Knowledge of NY-ESO-1 Expression has depended on RT-PCR detection of mRNA and there is a need for detecting NY-ESO-1 at the protein level. In the present study, a method for the immunochemical detection of NY-ESO-1 in paraffin-embedded tissues has been developed and used to define the Expression pattern of NY-ESO-1 in normal tissues and in a panel of human tumors. No normal tissue other than testis showed NY-ESO-1 reactivity, and Expression in testis was restricted to germ cells particularly spermatogonia. In human tumors, the frequency of NY-ESO-1 Antigen Expression corresponds with past analysis of NY-ESO-1 mRNA Expression e.g., 20-30% of lung cancers, bladder cancers and melanoma, and no Expression in colon and renal cancer. Co-typing of NY-ESO-1 Antigen and mRNA Expression in a large panel of lung cancers showed a good correlation. There is great variability in NY-ESO-1 Expression in individual tumors, ranging from an infrequent homogeneous pattern of staining to highly heterogeneous Antigen Expression.

To our knowledge, the possibility that the levels of these Antigens vary with time has not been previously assessed. We determine if the pattern and/or intensity of blood group Antigen Expression on vaginal epithelial cells and mucus changed with time. Cell and mucus specimens were collected weekly for 3 months from 10 women: 5 (2 secretors and 3 nonsecretors) with and 5 (3 secretors and 2 nonsecretors) without a history of urinary tract infections. In addition, samples were collected on 5 consecutive days from 5 of these individuals. The cell and mucus samples were assayed for ABH and Lewis blood group Antigens using monoclonal antibodies in cell concentration immunofluorescence and enzyme-linked fluorogenic assays, respectively. Blood group Antigens, carbohydrate structures bound to lipids or proteins, constitute an important component of the uroepithelial cell membrane and may affect susceptibility to urinary tract infections. To determine if the ABH and Lewis Antigen Expression in women with a history of urinary tract infections differed from that of women without such a history, vaginal specimens from 87 women (42 patients and 45 controls) were analyzed for the detection of these Antigenic determinants using monoclonal antibodies in quantitative immunoassays. The profile of ABH Antigen Expression was generally consistent with the ABO phenotype of the individual and appeared to be influenced by the secretor status. Their Expression in various human cancer types has been interpreted as ‘re-Expression’ or as intratumoral progenitor cell signature. Cancer testis Antigen Expression patterns have not yet been studied in germ cell tumorigenesis with specific emphasis on intratubular germ cell neoplasia unclassified as a precursor lesion for testicular germ cell tumors. Immunohistochemistry was used to study MAGEA3, MAGEA4, MAGEC1, GAGE1 and CTAG1B Expression in 325 primary testicular germ cell tumors, including 94 mixed germ cell tumors. Seminomatous and non-seminomatous components were separately arranged and evaluated on tissue microarrays. Spermatogonia in the normal testis were positive, whereas intratubular germ cell neoplasia unclassified was negative for all five CT Antigens. Cancer testis Antigen Expression was only found in 3% (CTAG1B), 10% (GAGE1, MAGEA4), 33% (MAGEA3) and 40% (MAGEC1) of classic seminoma but not in non-seminomatous testicular germ cell tumors. In contrast, all spermatocytic seminomas were positive for cancer testis Antigens. These data are consistent with a different cell origin in spermatocytic seminoma compared with classic seminoma and support a progression model with loss of cancer testis Antigens in early tumorigenesis of testicular germ cell tumors and later re-Expression in a subset of seminomas.

Frequently, immunophenotyping identifies myeloid Antigen Expression in ALL, causing a potential diagnostic problem. To evaluate the immunophenotype of ALL, we studied 210 cases of pediatric and adult ALL by flow cytometry and compared the results with the French-American-British (FAB) Cooperative Group classification and the karyotypic findings. Myeloid-associated Antigens were expressed in 78 (45.6%) of precursor B-cell ALL cases. Pediatric precursor B ALLs had a higher frequency of myeloid Antigen Expression than did adult cases. All mature B-cell ALL cases were negative for TdT and myeloid Antigens. Myeloid Antigen Expression was less frequent in T-cell ALL cases compared with precursor B-cell ALL cases. Of the 192 cases submitted for cytogenetic analysis, 147 were abnormal. The most common chromosomal translocation was the Philadelphia chromosome, which was more likely to have L2 blast morphology and a precursor B immunophenotype. Myeloid Antigen Expression was present in 70.8% of Ph-positive cases (P = .008). Chromosome rearrangements involving 11q23 also showed an increased frequency of myeloid Antigen Expression. Chromosome translocations involving regions of T-cell receptor genes were present in 24% of T-cell ALL cases. A high percentage of ALL cases, however, had various other cytogenetic abnormalities, many of which involved less well-studied chromosomal regions.

Two tissue microarrays constructed with 220 and 778 CRC tumors were stained with HLA-DR, DQ, and DP Antigen-specific monoclonal antibody LGII-612.14, using the immunoperoxidase staining technique. The immunohistochemical staining results were correlated with the clinical course of the disease. The functional role of HLA class II Antigens expressed on CRC cells was analyzed by investigating their in vitro interactions with immune cells. HLA class II Antigens were expressed in about 25% of the 220 and 21% of the 778 tumors analyzed with an overall frequency of 23%. HLA class II Antigens were detected in 19% of colorectal adenomas. Importantly, the percentage of stained cells and the staining intensity were significantly lower than those detected in CRC tumors. However, HLA class II Antigen staining was weakly detected only in 5.4% of 37 normal mucosa tissues. HLA class II Antigen Expression was associated with a favorable clinical course of the disease. In vitro stimulation with interferon gamma (IFNγ) induced HLA class II Antigen Expression on two of the four CRC cell lines tested. HLA class II Antigen Expression on CRC cells triggered interleukin-1β (IL-1β) production by resting monocytes. HLA class II Antigen Expression in CRC tumors is a favorable prognostic marker. This association may reflect stimulation of IL-1β production by monocytes.
