2020Co-Authors: Aulia Muliana InsaniAbstract:The purpose of this research was to discover the effect of Eurycoma longifolia extract towards the number of sertoli and Spermatogonium cell of Rattus norvegicus which was exposed by monosodium glutamate. The research was conducted for 30 days with white male rat as experimental animal with species Rattus norvegicus. This research used complete randomized design factorial. This research used 20 white male rats which was divided into five groups of treatment, each treatment consisted of P- as negative control which was given cmc-na 2 ml and aquadest 1 ml, P+ which was given cmc-na 0,5% 1 ml and MSG 4g/kg bw/day, P1 which was given the extract of Eurycoma longifolia 600mg/kg bw/day and MSG 4g/kg bw/day, P2 which was given the extract of Eurycoma longifolia 900mg/kg bw/day and MSG 4g/kg bw/day. The result showed that the average number of sertoli cell on P-, P+, P1, and P2 was 6,440 ± 0,81731, 1,240 ± 0,08944, 2,640 ± 0,38471, 4,080 ± 0,91214 and the number of Spermatogonium on P-, P+, P1, and P2 was respectively 62,760 ± 4,6376, 22,400 ±11,439, 44,320 ± 8,8324, 59,000 ± 9,2628. THE EFFECT OF CHRONIC EXPOSURE OF NICOTINE INHALATION TO THE COUNT OF SPERMATOGONIA, SERTOLI CELLS AND LEYDIG CELLS OF YOUNG WHITE RAT WISTAR STRAIN

Material & Method: Laboratory experimental study with post test only control group design, measurement of Spermatogonium, leydig cell, sertoli cell in 5 groups of young male Wistar strain, negative control group and treatment group given nicotine exposure 0.5 mg, 1 mg, 2 mg, and 4 mg/kg body weight/day for 30 days. Results: A significant reduction in Spermatogonium was found in the group given nicotine 0.5 mg/kgBW/day (p=0.048), 1 mg/kgBW/day (p=0.002), 2 mg/kgBW/day (p=0.002) and 4 mg/kgBW/day (p=0.000) when compared to the control group. Significant decreases were also seen in the group receiving 4 mg of nicotine exposure compared with 0.5 mg (p=0.018). Significant decrease in sertoli cell count was seen only in the nicotine group of 4 mg/kgBW/day compared with the control group (p=0.047). A significant decrease in leydig cell count was found in the nicotine 2 mg/kgBW/day (p=0.037) and nicotine group 4 mg/kgBW/day (p=0.023) when compared with the control group. Significant decreases were also found in the 4 mg/kgBW/day group compared to the 0.5 mg/kgBW/day group (p=0.004). In this study there were also a decrease in the number of spermatogonia, sertoli cells, and leydig cells in the increased dose of nicotine given although not statistically significant. Conclusion: Chronic exposure of nicotine per inhalation may decrease the number of spermatogonia, sertoli cells, and leydig cells. The higher the dose of nicotine given the greater the decrease in the number of Spermatogonium cells, sertoli cells, and leydig cells that occur. This proves that nicotine is one of the causes of infertility in men.

2018Co-Authors: Aril RizaldiAbstract:Tujuan: Untuk menganalisis perbedaan jumlah Spermatogonium, sel leydig dan sel sertoli pada tikus putih strain Wistar usia muda paska paparan kronis nikotin secara inhalasi. Metode: Penelitian eksperimental laboratorium dengan post test only control group design, dengan pengukuran variabel jumlah Spermatogonium, sel leydig, sel sertoli pada 5 kelompok tikus witar jantan muda yaitu kelompok kontrol negatif dan kelompok perlakuan yang diberikan paparan nikotin 0,5 mg, 1 mg, 2 mg, dan 4 mg/kgBB/hari selama 30 hari. Hasil: Didapatikan penurunan jumlah Spermatogonium yang signifikan pada kelompok yang diberikan nikotin 0,5 mg/kgBB/hari (p=0,048), 1 mg/kgBB/hari (p=0,002), 2 mg/kgBB/hari (p=0,002) dan 4 mg/kgBB/hari (p=0,000) jika dibandingkan dengan kelompok kontrol. Penurunan yang signifikan juga terlihat pada kelompok yang mendapatkan paparan nikotin 4 mg dibandingkan dengan 0,5 mg (p=0,018). Penurunan jumlah sel sertoli yang signifikan hanya terlihat pada kelompok nikotin 4 mg/kgBB/hari dibandingkan dengan kelompok kontrol (p=0,047). Penurunan jumlah sel leydig yang signifikan didapati pada kelompok nikotin 2 mg/kgBB/hari (p=0,037) dan kelompok nikotin 4 mg/kgBB/hari (p=0,023) jika dibandingkan dengan kelompok kontrol. Penurunan yang signifikan juga didapati pada kelompok 4 mg/kg/BB dibandingkan denga kelompok 0,5 mg/kgBB/hari (p=0,004). Pada penelitian ini juga tampak penurunan jumlah Spermatogonium, sel sertoli, dan sel leydig pada peningkatan dosis nikotin yang diberikan walaupun tidak signifikan secara statistik. Kesimpulan: Pemberian paparan kronis nikotin per inhalasi dapat menurunkan jumlah Spermatogonium, sel sertoli, dan sel leydig. Semakin tinggi dosis nikotin yang diberikan maka semakin besar penurunan jumlah sel Spermatogonium, sel sertoli, dan sel leydig yang terjadi. Hal ini membuktikan bahwa nikotin merupakan salah satu penyebab infertilitas pada pria.

