    Abstract:

    An actinobacterial strain designated as BN-6 was isolated from Swallow Bird mud nest by agar plate dilution method. It showed the morphological, physiological and biochemical characteristics of the genus Streptomyces . On the basis of 16S rRNA gene sequence analysis and physiological tests, this isolate was found to be most closely related with Streptomyces vinaceusdrappus NRRL2363. The strain BN-6 produced a broad spectrum active metabolite in Glucose Soybean meal (GS) medium and was extracted with HP Dianion resin. The resin extracted product showed activity against Gram-positive, Gram-negative bacteria, acid fast bacilli and fugal cultures, suggesting it broad spectrum characteristics. The bioautography on silica gel plates using Micrococcus luteus MTCC106 as test organism indicated the presence of a single metabolite having a Rf value of 3.2 and 9.1 in two different solvent systems. The characteristic peak obtained in UV–VIS spectrophotometer at 214 nm and chemical analysis indicates that it may be peptide class of metabolite.

    Abstract:

    The aim of this study was to isolate and screen actinomycetes from solitary wasp and Swallow Bird mud nests for antimicrobial activity. The actinomycetes were isolated from soil of nests of solitary wasp and Swallow Bird, and identified on the basis of morphological characteristics and molecular biological methods. A total of 109 actinomycetal isolates were obtained from 12 soil samples (6 from each habitat) using two media. The highest number of actinomycetes were recovered on Humic acid vitamin agar media (65.13%, n  = 71) as compared to actinomycetes isolation agar media (34.86%, n  = 38). The antimicrobial activity of actinomycetes isolates was determined using the agar plug method. Among 109 isolates, 51 isolates (46.78%) showed antibacterial activity by agar plug assay. The morphological and molecular characteristics confirmed that the most of active isolates in both sample belonged to the genus Streptomyces , the other potential genera like Streptosporangium , Actinomadura , Saccharopolyspora , Thermoactinomycetes and Nocardia were also recovered, but in a low frequency. The isolates designated as 8(1)*, BN-6, MN 2(6), MN 2(7) and MN 9(V) showed most promising activity against various drug resistant bacterial pathogens. It seems that the promising isolates from these unusual/unexplored habitats may prove to be an important step in development of drug for treating multi-drug resistant bacterial pathogens.

    Abstract:

    This thesis discusses the tort that builds the Swallow Bird house above the shop house where his business is in Sintang District. In addition it also has a purpose that is to disclose and analyze the causes of traders who build houses Swallow Bird above the shop house where business can be categorized as an tort and the policy made by the Government of Sintang District against the construction of Swallow Birdhouse on top shop house. Through literature study using empirical legal approach method, it is concluded that the causes of merchants who built swiftlet houses above shop house where their business can be categorized as unlawful act due to the actions of traders who build Swallow Bird house above shop house where his business in Sintang District has violated the provisions of Article 1365 of the Civil Code and fulfills elements of unlawful acts and policies perpetrated by the Government of Sintang Regency against the construction of swiftlet houses above shop houses by: (a) Socialization of Regional Regulation of Sintang Regency Number 8 Year 2010 about Building Building so that people know clearly what matters regulated in the Regional Regulation; (b) appeal to merchants who construct Swallow houses above shop houses (ruko) where their business is to take care of permit changes or additions to buildings in previous Building Permit (IMB) documents; (c) ordering the buildings of Swallow Bird houses established on top of shop houses (ruko) where businesses by traders who are inconsistent with Building Establishment Permit early shop house (ruko) which made the place of business; and (d) seeks to make Regional Regulations specifically regulating swiftlet house construction and control of unauthorized buildings.

    This study aims to find out how the Effectiveness of East Kotawaringin Regent Regulation Number 29 of 2015 Concerning the System and Procedure of Swallow Bird Nest Tax Collection. Therefore this study uses Jurisdiction Sociology namely Legal Research by looking at the prevailing legal norms in the community and relating them to facts that exist in the community and associated with the problems found in this study by measuring certain laws and regulations in response to legal events. happening in the community. The indicators used to describe the Effectiveness of East Kotawaringin Regent Regulation No. 29 of 2015 concerning the System and Procedure of Swallow Bird Nest Tax Collection are the implementation of policies in the form of targets or realization and efforts of regional governments and the inhibiting factors for the implementation of these policies. The results of the research and analysis show that the main problem is the Effectiveness of East Kotawaringin Regent Regulation No. 29 of 2015 concerning the System and Procedure of Swallow Bird Tax Collection. The lack of uneven and not intense data collection and outreach and the lack of awareness of employers to pay taxes. this study illustrates that the Effectiveness of East Kotawaringin Regent Regulation No. 29 of 2015 concerning the System and Procedure of Swallow Nest Tax Collection is effective enough to meet the target of regional income but when conducting field research there are still entrepreneurs who do not pay Swallow tax means that the regulation has been effective but not evenly distributed. the findings of this study indicate that the implementation and implementation factors are still weaknesses that must be addressed by the regional government in the future
