    Abstract:

    Abstract Ethnopharmacological relevance Veratrum patulum L. (Liliaceae) is used as one source of the Chinese traditional drug “Lilu” which has been used in the treatment of aphasia arising from apoplexy, wind type dysentery, headache, etc . for thousands of years. Aim of the study To evaluate the antithrombotic effect of Veratrum patulum L. (Liliaceae) total steroidal alkaloids (VpA) and explore the potential mechanisms. Materials and methods The antithrombotic effect of VpA was evaluated in two experimental thrombosis models in rat. The potential antithrombotic mechanisms of VpA were explored by determining the coagulation parameters and platelet aggregation induced by various agonists. Cutting-tail rat model was used to evaluate the influence of VpA on bleeding time. Results VpA significantly inhibit the formation of arterial and venous thrombosis and showed different inhibitory effect on the platelet aggregation induced by various agonists with the following potency order: collagen > ADP > thrombin. VpA showed no influence on the coagulation parameters in rat. But VpA decreased the cutting-tail bleeding time in rat. Conclusion VpA possess antithrombotic effect on venous and arterial thrombosis. The antithrombotic effect of VpA is due to its inhibition to platelet aggregation, especially induced by collagen.

    Abstract Two new steroidal alkaloids, veraussines A ( 1 ) and B ( 2 ) were isolated from the roots and rhizomes of Veratrum nigrum var. ussuriense . Their structures were determined as N-(ethoxycarbonyl)-1 α ,2 β ,3 α ,15 α -tetrahydroxy-5 β -jervanin-12-en-11-one ( 1 ) and N -(methoxycarbonyl)-1 α ,2 β ,3 α ,15 α -tetrahydroxy-5 β -jervanin-12-en-11-one ( 2 ) by spectroscopic analysis.

    Abstract:

    Germidine is one of ester type steroidal alkaloids extracted from Veratrum Nigrum L. var. ussuriense Nakai ("Wusuli Lilu" in Chinese), a herbal medicine that has long been used as an anti-hypertensive remedy in Chinese traditional medicine. Although the antithrombotic effects of the Veratrum nigrum alkaloids (VnA) have been reported , the effective chemical constituent of VnA is still unclear. In view of this fact, we investigated the inhibitory activity of germidine on thrombosis in rats. In rat carotid artery thrombosis model, germidine (iv 1.25-20.0 µg·kg -1 ) significantly and dose-dependently prolonged the occlusion time (OT) of carotid artery injured by electrical stimulation. In rat inferior vena thrombosis model, germidine (iv 2.5-20.0 µg·kg -1 ) decreased the thrombus dry weight in a significant and dose-dependent manner. LD50(iv) in mice of germidine was found to be 6.6619 mg·kg -1 , a value much greater than that of VnA. We conclude that germidine has powerful inhibitory effects against both arterial and venous thrombosis in rats and plays an important role in the antithrombotic effects of VnA.

    Abstract:

    Veratrum nigrum L., a traditional Chinese herb, has been used for treatment of hypertension, blood-stroke, excessive phlegm, epilepsy, etc. Steroidal alkaloids were well-known as both bioactive and toxic constituents of Veratrum species, the toxicity of which the traditional processing procedure can reduce. To reveal the mechanism of processing V. nigrum L., a high performance liquid chromatography (HPLC) with evaporative light scattering detector (ELSD) method was developed for the simultaneous determination of ten steroidal alkaloids in crude and processed V. nigrum L., comparison with ultrasound extract of crude V. nigrum L. With a Venusil XBP-C(18) analytical column, the analytes were separated efficiently using the mobile phase consisted of (A) 0.03% aqueous triethylamine (TEA) and (B) acetonitrile in a gradient program. The parameters for ELSD were set: S.C. (Spray Chamber)=35 degrees C, D.T. (Drift Tube)=70 degrees C, GAS=50 psi. All calibration curves showed good linear regression (gamma>or=0.9990) within the tested range. Additionally, reproducibility for the quantification of ten alkaloids in V. nigrum L. with intra- and inter-day variations of less than 5.0% was observed. The obtained alkaloid profiles performed by this newly established method, provided valuable information for the differentiation of crude and processed V. nigrum L. and for the explanation of the different toxicity.

    A new steroidal alkaloid ( 1), together with three known steroidal alkaloids ( 2 - 4), has been isolated from the roots and rhizomes of Veratrum japonicum (Baker) Loes. f.., their structures were established as neogermine ( 1), germine ( 2), germerine ( 3), and neogermbudine ( 4) by means of physicochemical properties and spectroscopic analysis. Compounds 1 - 4 exhibited genotoxicity on brain cells in mice by using single-cell gel electrophoresis (Comet assay).

    Two new steroidal alkaloids, veramanine and neojerminalanine, have been isolated from the rhizomes of Veratrum album, along with the known glycoalkaloid, pseudojervine, whose NMR spectral data are reported for the first time. The structures of 1 and 2 were elucidated on the basis of extensive spectroscopic investigations.

    Abstract Leaves of Veratrum album of Turkish origin have yielded two new steroidal alkaloids, O -acetyljervine and methyljervine- N -3′-propanoate along with the known alkaloid jervine. The structures of these new bases were established on the basis of 2D NMR and other spectroscopic techniques.

    Alkaloids comprise one of the largest groups of plant secondary metabolites including vinca alkaloids. The ability of six alkaloids from Veratrum lobelianum, one from Veratrum nigrum and three from Peganum nigellastrum to modify transport activity of MDR1 was studied. Flow-cytometry in a multidrug-resistant human MDR1-gene-transfected mouse lymphoma cells (L5178Y) was applied. The inhibition of multidrug resistance was investigated by measuring the accumulation of rhodamine-123 in cancer cells. Veralosinine and veranigrine were the most effective resistance modifiers. In a checkerboard method veralosinine and veranigrine enhanced the antiproliferative effects of doxorubicin on MDR cells in combination. The structure-activity relationships were discussed.

    Twelve steroidal alkaloids were isolated from four populations of Veratrum lobelianum Bernh. and Veratrum nigrum L. Full NMR data for veralosinine (1), and extensive 1H NMR data for veralosine (3) and teinemine (5) are presented here for the first time. (+/-)-15-O-(2-Methylbutyroyl)germine (10) is undescribed up to now. The antiproliferative activities of veranigrine, veralosinine, and neogermitrine have shown that they are a perspective for further studies.
