    Abstract:

    A strain of Bacillus licheniformis 8-37-0-1 with high exopolysaccharide (EPS) production ability was isolated and identified based on morphological and physiological characteristics and phylogenetic analysis of 16S rDNA sequences. A new type of EPS was isolated from the strain fermentation broth by enzymolysis, isopropanol precipitation, anion-exchange, and gel-filtration chromatography. The new EPS was determined as homogeneous, with a molecular weight of 2.826 x 10(4), as determined by High-Performance Size-Exclusion Chromatography Multi-Angle Laser Light Scattering analysis. Its structural characteristics were investigated and elucidated by methylation analysis, partial acid hydrolysis, gas-liquid chromatography mass spectrometry, Fourier transform infrared, and nuclear magnetic resonance spectroscopy. Based on obtained data, the EPS was found to be a levan containing a (2-->6)-linked backbone with a single beta-d-fructose at the C-1 position every seven residue, on average, along the main chain. Preliminary in vitro tests revealed that EPS could significantly stimulate the proliferation of Spleen Lymphocyte.

    Abstract:

    Abstract A water-soluble polysaccharide (HCP) with a molecular mass of 6.7 × 10 5  Da determined by high performance size-exclusion chromatography (HPSEC), was isolated from the fruit bodies of Sarcodon aspratus (Berk.) S. Ito., an edible mushroom. HCP was elucidated as a liner glucan with a backbone structure of (1 → 6)-linked-α- d -glucopyranosyl residues by interpretation of the composition analysis, methylation analysis, periodate oxidation experiment, FT-IR, and NMR spectroscopy. Immunological activity evaluation using H 3 -thymidine incorporation method revealed that HCP could significantly stimulate the proliferation of the cultured mice Spleen Lymphocyte in a dose-dependent manner . HCP might be a potential immunomodulator which can be used against pathogens and tumors in health-care food or medicine. This is the first report on the detailed structure elucidation of the polysaccharide from S. aspratus .

    Abstract:

    A water-soluble polysaccharide with a molar weight of 4.3 x 10(5) Da, termed as HBP was isolated from the fruit bodies of an edible mushroom, Sarcodon aspratus (Berk.) S. Ito. Composition, methylation analysis, Fourier transform infrared spectroscopy, and nuclear magnetic resonance spectroscopy experiments were conducted to elucidate its structure. The results indicated that HBP is a glucan featuring a backbone of (1 -> 6)-linked-beta-D-glucopyranosyl residues, which occasionally branched at O-3 position on along the backbone and substituted by the side chains that consisting of (1 -> 3)-linked-beta-D-glucopyranosyl, (1 -> 4)-linked-beta-D-glucopyranosyl and non-reducing end beta-D-glucopyranosyl residues. Immunological activity evaluation by H-3-thymidine incorporation method revealed that HBP can significantly stimulate the proliferation of the cultured mice Spleen Lymphocyte in a dose-dependent manner, thus, HBP is a promising potential immunomodulator that can be used as healthcare food or medicine against pathogens and tumors.     Abstract A water-soluble polysaccharide named as PRP was isolated from the fruiting bodies of Phellinus ribis by hot water extraction, DEAE-cellulose and Superdex 30 column chromatography. Its structural characteristics were investigated by FT-IR, NMR spectroscopy, GLC-MS, methylation analysis, periodate oxidation and Smith degradation. Based on the data obtained, PRP was found to be a β- d -glucan containing a (1 → 4), (1 → 6)-linked backbone, with a single β- d -glucose at the C-3 position of (1 → 6)-linked glucosyl residue every eight residues, along the main chain. The glucan has a weight-average molecular weight of about 8.59 kDa by HPGPC determination using dextran samples as the standards. Preliminary activity tests in vitro revealed that PRP could stimulate the proliferation of Spleen Lymphocyte.

    Vegetable oils containing α-linolenic acid (ALA; 18 : 3n-3) have been shown to modulate the functions of immunocompetent cells. The aim of the present study was to understand the modulatory effect of ALA-rich garden cress (Lepidium sativum L.) seed oil (GCO) on lipid composition, Spleen Lymphocyte (SL) proliferation and inflammatory mediator production by peritoneal macrophages (PMΦ) in rats. Female Wistar rats were fed diets containing either GCO (2·5, 5·0 and 10 %, w/w) or sunflower oil (SFO, 10 % w/w) for 8 weeks. Ex vivo proliferation of SL was measured by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. IL-2 and TNF-α in SL and PMΦ were analysed by ELISA. Inflammatory mediators such as NO, leukotriene B(4) (LTB(4)) and H(2)O(2) were measured in mitogen-activated PMΦ. GCO significantly increased the levels of ALA, EPA and DHA, but reduced linoleic acid and arachidonic acid in SL and PMΦ lipids. GCO (10 %) significantly decreased the concanavalin A (Con-A)- and phytohaemagglutinin-mediated proliferation of SL by 54 and 38 %, respectively, in comparison with SFO. A marginal decrease in IL-2 and TNF-α was observed in Con-A-stimulated SL and lipopolysaccharide-stimulated PMΦ. LTB(4) levels in Ca ionophore-stimulated PMΦ were reduced by 40 % in GCO-fed rats. NO release in response to various stimuli was significantly decreased in PMΦ of GCO-fed rats. The present study is the first report on the modulatory efficacy of GCO on immunomediators in rats. GCO modulated inflammatory mediators such as NO and LTB(4), and thus may play a role in alleviating inflammatory conditions favourably.
