    Abstract:

    Summary.  We report the case of a 36-year-old man who presented large erythematous plaques involving the trunk, face and extremities, since one month. Some of these lesions had a psoriasis-like aspect, with abundant superficial scaling. Direct microscopic examination of scales removed from the plaques revealed numerous hyphae. Cultural examination lead to the identification of Trichophyton Rubrum species. Our case of tinea corporis presented some peculiarities related both to the extension of the lesions and their clinical aspects. Zusammenfassung.  Wir berichten uber einen 36-jahrigen Mann, der grosseren rotlaufartigen Ausschlag an Rumpf, Gesicht und Gliedmassen seit einem Monat aufwies. Einige dieser Lasionen sahen wie Psoriasis mit reichlichem Abschuppen aus. In der direkten mikroskopischen Kontrolle der kleinen Schuppen aus den Flecken fanden wir viele Hyphen Kulturell wurde Trichophyton Rubrum nachgewiesen. Unser Fall von Tinea corporis zeigte einige Eigenheiten in Hinblick auf den Umfang und das klinische Bild.

  • Detection of Xanthomegnin in Epidermal Materials Infected with Trichophyton Rubrum

    Abstract:

    Xanthomegnin, a mutagenic mycotoxin best known as an agent of nephropathy and death in farm animals exposed to food-borne Penicillium and Aspergillus fungi, was first isolated about 35 y ago as a diffusing pigment from cultures of the dermatophyte, Trichophyton megninii . This study investigates the production of xanthomegnin by the most common dermatophytic species, Trichophyton Rubrum , both in dermatologic nail specimens and in culture. In view of the labile nature of xanthomegnin, a chromatographic procedure was developed to allow high-performance liquid chromatography analysis within 1 h of sample extraction. In cultures, Tricho- phyton Rubrum produced xanthomegnin as a major pigment that appears to give the culture its characteristic red colony reverse. Xanthomegnin was also repeatedly extracted from human nail and skin material infected by Trichophyton Rubrum . The level of xanthomegnin present, however, varied among the clinical samples studied. Xanthomegnin was not detected in uninfected nails. These results show that patients with Trichophyton Rubrum infections may be exposed to xanthomegnin, although the consequences of such an exposure are not currently known.

    Abstract:

    The Trichophyton Rubrum species complex comprises commonly encountered dermatophytic fungi with a worldwide distribution. The members of the complex usually have distinct phenotypes in culture and cause different clinical symptoms, despite high genome similarity. In order to better delimit the species within the complex, molecular, phenotypic, and physiological characteristics were combined to reestablish a natural species concept. Three groups, T. Rubrum, T. soudanense, and T. violaceum, could be distinguished based on the sequence of the internal transcribed spacer (ITS) ribosomal DNA barcode gene. On average, strains within each group were similar by colony appearance, microscopy, and physiology, but strains between groups showed significant differences. Trichophyton Rubrum strains had higher keratinase activity, whereas T. violaceum strains tended to be more lipophilic; however, none of the phenotypic features were diagnostic. The results of matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) and amplified fragment length polymorphism (AFLP) were partially consistent with the ITS data but failed to distinguish the species unambiguously. Despite their close similarity, T. violaceum, T. soudanense, and T. Rubrum can be regarded as independent species with distinct geographical distributions and clinical predilections. Trichophyton soudanense is pheno- and genotypically intermediate between T. Rubrum and T. violaceum For routine diagnostics, ITS sequencing is recommended.

    Abstract:

    The validity of taxa around Trichophyton Rubrum was evaluated by a combination of phenetic and molecular methods. Morphological and physiological features were compared to results of sequencing of the internal transcribed spacer region of the ribosomal operon, PCR fingerprinting, and amplified fragment length polymorphism analysis. The 15 species and varieties investigated (Trichophyton circonvolutum, Trichophyton fischeri, Trichophyton fluviomuniense, Trichophyton glabrum, Trichophyton gourvilii, Trichophyton kanei, Trichophyton kuryangei, Trichophyton megninii, Trichophyton pedis, Trichophyton raubitschekii, Trichophyton rodhaini, Trichophyton Rubrum var. nigricans, Trichophyton soudanense, Trichophyton violaceum var. indicum, and Trichophyton yaoundei) were reclassified or synonymized as T. Rubrum or T. violaceum.

    Abstract:

    Genotypic variability among 96 Trichophyton Rubrum strains which displayed different colony morphologies and were collected from four continents was investigated. Twelve markers representing 57 loci were analyzed by PCR fingerprinting, amplified fragment length polymorphism, and random amplified monomorphic DNA markers. Interestingly, none of the methods used revealed any DNA polymorphism, indicating a strictly clonal mode of reproduction and a strong adaptation to human skin.
