ErwinAbstract:Abstract In vitro assays demonstrated that clinical yeasts were significantly more inhibited by Tioconazole (MIC 50 , ⩽0.5 μ g/ml) than by fluconazole (MIC 50 , 8 μg/ml). Tioconazole also exhibited potency against most molds ( Alterneria spp. and Acremonium spp.). All Candida tropicalis isolates had MICs of 8 μg/ml, four-fold greater than any other Candida spp. Generally Gram-negative bacteria were less susceptible to Tioconazole. Moraxella catarrhalis (MIC 90 , 2 μg/ml) was the most susceptible Gram-negative species. Staphylococci and enterococci were the most susceptible to Tioconazole Gram-positive species (MIC 50 s, 1–8 μg/ml). Bacterial species associated with vaginosis. [ Gardnerella vaginalis (MIC 90 , 16 μg/ml), Mobiluncus spp. (MIC 90 , 16 μg/ml) and Prevotella biviadisiens (MIC 90 , 64 μg/ml)] were inhibited by Tioconazole. Isolates of Lactobacillus spp. were most resistant (MIC 90 , ⩾256 μ g/ml) to Tioconazole. Vaginal fluid levels of Tioconazole (mean, 91.4 μg/ml) persisted above the MIC 90 levels (1–64 μg/ml) for most fungal and bacterial pathogens for 72 h in 19 evaluable female human subjects receiving 300 mg Tioconazole in an intravaginal ointment.

Tioconazole in the treatment of vaginal candidosis
Mycoses, 2009Co-Authors: A SchaetzingAbstract:Summary: A 3-day treatment with Tioconazole vaginal tablets (Gyno-Trosid; Pfizer) was used in 30 patients with vaginal candidosis during a prospective, mycologically controlled study. Candida yeasts were identified by microscopy and culture before initiation of treatment and then on days 3, 6 and 28. On the 6th day, all patients had responded to the treatment and there was a 100% cure rate on microscopic and mycological examination. After 4 weeks, 83% of the patients still had relief of symptoms, although a recurrence of Candida on culture and microscopy was detected in 14 patients (46%). Seven of these were available for further assessment and, when given a second course of treatment, they were cured both clinically and microbiologically. There was no evidence of any adverse systemic reaction and local side effects were limited to a transient sensation of mild to moderate burning. Problems relating to recurrence of symptoms and reappearance of Candida are discussed. Zusammenfassung: In einer prospektiven, mykologisch kontrollierten Untersuchung sind 30 an Vaginalmykose leidende Patientinnen 3 Tage lang mit Tioconazol Vaginaltabletten (Gyno-Trosid; Pfizer) behandelt worden. Untersuchungen auf Candida Hefen wurden mittels Mikroskopie und Pilzkultur vor Behandlungsbeginn und danach am 3., 6. und 20. Tag durchgefuhrt. Am 6. Tag hatten alle Patientinnen auf die Behandlung angesprochen, und die mikroskopischen wie auch mykologischen Untersuchungen ergaben eine 100%ige Heilungsrate. Vier Wochen spater waren 83% der Patientinnen noch symptomfrei, obwohl sich bei 14 Frauen (46%) ein erneutes Auftreten von Candida kulturell und mikroskopisch nachweisen lies. Sieben dieser Patientinnen unterzogen sich einer weiteren Kontrolle und wurden klinisch und mikrobiologisch nach einer zweiten Behandlung geheilt. Es ergaben sich keine therapiebedingten nachteiligen Allgemeinerscheinungen, und lokale Nebenwirkungen wurden lediglich als ein vorubergehendes, leichtes bis masig starkes Brennen angegeben. Die Problematik bezuglich des Symptomruckfalls und des erneuten Candida-Nachweises wird diskutiert.