2018Co-Authors: Nim Poppy Desra Syahfitri NasutionAbstract:Pendahuluan : MSG adalah salah satu penyedap makanan yang sering disalah gunakan dan diduga MSG dapat menyebabkan terganggunya proses spermatogenesis terutama bila diberikan secara terus menerus pada usia anak-anak. Tujuan : mengetahui jumlah sel Spermatogonium, sel sertoli dan sel leydig paska pemberian MSG secara kronis pada tikus usia muda Metode dan material : Tiga puluh tikus putih strain wistar usia 12 minggu secara acak dibagi menjadi 5 kelompok, masing-masing kelompok terdiri dari 6 ekor tikus. Tikus pada kelompok kontrol negatif menjalani operasi sham kemudian diambil testis kirinya. 4 kelompok lain diberikan dosis MSG masing –masing 5mg/hari, 6 mg/hari, 12mg/hari dan 18mg/hari selama 30 hari kemudian diambil testis kirinya. Jumlah Spermatogonium sel sertoli dan sel leydig diamati dengan pengecatan hematoxylin eosin Hasil : terdapat penurunan jumlah Spermatogonium, sel sertoli dan sel leydig secara bermakna pada kelompok perlakuan pemberian MSG 18mg/hari selama 30 hari dibandingkan dengan kelompok kontrol negative (p<0,005) dan tidak didapatkan perbedaan jumlah sel Spermatogonium, sel sertoli dan sel leyding pada kelompok perlakuan pemberian MSG 5mg/hari dan 6mg/hri selama 30 hari bila dibandingkan dengan kelompok kontrol negative (p>0,005). Kesimpulan : Pemberian MSG dengan dosis 18mg/hari secara kronis (1 bulan) pada tikus wistar jantan usia muda secara signifikan menurunkan jumlah sel Spermatogonium, sel sertoli dan sel leydig.

This research using completely randomized design with eighteen male Mojosari ducks which divided into three group randomly. Group I (P0) without any treatment of laserpuncture, Group II (P1) with 0,2 J dose of laserpuncture, and Group III (P2) with 0,4 J dose of laserpuncture. Laserpuncture shooting was done on reproduction point with three days interval in one month. On the thirty first day, the duck was dissected so that the testes can be taken then made by histological slide of testes. The obtained data processed by Analysis of Variant (ANOVA) followed with Duncan. Research result showed that laserpuncture shoot on the reproduction point of five month old male Mojosari duck decrease the number of Spermatogonium cells and seminiferous tubule diameter of the testes.

2015Co-Authors: Kevin AperiaginadiAbstract:Infertilitas adalah kegagalan atau ketidakmampuan pasangan suami istri untuk hamil setelah 1 tahun beruhubungan tanpa alat kontrasepsi. Salah satu penyebab infertilitas pada pria adalah radikal bebas yang menyebabkan gangguan pada spermatogenesis. Salah satu herbal yang sering digunakan dalam berbagai terapi adalah esktrak kulit manggis karena khasiatnya sebagai antioksidan dan antiproliferasi. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh berbagai dosis ekstrak etanol kulit manggis terhadap jumlah sel Spermatogonium dan diameter tubulus seminiferus tikus Wistar. Penelitian ini adalah penelitian eksperimental laboratorium sungguhan. Tiga puluh ekor tikus dibagi menjadi 5 kelompok. Kelompok tersebut diberikan perlakuan Ekstrak Etanol Kulit Manggis (EEKM) I dosis 200 mg/kgBB, EEKM II 400 mg/kgBB, EEKM III 600 mg/kgBB, Tribulus terrestris 13,5 mg sebagai kontrol pembanding, dan kontrol negatif CMC 1% 5 mL secara oral. Analisis data menggunakan uji ANAVA satu arah dilanjutkan dengan uji beda rata-rata LSD. Hasil analisis statistik jumlah sel Spermatogonium dan diameter tubulus seminiferus menunjukan bahwa EEKM I memiliki perbedaan yang bermakna (p<0,05) dengan kelompok EEKM II, EEKM III, kontrol pembanding dan kontrol negatif. Pada analisis statistic jumlah sel Spermatogonium menunjukan hasil yang sangat bermakna dibandingkan dengan kontrol pembanding. Kesimpulan ekstrak etanol kulit manggis mempengaruhi jumlah sel Spermatogonium dan diameter tubulus seminiferus. EEKM dosis 200 mg/kgBB dapat meningkatkan jumlah Spermatogonium dan diameter tubulus seminiferus.
