The protective effect of Salvia Miltiorrhiza(danshen) on high energy shock wave induced renal damage

Methods:Thirty white male rabbits were divided into 2 groups at random.Group 1 received Salvia Miltiorrhiza and the others received physiological saline as control before ESWL. Plasma ET-1, SOD and MDA were determined and renal morphlogy were observed. Results: After ESWL there was a significant increase in plasma ET-1 and MDA levels (P 0.05), and plasma SOD decreased significantly (P 0.05) compared with the pre-ESWL levers in control; the Salvia miltriorrhiza treated animals showed only slightly not significantly elevated levels in post-ESWL plasma ET-1 and serum MDA. the peak of plasma ET-1 and MDA levels were greatly lower and the plasma SOD were significantly higher versus the control group (P 0.05). It is most important that renal morphological injury was slight on Salvia Miltiorrhiza group. Conclusion: Salvia Miltiorrhiza has protective effect on high energy shock wave induced renal damage.

IEEE Journal of Solid-state Circuits, 2015Co-Authors: Weijun Kong, Yichen Hu, Ping Chen, Xiaoru Wu, Meihua YangAbstract:UNLABELLED: An ultra high performance liquid chromatography with photodiode array detection method is developed for the simultaneous quantitative determination of five water-soluble compounds including danshensu, protocatechualdehyde, rosmarinic acid, Salvianolic acid B, and Salvianolic acid A in Salvia Miltiorrhiza Bge. SAMPLES: Through method optimization, the five compounds all expressed good linearity (R(2) > 0.9990) in a wide concentration range together with satisfactory accuracy, precision, and stability. Moreover, through qualitative analysis of the chemical fingerprint combined with similarity analysis, hierarchical cluster analysis, principle component analysis, and partial least-squares discriminate analysis, we determined that the 13 batches of Salvia Miltiorrhiza Bge. were similar in internal quality and the differences resulted from various cultivation environments, recovery elements, and others. Seen from the results of hierarchical cluster analysis and principle component analysis, the classification of 13 batches was in accordance, and partial least-squares discriminate analysis technique was more suitable than the principle component analysis model to provide a distinct classification of test samples on the basis of their different components. Moreover, a permutation test verified the rationality of partial least-squares discriminate analysis and variable importance plot showed that peaks 37 and 38 were the most significant variables in distinguishing the Salvia Miltiorrhiza Bge. SAMPLES: The idea of the quantitative and qualitative analysis of Salvia Miltiorrhiza Bge. was convenient, sensitive, and comprehensive, which could be applied to evaluate the quality of more traditional Chinese medicines.

Protective effects of Salvia Miltiorrhiza and verapamil on high energy shock wave induced renal damage in rabbits

METHODS: A total of 45 rabbits was randomly assigned to 3 groups. Control received physiological saline and the others received Salvia Miltiorrhiza and verapamil for 3 days before extracorporeal shock wave lithotripy (ESWL), respectively. Plasma ET 1, SOD and serum MDA were measured. RESULTS: After ESWL, there was a significant increase in plasma ET 1 ( P =0.015) and serum MDA levels ( P =0.01) and the activity of SOD in plasma decreased significantly ( P =0.003) compared with that before ESWL in control. In Salvia Miltiorrhiza and verapamil groups, however, plasma ET 1 and serum MDA after ESWL were not significantly different compared with the values before ESWL and were significantly lower than those of the controlled groups. Although the levels of the plasma SOD after ESWL decreased compared with the pre ESWL levels, the activity of SOD in plasma was significantly higher than that of the controlled ones (Salvia Miltiorrhiza: P =0.001, verapamil: P =0.022). All parameters were not significantly different between Salvia Miltiorrhiza and verapamil groups. CONCLUSION: Salvia Miltiorrhiza and verapamil both have protective effects on shockwave induced renal injury in rabbits.

Journal of Agricultural and Food Chemistry, 2009Co-Authors: Guoxin Zhong, Lingjie Zeng, Jia GuanAbstract:The chemical characteristics of Salvia Miltiorrhiza , also called "danshen" in China, were investigated on the basis of the simultaneous quantitative determination of 13 hydrophilic and lipophilic compounds, namely, protocatechuic acid, protocatechuic aldehyde, caffeic acid, ferulic acid, isoferulic acid, rosmarinic acid, Salvianolic acid B, Salvianolic acid A, dihydrotanshinone I, przewalskin, cryptotanshinone, tanshinone I, and tanshinone IIA, in 74 samples collected from different locations using ultraperformance liquid chromatography (UPLC). Hierarchical clustering analysis based on 13 investigated compounds and the similarity of the entire chromatographic pattern showed that S. Miltiorrhiza was significantly different from Salvia przewalskii , an adulterant of danshen. The chemical characteristics of S. Miltiorrhiza collected from different locations in China were revealed, and Salvianolic acid B, rosmarinic acid, cryptotanshinone, and tanshinones I and IIA were optimized as markers for the evaluation, which is helpful in the quality control of S. Miltiorrhiza.

A set of three solvent systems and other experimental conditions were determined by analytical HSCCC. Using the optimized conditions, the preparative HSCCC separation was performed on 50 mg of crude light petroleum extract yielding pure tanshinones of tanshinone IIA (7 mg), tanshinone I (3 mg) and cryptotanshinone (4 mg) all at purities of over 95% in a single run.
