The previously undescribed sulfated polygalactopyranosyl-fucopyranan could function as a potential pharmacophore lead against inflammation, type 2 diabetics, hypertension and utilization as natural antioxidant.

Previously undescribed fridooleanenes and oxygenated labdanes from the brown seaweed Sargassum wightii and their protein tyrosine phosphatase-1B inhibitory activity.
Phytochemistry, 2017Co-Authors: Anusree Maneesh, Kajal ChakrabortyAbstract:Previously undescribed fridooleanene triterpenoids 2α-hydroxy-(28,29)-frido-olean-12(13), 21(22)-dien-20-propyl-21-hex-4'(Z)-enoate, 2α-hydroxy-(28,29)-frido-olean-12(13), 21(22)-dien-20-prop-2(E)-en-21-butanoate and oxygenated labdane diterpenoids 2α-hydroxy-8(17), (12E), 14-labdatriene, 3β, 6β, 13α-tri hydroxy 8(17), 12E, 14-labdatriene were purified from the ethyl acetate-methanol and dichloromethane fractions of the air-dried thalli of Sargassum wightii (Sargassaceae), a brown seaweed collected from the Gulf-of-Mannar of Penninsular India. Inhibitory potential of Δ12 oleanenes towards protein tyrosine phosphatase-1B, the critical regulator of insulin-receptor activity were found to be significantly greater (IC50 0.1 × 10-2 and 0.09 × 10-2 mg/mL, respectively) than the standard sodium metavanadate (IC50 0.31 × 10-2 mg/mL). Fridooleanene triterpenoids displayed greater antioxidant activities (IC50DPPH 0.16-0.18 mg/mL) than the commercially available antioxidants, butylated hydroxytoluene and α-tocopherol (IC50DPPH 0.25 and 0.63 mg/mL, respectively). In general, the oxygenated labdane diterpenoids displayed significantly lesser antioxidant and tyrosine phosphatase-1B inhibitory properties than those exhibited by the fridooleanenes. Bioactivities of the titled compounds were primarily determined by the electronic and lipophilic parameters and not by the steric descriptors. Molecular docking simulations and kinetic studies were employed to describe the tyrosine phosphatase-1B inhibitory mechanism. The previously undescribed fridooleanene triterpenoids might be used as potential anti-hyperglycaemic pharmacophore leads to reduce the risk of elevated postprandial glucose levels.

Unprecedented antioxidative and anti-inflammatory aryl polyketides from the brown seaweed Sargassum wightii.
Food research international (Ottawa Ont.), 2017Co-Authors: Anusree Maneesh, Kajal ChakrabortyAbstract:Previously undescribed aryl polyketide lactones, 4-(8-ethyl-tetrahydro-7-oxo-2H-pyran-5-yl)-propyl-4'-methylbenzoate (compound 1) and methyl-2-(12-oxo-7-phenyl-8-vinyl-1-oxa-4,9-cyclododecadien-3-yl)-acetate (compound 2) were purified from ethyl acetate-methanol fraction of the brown seaweed Sargassum wightii. The structures were proposed based on their NMR and mass spectrometric data. The antioxidative activities of the lactones were significantly greater (P 1) compared to non-steroidal anti-inflammatory drugs (SIaspirin 0.03, SIibuprofen 0.43). Putative biosynthetic pathway of title polyketide products through polyketide synthase enzyme cascade catalyzed reactions substantiated the structural attributions of the hitherto unreported aryl polyketides. This is the first report of the occurrence and characterization of two rare skeletal types, oxo-2H-pyranyl and oxa-cyclododecadienyl macrolactone featuring the aryl substituent from marine organisms with potential antioxidative and anti-inflammatory activities.

The maximum zone of inhibition observed was 15.66 mm in 20 mg mL−1 concentration against Vibrio parahaemolyticus. The Minimum Inhibitory Concentration (MIC) of the fucoidan was 12 mg mL−1 against V. parahaemolyticus. The fucoidan was then enriched with Artemia nauplii at four different concentrations such as 100, 200, 300 and 400 mg L−1 for 12 h. The enriched Artemia nauplii were fed to Penaeus monodon post-larvae for 20 days and the growth performance was assessed. The weight gain and SGR of the control group were 0.2432 g and 15.78%, respectively. But, in experimental groups fed with fucoidan enriched Artemia nauplii, the weight gain and SGR were increased and were respectively ranged from 0.2602 to 0.3161 g and from 16.11 to 17.05%. The P. monodon post-larvae were challenged with V. parahaemolyticus for a period of 30 days showed a reduction in mortality percentage of experimental groups over the control group and it was ranged between 36.97 and 89.86%. During the challenge test, the V. parahaemolyticus load was also enumerated from the infected shrimp at every 10 day intervals. In the control group, the Vibrio load showed a linear increase in hepatopancreas and muscle tissues from 10th to 30th days of challenge test, whereas in the experimental groups, the Vibrio load established a declining trend with the advancement of challenged test.

