  • Taonella mepensis gen. nov., sp. nov., a member of the family Rhodospirillaceae isolated from activated sludge
International Journal of Systematic and Evolutionary Microbiology, 2013
Co-Authors: Wei-liang Dong, Jun Zhang, Yan Huang, Zhong-li Cui
    Abstract:

A novel Gram-negative, non-spore-forming, rod-shaped strain, H1T, was isolated from activated sludge by micromanipulation. No close relatives among cultured bacterial isolates were found; phylogenetic analysis based on 16S rRNA gene sequences revealed that strain H1T forms a deep single branch in the family Rhodospirillaceae . Cells of strain H1T were slightly curved to straight rods (1.2–1.4×1.5–1.7 µm) and motile by a single polar flagellum. Strain H1T was able to grow in the presence of 0–4 % NaCl and grew optimally at 37 °C and pH 6.0–7.0. Chemotaxonomic analysis revealed that strain H1T possessed Q-10 as the predominant ubiquinone and C18 : 1ω7c, C16 : 0 and C19 : 0 cyclo ω8c as the major fatty acids. The DNA G+C content of strain H1T was 65.1 mol%. Comparative analysis of 16S rRNA gene sequences, and phenotypic and chemotaxonomic data, indicate that strain H1T should represent a novel genus and species of the family Rhodospirillaceae . The name Taonella mepensis gen. nov., sp. nov. is proposed. The type strain of Taonella mepensis is H1T ( = CICC 10529T  = CCTCC AB 2012861T  = KACC 16940T).

  • Thalassobaculum salexigens sp. nov., a new member of the family Rhodospirillaceae from the NW Mediterranean Sea, and emended description of the genus Thalassobaculum
International Journal of Systematic and Evolutionary Microbiology, 2010
Co-Authors: Laurent Urios, Valérie Michotey, Laurent Intertaglia, Françoise Lesongeur, Philippe Lebaron
    Abstract:

A novel Gram-negative bacteria, named CZ41_10a(T), was isolated from coastal surface waters of the north-western Mediterranean Sea. Cells were motile, pleomorphic rods, 1.6 mum long and 0.7 mum wide and formed cream colonies on marine agar medium. The G+C content of the genomic DNA was 65 mol%. Phylogenetic analysis of 16S rRNA gene sequences placed the new isolate in the genus Thalassobaculum, a member of the family Rhodospirillaceae, class Alphaproteobacteria. Unlike Thalassobaculum litoreum CL-GR58(T), its closest relative, strain CZ41_10a(T) was unable to grow anaerobically and did not exhibit nitrate reductase activity. On the basis of DNA-DNA hybridization, fatty acid content and physiological and biochemical characteristics, this isolate represents a novel species for which the name Thalassobaculum salexigens sp. nov. is proposed. The type strain is CZ41_10a(T) (=DSM 19539(T)=CIP 109604(T)=MOLA [corrected] 84(T)). An emended description of the genus Thalassobaculum is also given.

  • Pelagibius litoralis gen. nov., sp. nov., a marine bacterium in the family Rhodospirillaceae isolated from coastal seawater.
INTERNATIONAL JOURNAL OF SYSTEMATIC AND EVOLUTIONARY MICROBIOLOGY, 2009
Co-Authors: Dong Han Choi, Chung Yeon Hwang, Byung Cheol Cho
    Abstract:

A Gram-negative, strictly aerobic, slightly curved rod-shaped bacterial strain, designated CL-UU02T, was isolated from coastal seawater off the east coast of Korea. 16S rRNA gene sequence analysis revealed a clear affiliation of this novel strain with the family Rhodospirillaceae. Strain CL-UU02T formed a robust cluster with the type strains of species of the genus Rhodovibrio at 16S rRNA gene sequence similarity levels of 89.9–90.4 %. Strain CL-UU02T shared no more than 89 % 16S rRNA gene sequence similarity with the type strains of other species in the family Rhodospirillaceae. Strain CL-UU02T was able to grow in the presence of 2–6 % sea salts, and grew optimally at 28–30 °C and pH 7–8. The DNA G+C content of strain CL-UU02T was 66.3 mol%. On the basis of phylogenetic analyses and chemotaxonomic and physiological data, strain CL-UU02T is considered to represent a novel species of a new genus in the family Rhodospirillaceae, for which the name Pelagibius litoralis gen. nov., sp. nov. is proposed. The type strain of Pelagibius litoralis is CL-UU02T (=KCCM 42323T=JCM 15426T).

  • Thalassobaculum litoreum gen. nov., sp. nov., a member of the family Rhodospirillaceae isolated from coastal seawater.
International journal of systematic and evolutionary microbiology, 2008
Co-Authors: Gwang I Zhang, Chung Yeon Hwang, Byung Cheol Cho
    Abstract:

A Gram-negative, facultatively anaerobic strain with slightly curved and straight rod-shaped cells, strain CL-GR58(T), was isolated from coastal seawater (near Gori, Korea). Analyses of the 16S rRNA gene sequence revealed that strain CL-GR58(T) belonged to the family Rhodospirillaceae with Azospirillum lipoferum as its closest relative (gene sequence similarity of 90.9 %). Phylogenetic analyses of the 16S rRNA gene sequences showed that strain CL-GR58(T) was not associated with any known genera in the family Rhodospirillaceae. The novel strain grew in the presence of 1-10 % sea salts, optimally at 30-35 degrees C and pH 8. The major cellular fatty acids consisted of C(18 : 1)omega7c (48.5 %), C(16 : 0) (14.8 %), C(17 : 0) (12.2 %), C(19 : 0) cyclo omega8c (6.3 %) and summed feature 3 (C(16 : 1)omega7c and/or iso-C(15 : 0) 2-OH, 6.0 %). Among the phylogenetically related genera, the fatty acid C(17 : 0) was found only in strain CL-GR58(T). The DNA G+C content of the novel strain was 68.0 mol%. According to phylogenetic analyses of the 16S rRNA gene sequence, fatty acid content and the physiological data, strain CL-GR58(T) represents a novel species in a new genus of the family Rhodospirillaceae, for which the name Thalassobaculum litoreum gen. nov., sp. nov. is proposed. The type strain of the type species is CL-GR58(T) (=KCCM 42674(T)=DSM 18839(T)).
