    Abstract:

    Cryptosporidiosis affects the gastrointestinal and respiratory tract of humans as well as of a wide range of companion, farm, laboratory and wild animals. In the past few years, three independent studies have provided strong evidence for the existence of a distinct Cryptosporidium species affecting Tortoises and likely circulating in other reptile species as well. A new Cryptosporidium genotype was firstly detected and genetically characterized in a marginated Tortoise in Italy in 2007 and named Cryptosporidium sp. ex Testudo marginata CrIT-20. The phylogenetic analysis of this isolate indicated that this Cryptosporidium was unique and belonged to the intestinal clade. These findings were later on confirmed by the detection of genetic homologies of isolates from a python and a chameleon from Spain and by recent research in the United States. The latter study presented both the occurrence of intestinal lesions in a pancake Tortoise and a Russian Tortoise and the genetic characterization of the isolates, together with the first pictures of the endogenous stages of Cryptosporidium CrIT-20. Phylogenetic inference based on the sequences representing small subunit of the nuclear ribosomal RNA gene (SSU) of these isolates confirmed the pathological findings because this Cryptosporidium was related to the intestinal group and supported previous results in T. marginata from Italy.     Abstract:

    Stomatitis (mouth infection) is common disease in reptiles. The number of reptiles kept as house pets is increasing so it is likely that clinical cases in veterinary clinics will also increase. It is necessary to identify infective agents and start optimal antimicrobial therapy. In this study microflora was isolated from reptiles kept at Lithuanian zoo: 8 corn snakes, 2 royal snakes, 13 Russian Tortoises. Isolated microflora was: corn snakes- Staphylococcus spp., Streptococcus spp., Pseudomonas spp., Bacillus spp.; royal snakes: Staphylococcus spp., Streptoccocus spp.; Russian Tortoises: Staphylococcus spp., Pseudomonas spp., Bacillus spp., Klebsiella spp. Resistance to antimicrobial substances was identified using modified Kirby-Bauer (indicatory discs) method. Antimicrobial substances were: penicillins, cephalosporins, aminoglycosides, sulphonamides. Resistance to antimicrobial substances of healthy corn snakes mouth microflora was: highest resistance to sulphonamides. Mouth microflora of corn snake with stomatitis was most resistive to cephaclor, oxitetracyclin, amoxicillin. Russian Tortoise mouth microflora was most resistive to amoxicillin and clavulanic acid

    Russian Tortoise (Testudo horsfieldii) is one of the most commonly kept in captivity species of reptiles. It is a reason for incorrect opinion that it is easy to maintain proper environment conditions and nutrition for it. The aim of the presented study was to investigate nutritional preferences of adult Russian Tortoises kept in captivity. The scientific hypothesis was assumed that Russian Tortoises select their feed sources. The experiment was carried out in free choice feeding model and lasted 6 weeks. In this time Tortoises were fed with easy accessible and advised in literature groups of feed: fruits, vegetables and herbs. Each of them was used for 2 weeks. Nutritional preferences of Tortoises were examined on the basis of the weight of remaining food. All used components were chemically analysed for dry matter, crude protein, crude fat, and crude fibre to find compound or nutrient which is key factor for its selection by Tortoises. The results of the experiment confirm the hypothesis that Russian Tortoises are selective in terms of feed choice. The main factor which affects their intake is water content in their fresh matter. It can be interpreted as the result of their evolutionary adaptation for dry environment and rare availability plants with high moisture in natural environment of these reptiles. Additionally, observation of many keepers that turtles prefer yellow and red colours of feed plants were confirmed

    Cryptosporidiosis in squamates is well documented, but there is very limited information available on cryptosporidiosis in testudines. We describe three cases of cryptosporidiosis in Tortoises with associated pathology. Two Russian Tortoises (Agrionemys [Testudo] horsfieldii) and a pancake Tortoise (Malacochersus tornieri), all from separate collections, were found dead. At necropsy, two had histological evidence of intestinal cryptosporidiosis and one had gastric cryptosporidiosis. Consensus Cryptosporidium sp. PCR and sequencing was used to identify the Cryptosporidium sp. present in these three Tortoises. In the juvenile Russian Tortoise with gastric cryptosporidiosis, the organism had 98% homology with a previously reported sequence from an Indian star Tortoise isolate. A second chelonian Cryptosporidium sp. was identified in the pancake Tortoise and the second Russian Tortoise. This sequence was 100% identical to a shorter gene sequence previously reported in a marginated Tortoise. This is the first report coordinating pathology with Cryptosporidium characterization in chelonians. The two Cryptosporidium sp. found in Tortoises segregate according to site of infection, and there may be further differences in pathology, host range, and transmission. These Cryptosporidium sp. appear to be able to infect diverse Tortoise host species. This may be an under-recognized problem in Tortoises.
