein concentration of iNOS-NO signaling. Under thermal stress, AE250 and AE500 improved the heat tolerance, and Toll-Relish signaling was active to the resistance. These results indicate anthraquinone extract could be used as an effective immunostimulant to improve growth performance, physiological balance and protect organism form environmental hyperthermia stress. This may provide insights for immunostimulant development in aquaculture production.

Comparison Study of the Effects of Anthraquinone Extract and Emodin from Rheum Officinale Bail on the Physiological Response, Disease Resistance of Megalobrama amblycephala under High Temperature Stress

The results showed that compared with the control group, the two treatment groups significantly increased serum lysozyme activities, superoxide dismutase activities and the relative HSP70 mRNA levels of liver and reduced serum alanine aminotransferase activities before stress. After 12 h high temperature stress, the two treatment groups significantly increased serum triglycerides contents 6 h after stress, liver superoxide dismutase activities 2 and 6h after stress and the relative HSP70 mRNA levels of liver 2 h after stress compared with the control group. And there are lower in the serum cortisol content 2 and 12 h after stress, serum aspartate aminotransferase activity 12 h after stress and liver malondialdehyde content 6 h after stress in the two treatment groups than those of the control group. Artificial infection with Aeromonas hydrophila showed that the cumulative mortality in the control was higher than that of the treatment group. The results suggest that ingestion of supplements with 0.1% anthraquinone extract and 60 ppm emodin can increase immune ability, enhance resistance against high temperature stress and A. hydrophila infection.

Effects of anthraquinone extract from Rheum Officinale Bail on the physiological responses and HSP70 gene expression of Megalobrama amblycephala under Aeromonas hydrophila infection.

The fish were randomly divided into two groups: a control group (fed a standard diet) and a treatment group (standard diet supplemented with 0.1% anthraquinone extract) and fed for 10 weeks. We then challenged the fish with A. hydrophila and recorded mortality and changes in serum cortisol, lysozyme, alkaline phosphatase (ALP), total protein, aspartate aminotransferase (AST), alanine aminotransferase (ALT), and hepatic catalase (CAT), superoxide dismutase (SOD), malondialdehyde (MDA) and the relative expression of heat shock protein 70 (HSP70) mRNA for a period of 5 d. Supplementation with 0.1% anthraquinone extract significantly increased serum lysozyme activity before infection, serum ALP activity at 24 h after infection, serum total protein concentration 12 h after infection, hepatic CAT activity 12 h after infection, hepatic SOD activity before infection, and the relative expression of hepatic HSP70 mRNA both before infection and 6 h after infection. In addition, the supplemented group had decreased levels of serum cortisol 6 h after infection, serum AST and ALT activities 12 h after infection, and hepatic MDA content 12 h after infection. Mortality was significantly lower in the treatment group (86.67%) than the control (100%). Our results suggest that ingestion of a basal diet supplemented with 0.1% anthraquinone extract from R. Officinale Bail can enhance resistance against pathogenic infections in M. amblycephala.

Comparison Study of the Effects of Anthraquinone Extract and Emodin from Rheum Officinale Bail on the Physiological Response, Disease Resistance of Megalobrama amblycephala under High Temperature Stress

The results showed that compared with the control group, the two treatment groups significantly increased serum lysozyme activities, superoxide dismutase activities and the relative HSP70 mRNA levels of liver and reduced serum alanine aminotransferase activities before stress. After 12 h high temperature stress, the two treatment groups significantly increased serum triglycerides contents 6 h after stress, liver superoxide dismutase activities 2 and 6h after stress and the relative HSP70 mRNA levels of liver 2 h after stress compared with the control group. And there are lower in the serum cortisol content 2 and 12 h after stress, serum aspartate aminotransferase activity 12 h after stress and liver malondialdehyde content 6 h after stress in the two treatment groups than those of the control group. Artificial infection with Aeromonas hydrophila showed that the cumulative mortality in the control was higher than that of the treatment group. The results suggest that ingestion of supplements with 0.1% anthraquinone extract and 60 ppm emodin can increase immune ability, enhance resistance against high temperature stress and A. hydrophila infection.

Effects of anthraquinone extract from Rheum Officinale Bail on the physiological responses and HSP70 gene expression of Megalobrama amblycephala under Aeromonas hydrophila infection.