Artemia franciscana nauplii were enriched with extracted fucoidan at four different concentrations (100, 200, 300, and 400 mg/L) and fed to shrimp P. monodon PL for 20 d. After feeding experiment, the P. monodon PL were challenged with WSSV, and the mortality percentage was recorded daily up to 21 d. During the challenge test, the control PL showed 100% cumulative mortality within 9 d, but the fucoidan-enriched Artemia nauplii fed groups of PL exhibited 60–94% cumulative mortality within 21 d. The reduction in mortality percentage of experimental groups of PL over control PL was ranged between 33.71 and 61.65%. The polymerase chain reaction analysis confirmed the concentration-dependent variation in WSSV infection.

Sodium alginate from Sargassum wightii retards mortalities in Penaeus monodon postlarvae challenged with white spot syndrome virus.
Diseases of aquatic organisms, 2012Co-Authors: Grasian Immanuel, Madasasmy Sivagnanavelmurugan, Varatharajan Balasubramanian, Arunachalam PalavesamAbstract:Sodium alginate extracted from brown seaweed Sargassum wightii (16.35 ± 1.42%, mean [±SD] yield from 5 extractions) was prepared as a powder or beads and used to enrich Artemia nauplii at concentrations of 100, 200, 300 and 400 mg l-1. The alginate-enriched nauplii were fed to Penaeus monodon shrimp postlarvae (PL) stage 15 (PL15, i.e. 15 d old) for 20 d. Mean weight gain and specific growth rate over this period were 0.24 g and 15.8%, respectively, in PL groups not fed alginate, and 0.20-0.28 g and 14.7-16.5%, respectively, in PL groups fed alginate. Amongst PL35 then challenged with white spot syndrome virus (WSSV) by immersion, all PL not fed alginate died within 9 d. However, amongst PL fed the 4 concentrations of alginate powder or beads, mortality rates reduced with increasing alginate concentration, and between 25 and 32% PL remained alive when the bioassay was terminated on Day 21. Amongst alginate-fed PL groups compared with the control group, mortality was reduced by 26.5 to 58.4%. Nested PCR detection of WSSV revealed sodium alginate concentration-dependent reductions in infection loads. The data indicate that sodium alginate extracted from brown seaweed and fed to P. monodon can retard progression of WSSV disease.

Fucoidan--a α-D-glucosidase inhibitor from Sargassum wightii with relevance to type 2 diabetes mellitus therapy.

SuganyaAbstract:The present study was conducted to screen the α-d-glucosidase inhibitory activity of fucoidan extracted from Sargassum wightii collected at Mandapam coastal area, Tamil Nadu, India. Fucoidan was extracted from the sporophyll of S. wightii using ethanol, acetone and CaCl2 precipitation. The average yield was 1.8 ± 0.16% and the extracted fucoidan was found to contain 53 ± 0.52% of fucose and 36 ± 0.60% of sulphate. FT-IR, NMR and in vitro α-d-glucosidase activity of purified fucoidan were performed. Fucoidan at the concentration of 31.25, 62.5, 125 and 250 μg exhibited 19, 31, 38 and 71% inhibition against α-d-glucosidase respectively in a dose dependent manner. The IC50 value against α-D-glucosidase of fucoidan is found to be 132.9 μg which is more effective than that of acarbose (1mg). The diverse biological activities of Fucoidan include anticancer, anti inflammatory and antimicrobial but the α-d-glucosidase inhibitory activity of native fucoidan from S. wightii for type 2 diabetes therapy is first of its kind.

Antibacterial effect of fucoidan from Sargassum wightii against the chosen human bacterial pathogens

The major chemical constituents of the extracted fucoidan were analyzed by biochemical methods. It showe d that the extracted fucoidan contains 52.86 ± 0.64% of fucose and 29.26 ± 0.83% of sulphate. The antibacterial efficacy was performed by agar well diffusion, minimum inhibitory concentration (MIC) and minimum inhibitory concentration (MBC) method. The maximum antibacterial activity 18.6 ± 0.32 mm was obtained for Vibrio cholera and the minimum activity 8.6 ± 0.26 mm was obtained for Salmonella typhi. Result of this manifested the considerable antibacterial potentiality of fucoidan against human bacterial pathogens. Toxicity of fucoidan was evaluated by brine shrimp toxicity assay. No toxic effect was observed in fucoidan. Our study concluded that fucoidan might be used as natural and safe antibiotics in curing many bacterial diseases. Further study is req uired to get the better understanding of mode of action of fucoidan against the bacterial pathogens.

