This study extends the knowledge on bioactivities of F. palmata, A. eupatoria, A. procera, F. ulmaria and F. vulgaris by comprehensive characterization of their methanolic extracts. Antioxidant properties of extracts were evaluated by DPPH• (2,2-diphenyl-1-picrylhydrazyl), ABTS•+ 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) scavenging and oxygen radical absorbance capacities (ORAC). Genotoxicity of extracts was tested using alkaline single-cell gel electrophoresis (comet) and cytokinesis-block micronucleus assays in human lymphocytes in vitro and the Ames Salmonella/microsome test. All investigated Agrimonia and Filipendula extracts possessed strong antioxidant activity, which was comparable with that of a standard antioxidant trolox (6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid). Thirty five compounds belonging to the classes of phenolic acids, flavonoids, phenylpropanoids and ellagitanins were detected by ultra-performance liquid chromatography – mass spectrometry (UPLC-Q-TOF-MS). Agrimonia and Filipendula extracts induced an increase in a DNA damage in the comet assay expressed as mean percentage of DNA in the comet tail. However, these extracts did not produce reverse mutation in bacterial cells in the Ames test and were not genotoxic in the micronucleus test. However, a slight though significant decrease of nuclear division index values was determined. In general, this study proved that Agrimonia and Filipendula species are a good source of bioactive compounds; their extracts may be classified as non-mutagenic and non-clastogenic in vitro under conditions of the current study. Consequently, the plants may be a promising material for nutraceuticals and natural medicines.

Assessment of radical scavenging capacity of Agrimonia extracts isolated by supercritical carbon dioxide

Two agrimony species were extracted in laboratory scale equipment at various parameters and it was found that in case of A. eupatoria the increase in extraction pressure and the addition of a co-solvent ethanol remarkably increases the yield of the extracts, up to 4.25% at 25 MPa and 1% of ethanol. The highest yield from A. procera was 3.39% at the same conditions; however the dependence of pressure on extract yield was not so evident. In general, the extracts isolated at higher pressure were more effective free radical scavengers, although this dependence for some extracts, especially obtained from A. procera was rather complicated. The concentration of phenolics in A. eupatoria and A. procera SC-CO2 extracts was 2.3-8.3 and 2.5-7.1 mg/g, respectively. The compositions of SFE and organic solvent extracts were totally different. Radical scavenging capacity of agrimony extracts varied in a wide range (9.1−97.5% in DPPH reaction and 6.7−79.5% in ABTS reaction) depending on the polarity of the solvent used to obtain the extract.

Decoction can be used in traditional medicine for a treatment of diabetes mellitus or coronary diseases (Gray and Flatt, 1998; Correia et al., 2006). Inhibitors of α-glucosidase are promising drugs in the treatment of diabetes mellitus, and antioxidants from natural sources can prevent the oxidative stress associated with diabetes mellitus. The individual with diabetes has a 2- to 6-fold increased risk of cardiovascular disease (Wolff, 1993) and Agrimonia herbs could exert a beneficial effect in the diabetic environment. A. eupatoria is the most common Agrimonia species in Europe and the aerial parts are used in form of infusion, decoction or tincture. The scientific investigations are focused on research of the most common A. eupatoria and A. pilosa (Jung et al., 2010; Kato et al., 2010; Lee et al., 2010), there are no information about species growing in eastern Asia – A. japonica and A. coreana. We found the highest inhibition of cholinesterase in the extract of A. japonica with inhibition 70.4% for AChE and 79.8% for BuChE. These findings are statistically significant in comparison with those of other extracts (p < 0.001). The phytochemical analyses showed that the antioxidant activity of Agrimonia extracts can be affected especially by hexahydroxydiphenoyl (HHDP)-glucose and quercetin glycosides, and inhibition of cholinesterases by apigenin, luteolin and quercetin glycosides.

Plants were collected in Medicinal Herbs Centre of Masaryk university Brno and these were Agrimonia eupatoria L., Agrimonia procera Wallr., Agrimonia leucantha Kunze, Agrimonia japonica (Miq.) Koidz and Agrimonia coreana Nakai. HPLC profile of Agrimonia herbs shows high content of quercetin and apigenin glycosides. The content of total polyphenols correlates with antioxidant activity and the content of flavonoids correlates with inhibition of a- glucosidase . Agrimonia procera works as an excellent antioxidant, the most used in traditional medicine Agrimonia eupatoria is richer on flavonoid glycosides.
