    O maneio reprodutivo é extremamente importante, pois é a principal fonte de ingressos económicos. O controlo da actividade reprodutiva é essencial à organização e à gestão adequada de qualquer exploração animal, pois permite adequar os ciclos de nascimentos e de produção às exigências sazonais do mercado de produtos de origem animal e planificar da melhor o maneio alimentar, o maneio sanitário, a mão-de-obra, a ocupação dos espaços, entre outros. A inseminação artificial permite de forma sustentável aumentar rapidamente o valor genético dos animais. Neste trabalho realizou-se, primeiramente, uma breve revisão bibliográfica sobre a fisiologia da reprodução das ovelhas, os métodos de controlo da actividade reprodutiva, as principais análises seminais efectuadas e a inseminação artificial com sémen fresco e refrigerado e os efeitos de dois diluidores seminais – Andromed® e INRA 96® – sobre a fertilidade. Foram utilizadas 67 ovelhas adultas (2-8 anos) da raça autóctone portuguesa Churra Galega Bragançana. O ciclo éstrico de metade das ovelhas foi sincronizado com um tratamento curto de acetato de fluorgestona (FGA; 20 mg) e eCG (500 UI) e o da outra metade foi-o com um tratamento curto de acetato de acetato de medroxiprogesterona (60 mg) e eCG (500 UI). O sémen foi recolhido por Vagina artificial e depois de avaliado foi diluído com Andromed® ou INRA 96® e preparado para ser inseminado fresco ou refrigerado. Cada dose seminal continha cerca de 200 x 106 espermatozóides. A inseminação artificial foi realizada cerca de 55+1 horas pós-administração de eCG. Procurou-se avaliar alguns dos parâmetros que condicionam o sucesso da inseminação artificial: inseminador, cor da Mucosa da Vagina, grau de lubrificação Vaginal, tipo do oríficio (Os) cervical externo, local de deposição do sémen e se houve refluxo cervical. A deposição do sémen foi feita sempre o mais profunda possível, com o auxílio de fórceps Vaginais, luzes LED, pistoletes Quicklock com baínhas (Minitüb). O dignóstico de gestação foi feito 40 dias pós inseminação artificial. Quando estudo teve início, a maioria das fêmeas estavam cíclicas. Os tratamentos progestagénicos curtos + eCG foram igualmente máximos. A IA às 55+1 horas produziu excelentes resultados. Quarenta dias pós-inseminação 85,1% das ovelhas estavam gestantes. Verificaram-se diferenças estatisticamente significativas entre inseminadores, na forma de conservação do sémen, no local de deposição do sémen, entre a forma de XIII Os cervical e a corrência ou não de refluxo cervical. Os diluidores usados não afectaram a taxa de fertilidade.Reproduction management is highly important since it is the main source of money income. Reproduction control is essential to organize and manage every animal production, since it turns possible the adjustment between production and market demands and the best feed, health, labor and space use planning. Artificial insemination allows a fast and sustainable increase of animal genetic value. This thesis starts with a brief monography in ewes reproduction physiology, reproduction control methods, main semen analysis used, artificial insemination with fresh and cooled semen and the effects of two commercial extenders (Andromed® and INRA 96®) in fertility. Sixty-seven adult (2-8 years old) Portuguese Churra Galega Bragançana ewes were used in this study. Ovarian activity of half ewes was synchronized with a short-term treatment FGA (20 mg) + eCG (500 UI) and the other half with a short-term MAP (60 mg) + eCG (500 UI). Semen was collected with artificial Vaginas. After been analyzed semen was diluted with Andromed® or INRA 96® and prepared to be inseminated fresh or cooled. Each seminal dose had 200 x 106 sperm cells. Artificial insemination was performed 55+1 hours after eCG administration. Different parameters effects (inseminator, Vagina Mucosa color, degree of Vagina lubrication, external cervix Os form, sperm deposition local and cervical possible semen reflux) in fertility. Semen was deposited as deeper as possible in the cervix without damaging the Mucosa wall, using Vaginal forceps, LED lights, Quicklock guns and Minitüb sheaths. Pregnancy diagnosis was performed 40 days after artificial insemination. At the start of this study the majority of ewes were cycling. Both short-term progestogens and eCG treatments were absolute efficient. AI at 55+1 hours resulted in excellent fertility. Forty days after insemination, 85.1% of all ewes were pregnant. Significant statistical differences were found between inseminators, semen preservation, local of semen deposition, Os cervix form and cervical reflux in fertility. Extenders had no effect on fertility