During challenge experiment, the immunological parameters such as THC, prophenoloxidase activity, respiratory burst activity, superoxide dismutase activity and phagocytic activity were measured before injection of WSSV (0 day) and after the injection of WSSV on 10th and 21st days, respectively. All the immunological parameters of experimental groups were significantly (P < 0.05) increased than control group. RT-PCR analysis confirmed the considerable reduction of WSSV DNA copy numbers with respect to the concentration of fucoidan. It was concluded that P. monodon fed with fucoidan of S. wightii supplemented diet had enhanced the innate immunity and increased resistance against WSSV infection.

The maximum zone of inhibition observed was 15.66 mm in 20 mg mL−1 concentration against Vibrio parahaemolyticus. The Minimum Inhibitory Concentration (MIC) of the fucoidan was 12 mg mL−1 against V. parahaemolyticus. The fucoidan was then enriched with Artemia nauplii at four different concentrations such as 100, 200, 300 and 400 mg L−1 for 12 h. The enriched Artemia nauplii were fed to Penaeus monodon post-larvae for 20 days and the growth performance was assessed. The weight gain and SGR of the control group were 0.2432 g and 15.78%, respectively. But, in experimental groups fed with fucoidan enriched Artemia nauplii, the weight gain and SGR were increased and were respectively ranged from 0.2602 to 0.3161 g and from 16.11 to 17.05%. The P. monodon post-larvae were challenged with V. parahaemolyticus for a period of 30 days showed a reduction in mortality percentage of experimental groups over the control group and it was ranged between 36.97 and 89.86%. During the challenge test, the V. parahaemolyticus load was also enumerated from the infected shrimp at every 10 day intervals. In the control group, the Vibrio load showed a linear increase in hepatopancreas and muscle tissues from 10th to 30th days of challenge test, whereas in the experimental groups, the Vibrio load established a declining trend with the advancement of challenged test.

Antiviral Effect of Fucoidan Extracted from the Brown Seaweed, Sargassum wightii, on Shrimp Penaeus monodon Postlarvae against White Spot Syndrome Virus

The fucoidan was extracted from S. wightii, and the yield was observed as 2.832 ± 0.204%. Artemia franciscana nauplii were enriched with extracted fucoidan at four different concentrations (100, 200, 300, and 400 mg/L) and fed to shrimp P. monodon PL for 20 d. After feeding experiment, the P. monodon PL were challenged with WSSV, and the mortality percentage was recorded daily up to 21 d. During the challenge test, the control PL showed 100% cumulative mortality within 9 d, but the fucoidan-enriched Artemia nauplii fed groups of PL exhibited 60–94% cumulative mortality within 21 d. The reduction in mortality percentage of experimental groups of PL over control PL was ranged between 33.71 and 61.65%. The polymerase chain reaction analysis confirmed the concentration-dependent variation in WSSV infection.

Sodium alginate from Sargassum wightii retards mortalities in Penaeus monodon postlarvae challenged with white spot syndrome virus.
Diseases of aquatic organisms, 2012Co-Authors: Grasian Immanuel, Madasasmy Sivagnanavelmurugan, Varatharajan Balasubramanian, Arunachalam PalavesamAbstract:Sodium alginate extracted from brown seaweed Sargassum wightii (16.35 ± 1.42%, mean [±SD] yield from 5 extractions) was prepared as a powder or beads and used to enrich Artemia nauplii at concentrations of 100, 200, 300 and 400 mg l-1. The alginate-enriched nauplii were fed to Penaeus monodon shrimp postlarvae (PL) stage 15 (PL15, i.e. 15 d old) for 20 d. Mean weight gain and specific growth rate over this period were 0.24 g and 15.8%, respectively, in PL groups not fed alginate, and 0.20-0.28 g and 14.7-16.5%, respectively, in PL groups fed alginate. Amongst PL35 then challenged with white spot syndrome virus (WSSV) by immersion, all PL not fed alginate died within 9 d. However, amongst PL fed the 4 concentrations of alginate powder or beads, mortality rates reduced with increasing alginate concentration, and between 25 and 32% PL remained alive when the bioassay was terminated on Day 21. Amongst alginate-fed PL groups compared with the control group, mortality was reduced by 26.5 to 58.4%. Nested PCR detection of WSSV revealed sodium alginate concentration-dependent reductions in infection loads. The data indicate that sodium alginate extracted from brown seaweed and fed to P. monodon can retard progression of WSSV disease.

The extracted fucoidan was supplemented with pellet diets at three different concentrations (0.1, 0.2 and 0.3%). The fucoidan supplemented diets were fed to Penaeus monodon for 45 days, then challenged with WSSV and the mortality percentage was recorded daily up to 21 days. During the challenge test, the control group showed 100% mortality within 10 days, but in the experimental groups, the mortality percentage (51–72% within 21 days) was decreased considerably ( P P P. monodon fed with fucoidan of S. wightii supplemented diet had enhanced the innate immunity and increased resistance against WSSV infection.

