Determination of Neutral Urinary 17-Oxosteroids with Hydroxide of Hyamine 1O-X in the Zimmermann

The organic base, hydroxide of Hyamine lO-X,#{176}which is stable in methanol, has replaced the unstable ethanolic potassium hydroxide solution in the Zimmermann reaction. Maximal color development was studied by varying incubation temperature, time, m-dinitrobenzene, and concentration of hydroxide of Hyamine 10-X. An incubation temperature of 370 for 30 mm., with 1 % m-dinitrobenzene in 1 M Hyamine 10-X hydroxide solution, gave close to maximal color. The major urinary 17-Oxosteroids, dehydroepiandrosterone, androsterone, and etiocholanolone, gave similar chromogenic values. However, C3 and C20 Oxosteroids produced some color in the Zimmermann reaction, and thus if present in large amounts, interfere. The interference in vitro of 35 drugs was tested, and several drugs were found to interfere. An essentially pigment-free urinary extract was obtained during the washing step, by addition of water to the ethylene dichloride extract containing sodium hydroxide pellets. Comparative 17-Oxosteroid studies of urine specimens from laboratory personnel and from patients were performed, based on absorbance at a single wavelength (520 m) and at three wavelengths (Allen correction). MASSIVE PREGNANETRIOL EXCRETION DUE TO VIRILIZING ADRENAL TUMOUR
2016Co-Authors: W. H. Atwill, R. D. Ensor, J. F. GlennAbstract:Urinary 17-Oxosteroid values in excess of 100 mg./24 hr. are not uncommonly associated with virilizing tumours of adrenal origin. However, the secretion of pregnanetriol in amounts greater than 50 mg./24 hr. is unusual. The patient reported here exhibited the highest level of pregnanetriol excretion due to adrenal tumour without metastases ever recorded in the English literature. A 47-year-old Negro woman presented with a 10-month history of hirsutism, darkening of the facial skin, periorbital oedema, abdominal swelling, and right flank pain, progressively severe and debilitating. Twenty years previously, total abdominal hysterectomy and bilateral salpingo-oophorectomy had been accomplished for pelvic inflammatory disease. Physical examination revealed a round-faced hirsute Negress in no distress with blood pressure of 150/105 mm. Hg, atrophic breasts, masculine abdominal and pubic hair distribution and patchy hyperpigmentation of the face and arms. The liver was palpable 4 cm. below the right costal margin and was only slightly tender. The clitoris was 3 cm. in length and the vaginal cuff and adnexal regions were unremarkable

Prostaglandin Involvement in the Endocrine Activity of the Testis
2015Co-Authors: Akio AmanoAbstract:were investigated in mature male dogs. The secretory activity was determined by measuring 17-Oxosteroids in spermatic vein blood. Seventeen-Oxosteroid secretion in animals which received either PGE2 (5 µg/kg) or the administration. Concomitantly blood flow through the spermatic vein increased to 3.4 times of preinjection levels 5 minutes after the injection of either prostaglandin. However, neither of these events occurred when either prostaglandin (10 µg/kg) was given by intravenous route. When either prostaglandin (1 µg/kg) was given intratestically at the time when the rate of testicular 17-Oxosteroid secretion caused by an intravenous injection of human chorionic gonadotrophin (HCG, 20 i.u./kg) was at a maximum, the response to HCG was significantly was extremely weak. Administration into the testicular tissue of adenosine-3 ' , 5'-cyclic monophosphoric acid (cyclic AMP, 1 mg/kg) in combination with PGE2 (5 µg/kg) produced marked increases in 17-Oxosteroid secretion and spermatic vein blood flow.
