    Abstract:

    The original protocol of two-dimensional electrophoresis with immobilized pH gradient (IPG-Dalt; Gorg et al., Electrophoresis 1988, 9, 531-546) is updated. Merits and limits of different methods for Sample solubilization, Sample Application (by cup-loading or ingel rehydration) with respect to the pH interval used for IPG-isoelectric focusing are critically discussed. Guidelines for running conditions of analytical and micropreparative IPG-Dalt, using wide IPGs up to pH 12 for overview patterns, or narrow IPGs for zoom-in gels for optimum resolution and detection of minor components, are stated. Results with extended separation distances as well as automated procedures are demonstrated, and a comparison between protein detection by silver staining and fluorescent dyes is given. A brief trouble shooting guide is also included.

    Abstract:

    This paper describes a method for extraction of tapazol, thiouracil, methylthiouracil, propylthiouracil and mercaptobenzimidazol (MBI) from thyroid tissue. The solid-phase extraction procedure is optimized to obtain the maximum results for the main thyreostats including MBI. Different combinations of Sample Application, column conditioning and wash steps were tested. The analytes were extracted from the matrix with methanol. After solid-phase extraction they were derivatised with 7-chloro-4-nitrobenzo-2-furazan. Determination is carried out using liquid chromatography-electrospray mass spectrometry. The identification of the analytes was performed according to the final revision of the EU criteria (93/256/EC decision). The detection capability was 20 microg kg(-1) for all mentioned thyreostats.

    Xface, the new version of our open source, platform independent toolkit for developing 3D embodied conversational agents is presented. The toolkit currently incorporates four pieces of software. The core Xface library is for developers who want to embed 3D facial animation to their Applications. XfaceEd editor provides an easy to use interface to generate MPEG-4 ready meshes from static 3D models. Xface-Player is a Sample Application that demonstrates the toolkit in action and XfaceClient is used as the communication controller over network.

    In this paper, we present our open source, platform independent toolkit for developing 3D talking agents, namely Xface. It relies on MPEG-4 Face Animation (FA) standard. The toolkit currently incorporates three pieces of software. The core Xface library is for developers who want to embed 3D facial animation to their software as well as researchers who want to focus on related topics without the hassle of implementing a full framework from scratch. XfaceEd editor provides an easy to use interface to generate MPEG-4 ready meshes from static 3D models. Last, XfacePlayer is a Sample Application that demonstrates the toolkit in action. All the pieces are implemented in C++ programming language and rely on only operating system independent libraries. The main design principles for Xface are ease of use and extendibility.
