    Abstract:

    Summary The present investigation was performed to determine the effect of long-term administration of sodium selenite on the biochemical indices and concentration of thyroid hormones in calves. Sodium selenite solution was drenched at 0.1 and 0.25 mg per kg body weight for 12 weeks. Higher dose produced characteristic symptoms of Selenosis whereas mild symptoms were observed with lower dose. The toxic symptoms appeared when blood selenium level was 1.68 ± 0.13 µg per ml. There was a significant (P<0.05) increase in the activities of the aspartate aminotransferase, alkaline phosphatase, blood urea nitrogen and creatinine levels in the treated animals. The repeated administration of sodium selenite resulted in a significant p-value decline in thyroxine levels on the 10th week and increase in triiodothyronine on the 8th week of treatment. The findings of the present study suggested that sodium selenite induced Selenosis alters thyroid hormone levels in plasma.

    Abstract:

    The present investigation reports the effect of long-term administration of sodium selenite on immune response in calves. Sodium selenite was administered orally at dose rate of 0.1 and 0.25 mg/kg b.wt. for 98 consecutive days. Higher dose produced characteristic symptoms of Selenosis whereas mild symptoms were observed with the lower dose. The toxic signs appeared when blood selenium levels were 1.68 ± 0.13mg/ml. The forced anti-body titre against the alum precipitate Haemorrhagic septicemia vaccine was significantly higher in the treated animals. The cell-mediated immune response studied by delayed type hypersensitivity test was significantly higher in the treated animals. Thus, raised blood selenium levels augmented the immune response i.e. humoral and cell mediated immune response in the treated animals. Prolonged exposure to selenium also produced marked histopathological changes in the lymph nodes of treated animals.

Nine calves, 9–12 months old, with an average body weight of 104 kg were divided into three groups. Calves of groups 2 and 3 were administered with sodium selenite at 0.1 and 0.25 mg/kg body weight for 98 consecutive days. The clinical signs characteristic of Selenosis viz. alopecia, cracking of hooves, intradigital lesions and discoloration of hard palate, started appearing from 45 to 60 days onwards with high dose, whereas only subtle indications of toxicosis were observed in the low-dose group. The prolonged administration of sodium selenite produced a progressive and dose-dependent decline in the circulating leucocyte count with concomitant decline in the circulating neutrophil count. There was a high negative correlation (0.94) between blood selenium levels and neutrophils. Granulocyte/agranulocyte ratio was also significantly reduced in the treated animals. Evaluation of bone marrow smears revealed a decline in the myeloid to erythroid ratio. In addition, there was also maturation arrest of neutrophils at promyelocyte or myelocyte level as shown by differential granulocyte count in the bone marrow. The results indicated that host's immune response may be adversely affected.

    Abstract:

    The present investigation reports the effect of sodium selenite-induced sub-chronic toxicity in crossbred cow calves on various antioxidant enzymes. Sodium selenite (0.25 mg/kg for 16 w) resulted in characteristic signs of sub-chronic Selenosis, ie alopecia, cracking and enlargement of hooves, interdigital lesions, ring formation on the coronet region, and gangrene at tip of the tail. The sodium selenite resulted in significant rise of blood selenium levels and concurrent increase in erythrocytic glutathione peroxidase (GPx) activity. Blood selenium levels and GPx activity had a high positive correlation (r = 0.97). Blood glutathione levels were lowered from 211.1 ′ 13.4 to 95.56 ′ 11.8 μg/ml. Selenosis caused oxidative stress as evidenced by a 3-fold increase in lipid peroxidation; activities of glutathione-S-transferase, glutathione reductase, superoxide dismutase and catalase were significantly increased. These findings support the hypothesis that the pro-oxidant attributes of selenium play important roles in its toxicity.

    Clinico-haematological alterations in experimentally induced chronic Selenosis by feeding selenium rich wheat straw (8.54 ppm) were studied in 18 non-descript buffalo calves. The clinical signs appeared after 35-45 days. The cardinal signs of induced Selenosis were malformation of hoov.es and horns along with inflammation of coronary band, anaemia, alopecia, emaciation followed by death. Haematological indices revealed macrocytic - normochromic anaemia.

    The present investigation was conducted on 23 non-descriptive healthy male buffalo of 1-1½ years of age. Chronic Selenosis was induced in 18 animals by feeding selenium (Se) enriched wheat straw (Se content 8.54 ppm) for 84 days while 5 animals were kept as healthy controls. Minerals profile of plasma samples reflected inverse relationship of Se and arsenic (As), which was associated with significant decline in mean concentration of magnesium (Mg), calcium (Ca), manganese (Mn) and potassium (K), whereas, effect on plasma copper (Cu) and sulphur (S) was over masked by their mobilization from tissues as confirmed on mineral tissue analysis. Heavy metals, viz. nickel (Ni), lead (Pb), mercury (Hg) and chromium (Cr) showed positive relationship except for cadmium (Cd) which revealed decreasing trend with increasing plasma Se levels in buffalo calves fed on Se enriched wheat straw.

    Blood glucose, plasma biochemicals, viz. plasma urea nitrogen, creatinine, total proteins and albumin; and plasma enzymes activities, viz. alkaline phosphatase (ALP), alanine transferase (AL T) and aspartate transferase (AST) were analysed in buffalo calves after induction of Selenosis by ad lib. feeding of selenium enriched (S.54 ppm) wheat straw for 84 days. The mean blood glucose values (48.70±1.45 mg/dl) reflected hypoglycaemia, whereas mean ALP (90.72±6.98 lU/L), ALT (43.5 1± 1.62 IU/L), AST (89.67±0.30 IU/L) activities and plasma urea nitrogen (13.08±0.69 mg/dl) showed significant increase after 14, 70, 56, 42, and 42 days, respectively, of feeding wheat straw, indicating chronic liver damage and nephritis respectively. However, mean plasma creatinine, total plasma proteins and albumin revealed nonsignificant alterations throughout the period of induction of Selenosis as compared to respective mean values of healthy controls.