The fish were randomly divided into two groups: a control group (fed a standard diet) and a treatment group (standard diet supplemented with 0.1% anthraquinone extract) and fed for 10 weeks. We then challenged the fish with A. hydrophila and recorded mortality and changes in serum cortisol, lysozyme, alkaline phosphatase (ALP), total protein, aspartate aminotransferase (AST), alanine aminotransferase (ALT), and hepatic catalase (CAT), superoxide dismutase (SOD), malondialdehyde (MDA) and the relative expression of heat shock protein 70 (HSP70) mRNA for a period of 5 d. Supplementation with 0.1% anthraquinone extract significantly increased serum lysozyme activity before infection, serum ALP activity at 24 h after infection, serum total protein concentration 12 h after infection, hepatic CAT activity 12 h after infection, hepatic SOD activity before infection, and the relative expression of hepatic HSP70 mRNA both before infection and 6 h after infection. In addition, the supplemented group had decreased levels of serum cortisol 6 h after infection, serum AST and ALT activities 12 h after infection, and hepatic MDA content 12 h after infection. Mortality was significantly lower in the treatment group (86.67%) than the control (100%). Our results suggest that ingestion of a basal diet supplemented with 0.1% anthraquinone extract from R. Officinale Bail can enhance resistance against pathogenic infections in M. amblycephala.

M. rosenbergii were randomly divided into five groups: a control group was fed with basal diet, and four treated groups fed with basal diet supplemented with 0.05%, 0.1%, 0.2%, and 0.4% anthraquinone extract for 8 weeks, respectively. The growth, the variations of haemolymph lysozyme, alkaline phosphatase, nitrogen monoxide, total anti-oxidative capacity, glutathione peroxidase, and malondialdehyde content and the relative level of hepatic HSP70 mRNA were investigated. The growth gain rate, length gain rate, haemolymph alkaline phosphatase activity and the relative level of hepatic HSP70 mRNA of M. rosenbergii fed with 0.1% anthraquinone extract for 6 or 8 weeks were higher than those of the control. Weight gain rate and haemolymph alkaline phosphatase activity of M. rosenbergii fed with 0.2% anthraquinone extract for 8 weeks were higher than those of the control. However feed conversion ratios of M. rosenbergii fed with 0.1% and 0.2% anthraquinone extracts for 8 weeks were lower compared to those of the control (P < 0.05). The haemolymph lysozyme activity and total anti-oxidative capacity in the group fed with 0.05% anthraquinone extract for 8 weeks were significantly higher than those of the control. However, there were no significant differences in haemolymph nitrogen monoxide concentrations and glutathione peroxidase activities between all treatment groups and the control. The present study suggested that the ingestion of 0.1%–0.2% anthraquinone extracts supplemented with a basal diet had the potential to stimulate immunity, increase the gene expression of HSP70, and promote the growth of prawns.

antioxidant phenolic constituents in roots of Rheum Officinale and rubia cordifolia structure radical scavenging activity relationships
Journal of Agricultural and Food Chemistry, 2004Co-Authors: Yizhong Cai, Mei Sun, Jie Xing, Harold CorkeAbstract:The phenolic constituents in the roots of Rheum Officinale and Rubia cordifolia were identified with the aid of high-performance liquid chromatography and liquid chromatography-mass spectrometry and by comparison with authentic standards. A total of 17 hydroxyanthraquinones, gallic acid, and tannins were separated, and 14 of them were identified, being the main phenolic constituents present. Their antioxidant activity (Trolox equivalent antioxidant capacity) was evaluated using the improved 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt method. Hydroxyanthraquinones were the predominant antioxidant phenolic constituents in the roots of R. cordifolia, and tannins and gallic acid were the predominant antioxidant phenolic constituents in the roots of R. Officinale. The structure-radical scavenging activity relationships of the tested hydroxyanthraquinones were systematically demonstrated as follows: Hydroxy groups on one benzene ring of the anthraquinone structure were essential for hydroxyanthraquinones to show activity, the ortho-dihydroxy structure in the hydroxyanthraquinone molecules could greatly enhance their radical scavenging effect, and glycosylation of the hydroxyanthraquinones reduced activity.
