Jeringau merah (Acorus calamus Linn.) dari famili Acoraceaemerupakan tanaman tropis di Indonesia. Data empiris menunjukkanbagian rimpang dari tanaman jeringau merah dapat digunakan untukmengobati Tinea versicolor dan beberapa penelitian telah melaporkanbahwa rimpang jeringau merah memiliki aktivitas antifungal. Tujuan:Penelitian ini bertujuan untuk mengetahui aktivitas antifungal ekstrakmetanol mentah rimpang jeringau merah (Acorus calamus Linn.) terhadappertumbuhan Malassezia furfur, menentukan kandungan senyawa, danmenentukan konsentrasi hambat minimum serta konsentrasi efektif dariekstrak metanol mentah rimpang jeringau merah. Metodologi: Skriningfitokimia menggunakan metode uji kualitatif. Rimpang jeringau merahdiekstraksi secara maserasi menggunakan pelarut metanol. Penelitian inimenggunakan sembilan konsentrasi yaitu 60%, 65%, 70%, 75%, 80%,85%, 90%, 95%, dan 100%. Kontrol positif menggunakan ketokonazol 20µg dan kontrol negatif menggunakan pelarut metanol. Uji aktivitasantifungal dilakukan dengan metode difusi cakram Kirby-Bauer terhadapMalassezia furfur. Hasil: Metabolit sekunder yang terkandung dalamekstrak metanol mentah rimpang jeringau merah yaitu minyak atsiri,alkaloid, saponin, tanin, dan triterpenoid. Ekstrak metanol mentah rimpangjeringau merah memiliki aktivitas terhadap pertumbuhan Malassezia furfurdengan konsentrasi hambat minimum dan dosis efektif pada konsentrasiekstrak 60%. Clinical features is in the form of fine spots that colored white to brown black, this infection is also called a skin fungus with blotches. Treatment of Tinea versicolor can use chemical or traditional medicine. Chemical treatments such as creams or ointments cause sticky taste, while one of the traditional medicine is the bulb and skin of the garlic bulb (Allium sativum Linn) containing allicin substances that are efficacious as an anti-fungal. This research is an experimental research with the aim to know the difference of inhibition zone to Malessezia furfur fungus. There are 3 treatments with 9 repetitions. The extraction method used is the soxhlet method. Anti-fungal activity test was done by using rating method, Saboroud Dextrose Agar (SDA) media which has been inoculated with suspension of Malassezia furfur with concentration 1.5x108 CFU/ml (Mc Farland standard). Result of data analysis obtained by ANOVA p = 0,000 α 0,05 which means bulb extract and garlic bulb skin (Allium sativum Linn) have different effect. International Journal of Pharmacy and Pharmaceutical Sciences, 2014Co-Authors: Yehia A G Mahmoud, Metwally A Metwally, Hanan Mubarak, Nesma El ZewawyAbstract:Objective: Pityriasis versicolor (PV) is a superficial fungal infection of the skin characterized by changes in skin pigment due to colonization of the stratum corneum by a dimorphic lipophilic fungus of the normal flora of the skin, known as Malassezia furfur . Currently, there is no antimycotic drug against Tinea versicolor with high fungicidal activity and without relapse. Methods: Dill seed hot water extract was chosen based upon preliminary studies which included different types of essential oils and other seed extracts where it produced the highest inhibition zone of 60 mm against M. furfur . Dill seed water extract antimicrobial activity was carried out by disc diffusion method. The minimal inhibitory concentration of dill extract was estimated and found to be 6 mg/ml which were used for the formulation of ointment to treat M. furfur infected albino rabbits as an animal model. Results: Topical application of Dill seed extract ointment (6 mg/g) two times daily succeeded in curing severe human Tinea versicolor in a time course of 2 weeks, and with complete healing, which might be used in the treatment regimen after testing its safety. Histopathological effects of Dill seed extract ointment in treating rabbit skin declared no significant toxic effects. Conclusion: Present study indicates that dill seed extract can be used as potential candidates for preparation of antidermatophyic drug formulations and thus may be useful in the treatment of Tinea versicolor inhuman.

molecular typing of malassezia furfur and malassezia globosa isolated from Tinea versicolor patients based on total protein extract and dna finger print profiles
JOURNAL OF ADVANCES IN BIOLOGY, 2014Co-Authors: Yehia A G Mahmoud, Nesma El ZewawyAbstract:The cellular proteins of the 40 studied isolates of M. furfur were electrophoretically analyzed on SDS-PAGE. Analysis of the total cellular protein from M. furfur isolates are grouped into two wide molecular weight range (11 - 112 kDa) . It is very interesting to notice that the band with molecular weight of 56 kDa may be specific to the type of Tinea versicolor. The cluster analysis based on cellular proteins data of M. globosa separated the seven isolates into 2 groups according to the band with molecular weight 56 kDa, which is present in 6 cases (hyperpigmented) and absent in case ( hypopigmented ) lesions. The polymorphism observed from RAPD markers revealed a high degree of genetic diversity in M. furfur and M. globosa isolates collected from different patients having Tinea versicolor infection. Only primer E (5'-TGCCGAGCTG-3') generated a RAPD PCR banding pattern which allowed the identification of 6 different amplification groups within M. furfur and M. globosa that could be used as diagnostic tool for different isolates of Malassezia ..