    This paper aims to study the efficiency of two short-term progestagen (FGA vs. MAP) + eCG treatments in estrus synchronization and artificial insemination (AI) with fresh or chilled semen in Assaf ewes fertility rate. All ewes received a subcutaneous implant of exogenous melatonin 45 days before been treated with short-term progestagens + eCG. By June 1st, ewes were divided in two groups: half was treated with an intraVaginal sponge impregnated with 20 mg of FGA and the other half with an intraVaginal sponge impregnated with 60 mg of MAP. Progestagen treatments lasted for 6 days. At sponge withdraw, all ewes were injected with 750 IU of eCG. Ovarian activity was assessed by plasmatic progesterone levels before and after progestagens + eCG treatment. Semen was collected by electro ejaculation and extended with Andromed® or OviXcell®. AI was performed 55 hours after eCG administration with fresh or chilled semen. During AI several factors were assessed: Vagina Mucosa color and lubrication, external cervical Os type, cervical mucous viscosity, semen deposition place and seminal cervix outflow. Semen was deposited as deep as possible without distress or trauma cervix Mucosa. All Assaf ewes presented cyclic activity before progestagen + eCG treatments (2nd fortnight of May). Short-term progestagen + eCG treatments were equally efficient (100.0%). About 76.5% of Assaf ewes were pregnant 41 days after AI. Fertility rate was influenced by external Os type, semen deposition place and seminal cervix outflow.     Mestrado de dupla diplomação com a UTFPR - Universidade Tecnológica Federal do ParanáNo Instituto Politécnico de Bragança a raça local Churra Galega Bragançana Branca tem sido sujeita, nos últimos 3 anos, a um programa sistemático de aplicação da técnica de inseminação artificial (IA) com sémen fresco ou refrigerado. O presente trabalho foi desenvolvido para testar a eficácia dos seguintes factores – administração prévia de melatonina exógena, tratamento progestagénico curto (FGA) + eCG, dois diluidores seminais (INRA 96® vs. OviXcell®) e dois processos de preservação do sémen (Fresco vs. Refrigerado) – e os seus efeitos sobre a taxa de fertilidade aos 41 dias pós-IA. Foram ainda avaliados alguns parâmetros registados quando da IA – cor e lubrificação da Mucosa Vaginal, tipo de Os externo, viscosidade do muco cervical, local de deposição do sémen, refluxo cervical e inseminador “A” X “B”. Na primeira quinzena de Abril, todas as ovelhas Churras Bragançanas Brancas estavam cíclicas. Todas elas responderam ao tratamento FGA + eCG, independentemente de terem sido tratadas ou não com melatonina exógena. Quarenta e um dias após a inseminação artificial 84,1% das ovelhas estavam gestantes. A taxa de fertilidade foi afectada pelo carneiro dador de sémen, pelo tipo de Os externo, pela viscosidade das secreções cervicais e pelo local de deposição do sémen. A taxa de fertilidade não foi influenciada pela administração prévia de melatonina, pelo método de preservação do sémen, pelo tipo de diluidor, pela cor e pelo grau de lubrificação da Mucosa Vaginal, pelo refluxo cervical ou pelo inseminador.Churra Galega Bragançana Branca ewes have been used in a program of artificial insemination (AI) with fresh or cooled semen developed at the Braganza Polytechnic Institute for the last 3 years. Present study aimed to test the efficiency of different factors – exogenous melatonin pre-treatment, short-term progestogen treatment (FGA) + eCG, two seminal extenders (INRA 96® vs. OviXcell®) and two semen preservation procedures (Fresh vs. Refrigerated) – on ovary activity and fertility rate at forty one days post-AI. During AI several issues were assessed: Vagina Mucosa colour and lubrication rate, external Os type, cervical mucus viscosity, semen allocation place, cervical reflux incidence and two inseminators’ skills “A” X “B”. All ewes were cycling at the first half of April. The efficiency rate of short-term progestogen treatment + eCG was 100%. Melatonin previous administration had no effect in the ratio of ovulated ewes. About 84.1% of ewes were pregnant at 41 days after AI. Fertility rate was influenced by male semen donor, external Os type, cervical mucus viscosity and semen allocation place. In opposition fertility rate was not affect by exogenous melatonin pre-treatment, semen preservation procedure, semen extender, Vagina Mucosa color and lubrication, cervical reflux prevalence or inseminator

    Objective:To investigate the anti-hCG antibody level in Vaginal lavage fluid after Vaginal Mucosa immunization with the recombinant lactobacillus excreting the human chorionic gonadotropin beta-subunit, hCGβ.Methods:The recombinant Lb. casei hCGβ was passaged in MRS containing no Erythromycin for 50 generations to get continual secretion. Radio immunol assay(RIA) was used to determine the level of hCGβ in the supernatant. Mice of 8 weeks old were immunized with the recombinant continual expressing strain through Vaginal inoculation. Two weeks later, level of the hCGβ and anti-hCGβ IgA antibody in the Vaginal lavage was determined by radio-immunology assay and indirect ELISA respectively.Results:The hCGβ was secreted by Lb. casei continually. The recombinant hCGβ and anti-hCGβ IgA antibody in the Vaginal lavage were higher than the negative control group(P0.01).Conclusion:Have got stable hCGβ excretion in Lb. casei. Vaginal immunization of recombinant lactobacillus secreting hCGβ could induce the anti-hCGβ IgA antibody at the Vaginal Mucosa.

    Objective In order to improve the treatment of Vagina relaxation, a modified colporrhaphy with the Vagina Mucosa intact is presented. Methods Being exposed with blunt and sharp section from Vaginan Mucosa, both of lateral muscular walls were plicated and tighten, therefore a valvulae was formed on Vagina wall with excessive Mucosa. Results From 1999 to 2004, 26 cases had undergone this modified colporrhaphy successfully and recovered from their Vagina relaxation. Conclusion The modified colporrhaphy with Vagina Mucosa intact is a safe and efficacy technique .

    Three to five per cent acetic acid is commonly used in the field of gynaecology for colposcopic examinations of the cervix. It gives an acetowhite effect that may assist clinicians in identifying neoplastic areas. A perimenopausal woman was treated with acetic acid for abnormal Pap smear report (cervical intraepithelial neoplasia 1). During application the patient complained of burning sensation of Vagina and vulva. The Vagina was saline-irrigated after realising that the acetic acid had not been diluted. Following this incident the patient was seen weekly and treated with conjugated Vaginal oestrogen cream. The patient recovered and the Vagina Mucosa healed within two weeks. Acetic acid is corrosive and may cause Vagina bleeding. Oestrogen cream can be used in an attempt to minimise the adverse reaction and speed the healing process. (authors)
