highly photoluminescent carbon dots based immunosensors for ultrasensitive detection of Aflatoxin M1 residues in milk

Then, a label-free immunosensor based on the inner filter effect of carbon dots was developed for ultrasensitive detection of Aflatoxin M1 residues in milk. The detection limit was 0.0186 ng/mL (equivalents to 18.10 ng/kg), which satisfied the most stringent maximum tolerable limit value of 25 ng/kg. Besides, the immunosensor showed a good linear relationship from 0.003 ng/mL to 0.81 ng/mL, and the average recoveries ranged from 79.6% to 112.5% for spiked milk samples, with relative standard deviations ranging from 6.7% to 13.3%. Compared with other immunoassays, the inner filter effect-based immunosensor incorporating fluorescent detection into conventional enzymatic cascade amplification systems and could be a reliable on-site screening method for Aflatoxin M1 residue analysis.

Nowadays, multiplex immunoassay is a primary goal throughout the world. Here, Aflatoxin M1 and pirlimycin were selected as models, and a novel dual colorimetric encoded frit-based immunoassay was developed for simultaneously screening of Aflatoxin M1 and pirlimycin residues in milk. This multiplex frit-based immunoassay combined two monoclonal antibodies to extend the spectrum of analytes and to enable detection of two classes of analytes in a single test. The cutoff values were 0.02 μg/kg for Aflatoxin M1 and 0.5 μg/kg for pirlimycin, which satisfied the requirement to measure the maximum residue levels. The novel colorimetric frit-based immunoassay has the advantage of high throughput, short analysis time, reduced overall cost per assay, and can be used as a rapid screening technique for simultaneously detecting Aflatoxin M1 and pirlimycin residues in milk.

Food Control, 2011Co-Authors: Kursat Kav, Ramazan Col, Kaan K TekinsenAbstract:Abstract Aflatoxin M1 (AFM1) is the hydroxylated metabolite of Aflatoxin B1 found in a variety of foods. In this study, 127 samples of white-brined Urfa cheese produced mainly in the southeast of Turkey from raw ovine and bovine milks were surveyed for the presence of AFM1 using a competitive Enzyme Linked Immunosorbent Assay (ELISA) technique. The results showed that at detectable levels (≥50 ng/kg), 36 cheese samples (28.3%) were contaminated with AFM1 ranging from 70.61 to 770.97 ng/kg. Of the 36 cheese samples, 13 (10.2%) were found to have levels that exceeded the legal limits of 250 ng/kg established by the Turkish Food Codex. Consequently, the AFM1 contamination levels determined in this study in white-brined Urfa Cheese, which is commonly consumed in the southeast part of Turkey, were not considered to be a serious public health hazard. It was considered to be a potential risk for customers, particularly for infant health.

Aflatoxin M1 levels in uht milk and kashar cheese consumed in turkey
Food and Chemical Toxicology, 2008Co-Authors: Kaan K Tekinsen, Semih H EkenAbstract:In the present study, 100 UHT milk and 132 kashar cheese samples were analyzed for Aflatoxin M1 (AFM1). They were obtained from retail outlets in five big cities (Istanbul, Izmir, Konya, Tekirdag, Edirne). The occurrence and concentration range of AFM1 in the samples were investigated by competitive enzyme-linked immunosorbent assay (ELISA) method. Sixty-seven percent of the UHT milk samples and 82.6% of the kashar cheese samples contained AFM1. The positive incidence of AFM1 in the UHT milk samples and the kashar cheese samples ranged from 10 to 630 ng/kg and from 50 to 690 ng/kg, respectively. AFM1 levels in 31 (31%) UHT milk and 36 (27.3%) kashar cheese samples exceeded the maximum tolerable limit of the EC and the TFC. AFM1 levels in the samples show that there is a presence of high Aflatoxin level that constitutes a human health risk in Turkey. Therefore milk and dairy products have to be controlled continuously for presence of AFM1 contamination by the Turkish public health authorities.