Extensive spectroscopic experiments in conjugation with monosaccharide compositional analysis attributed (1 → 3)-linked α-fucopyranose units in the polygalactofucan chain with C-2 sulfation and C-4 substituents as O-acetyl/O-methyl/(1 → 4)-linked β-galactopyranose. The previously undescribed sulfated polygalactopyranosyl-fucopyranan could function as a potential pharmacophore lead against inflammation, type 2 diabetics, hypertension and utilization as natural antioxidant.

Previously undescribed fridooleanenes and oxygenated labdanes from the brown seaweed Sargassum wightii and their protein tyrosine phosphatase-1B inhibitory activity.
Phytochemistry, 2017Co-Authors: Anusree Maneesh, Kajal ChakrabortyAbstract:Previously undescribed fridooleanene triterpenoids 2α-hydroxy-(28,29)-frido-olean-12(13), 21(22)-dien-20-propyl-21-hex-4'(Z)-enoate, 2α-hydroxy-(28,29)-frido-olean-12(13), 21(22)-dien-20-prop-2(E)-en-21-butanoate and oxygenated labdane diterpenoids 2α-hydroxy-8(17), (12E), 14-labdatriene, 3β, 6β, 13α-tri hydroxy 8(17), 12E, 14-labdatriene were purified from the ethyl acetate-methanol and dichloromethane fractions of the air-dried thalli of Sargassum wightii (Sargassaceae), a brown seaweed collected from the Gulf-of-Mannar of Penninsular India. Inhibitory potential of Δ12 oleanenes towards protein tyrosine phosphatase-1B, the critical regulator of insulin-receptor activity were found to be significantly greater (IC50 0.1 × 10-2 and 0.09 × 10-2 mg/mL, respectively) than the standard sodium metavanadate (IC50 0.31 × 10-2 mg/mL). Fridooleanene triterpenoids displayed greater antioxidant activities (IC50DPPH 0.16-0.18 mg/mL) than the commercially available antioxidants, butylated hydroxytoluene and α-tocopherol (IC50DPPH 0.25 and 0.63 mg/mL, respectively). In general, the oxygenated labdane diterpenoids displayed significantly lesser antioxidant and tyrosine phosphatase-1B inhibitory properties than those exhibited by the fridooleanenes. Bioactivities of the titled compounds were primarily determined by the electronic and lipophilic parameters and not by the steric descriptors. Molecular docking simulations and kinetic studies were employed to describe the tyrosine phosphatase-1B inhibitory mechanism. The previously undescribed fridooleanene triterpenoids might be used as potential anti-hyperglycaemic pharmacophore leads to reduce the risk of elevated postprandial glucose levels.

Unprecedented antioxidative and anti-inflammatory aryl polyketides from the brown seaweed Sargassum wightii.
Food research international (Ottawa Ont.), 2017Co-Authors: Anusree Maneesh, Kajal ChakrabortyAbstract:Previously undescribed aryl polyketide lactones, 4-(8-ethyl-tetrahydro-7-oxo-2H-pyran-5-yl)-propyl-4'-methylbenzoate (compound 1) and methyl-2-(12-oxo-7-phenyl-8-vinyl-1-oxa-4,9-cyclododecadien-3-yl)-acetate (compound 2) were purified from ethyl acetate-methanol fraction of the brown seaweed Sargassum wightii. The structures were proposed based on their NMR and mass spectrometric data. The antioxidative activities of the lactones were significantly greater (P 1) compared to non-steroidal anti-inflammatory drugs (SIaspirin 0.03, SIibuprofen 0.43). Putative biosynthetic pathway of title polyketide products through polyketide synthase enzyme cascade catalyzed reactions substantiated the structural attributions of the hitherto unreported aryl polyketides. This is the first report of the occurrence and characterization of two rare skeletal types, oxo-2H-pyranyl and oxa-cyclododecadienyl macrolactone featuring the aryl substituent from marine organisms with potential antioxidative and anti-inflammatory activities.

Pharmacological activities of brown seaweed Sargassum wightii (Family Sargassaceae) using different in vitro models

The ethyl acetate:methanol fraction registered greater Fe2+ ion chelating ability (IC50 0.52 mg/ml), and were effective in stabilizing the 2,2-azino-bis-3-ethylbenzothiozoline-6-sulfonic acid (IC50 0.82 mg/mL), and 1,1-diphenyl-2-picryl hydrazil radicals (IC50 0.32 mg/mL) than those derived from chloroform solvent fractions.