Food and Chemical Toxicology, 2010Co-Authors: Aziz A FallahAbstract:This study was conducted as a screening survey to determine the occurrence of Aflatoxin M1 (AFM1) in 225 commercial liquid milk samples composed of pasteurized milk (116 samples) and UHT milk (109 samples) obtained from popular markets in central part of Iran. The competitive enzyme-linked immunosorbent assay method was employed to determine level of the toxin in the samples. Aflatoxin M1 was detected in 151 (67.1%) samples, consisted of 83 (71.5%) pasteurized milk samples (mean: 52.8 ng/l; range: 5.8-528.5 ng/l) and 68 (62.3%) UHT milk samples (mean: 46.4 ng/l; range: 5.6-515.9 ng/l). Considering the US FDA and Iranian national standard limits for AFM1 in milk (500 ng/l), 2 samples (1.7%) of pasteurized and 3 samples (2.7%) of UHT milk had levels above the maximum tolerance limit. However, according to European Commission limit (50 ng/l), this figure increased to 31 (26.7%) and 19 (17.4%) samples for pasteurized and UHT milk, respectively. The results indicated that the contamination of the samples with AFM1 in such a level could be a serious public health problem at the moment. This paper represents the data of the first survey on the occurrence of AFM1 in commercial liquid milk marketed in central part of Iran.

Food and Chemical Toxicology, 2009Co-Authors: Aziz A Fallah, Tina Jafari, Ali Fallah, Mohammad Rahim RahnamaAbstract:A screening survey on the occurrence of Aflatoxin M1 (AFM1) was accomplished on 210 cheese samples composed of white cheese (116 samples) and cream cheese (94 samples) purchased from popular markets in central part of Iran (Esfahan and Yazd provinces). The quantitative analysis of AFM1 levels in the samples was performed by using the competitive enzyme-linked immunosorbent assay (ELISA) technique. Aflatoxin M1 at measurable level (50 ng/kg) was detected in 161 (76.6%) samples, consisting of 93 (80.1%) white and 68 (72.3%) cream cheese samples. The concentration of AFM1 in the samples ranged from 52.1 to 785.4 ng/kg. Comparing to legal regulation (250 ng/kg) accepted by some of the countries, 24.2% of the samples exceeded the accepted limit. Among these, the AFM1 levels in 28.4% of white and 19.1% of cream cheese samples were not in accordance with the safety limit. The results indicated that contamination of the samples with AFM1 in such a level appear to be a potential hazard for public health. This paper represents the data of the first survey on the occurrence of AFM1 in cheeses consumed in central part of Iran.

Aflatoxin M1 in milk and dairy products, occurrence and recent challenges: A review

Ahmad FaizalAbstract:Background: Milk is a highly nutritious food, and it is a source of necessary macro- and micronutrients for the growth, development and maintenance of human health. However, it may also be a source of natural food contaminants that may cause disease. The presence of Aflatoxin M1 (AFM1) in milk and dairy products throughout the world has been known since twenty to thirty ago. Milk and dairy products contamination with Aflatoxin M1 is important problem worldwide especially for developing countries for the last ten to twenty years. Scope and Approach: The presence of this mycotoxin in these products is important issue, especially for children and infants, who are more susceptible than adults. This review provides information regarding the occurrence of AFM1 in milk and dairy products in many regions of the world, its stability during processing and some reduction strategies. In this review the toxicity, occurrence of AFM1 in milk and dairy products (preferably for the last 5 years), regulations, strategies for its reduction, latest developments in detection methodologies and future challenges are described. Key Findings and Conclusions: Strict regulations and adapting good storage practices in developed countries have minimized the contamination of AFM1 in milk and dairy products. The current advancements in analytical techniques have helped the law enforcement agencies to implement strict regulations. Furthermore, the improvement in analytical facility and increasing the awareness related to the health effects of AFM1 in milk and dairy products could minimize its occurrence level in developing countries.

The results have shown that AFM1 was detected in 71% of milk samples with 58% samples were found above the permissible limit of European Union (EU). AFM1 was detected in 61% of yogurt, 78% white cheese, 59% cheese cream and 45% butter samples with 47, 15, 11 and 52% samples of yogurt, white cheese, cheese cream and butter, respectively were found above the recommended limit of EU. The data of the present study will be helpful for the implementation of regulatory limit for AFB1 in order to minimize or avoid AFM1 in milk and milk products from Pakistan and also gives insights that whether the occurrence of AFM1 in dairy products was considered as a possible risk for consumers health.
