    Abstract Background Atopic dermatitis (AD) is a common disease with genetic and environmental interactions. We previously reported lifetime exposure to cigarette smoke is associated with adult-onset AD. Aryl hydrocarbon receptor (AhR) is important in regulating environmental exposure to xenobiotics, including Benzopyrenes (BP), a major polycyclic aromatic hydrocarbon (PAH) present in cigarette smoke. However, how AhR regulates immune responses in sensitization phase of AD remained elusive. Methods We investigated how BP affects epicutaneous sensitization response through AhR axis. We compared AhR expression in skin from AD patients and healthy controls. We measured immune responses (Langerhans cell migration and T cell polarization in epicutaneous Ova sensitization in mice with or without AhR defect. Results We found AhR and ARNT (AhR nuclear translocator) are upregulated in AD skin. BP exposure increases Langerhans cell migration, and increases IL-5, IL-13, and IL-17 levels when lymph node cells were re-challenged with Ova. The increased cytokine levels were attenuated in AhR defected mice. AhR agonists (BP and ITE) decreased E -cadherin expression, while AhR antagonist (CH223191) increased it in human primary keratinocytes. Conclusions These results suggested AhR interacts with BP to polarize T cell responses, along with Langerhans cell migration. This study revealed a regulatory mechanism how cigarette smoking affects atopic sensitization through the Benzopyrene-AhR interaction.

    Abstract Evidence in humans and laboratory animals supports a cancer‐protective effect of vitamin A, but the mechanism remains unclear. While vitamin A deficiency causes squamous metaplasia, and lung cancer patients have lower vitamin A status, their serum vitamin A levels are not indicative of deficiency. We hypothesize that local enzymatic degradation of vitamin A can be induced by exposure to carcinogens such as Benzopyrene found in cigarette smoke. This study was designed to determine if Benzopyrene exposure depletes tissue vitamin A and whether β‐carotene might prevent the depletion. Weanling male Fischer rats were fed a nutritionally complete purified diet, supplemented with or without Benzopyrene at 400 mg/kg feed or β‐carotene at 2 g/kg feed. Vitamin A content of the liver, small intestine, and serum was determined by high‐performance liquid chromatography.     Cancer prevalence is increasing nowadays. In Indonesia, cancer prevalence reaches1.4% out of 1,000 people. Cancer is mainly caused by two factors; genetic and environment, including a carcinogenic agent known as Benzopyrene. Physical exercises can decrease the levels of blood glucose and fatty acid within the blood; can increase expression of Bax and caspase-3; but its effect on GADD45 and wild p53 expression, is still unknown. This study aimed to determine the expressions of GADD45 and wild p53 resulting from moderate swimming exercise on Mus musculus injected by Benzopyrene. 18 mice were divided into three study groups; control group 1 (K1) which was not treated with any physical exercise and Benzopyrene; control group 2 (K2) which was not treated with any physical exercise but induced with 0.08 mg of Benzopyrene; and treated group (K3) which wastreated with physical exercise in moderate intensity and induced with 0.08 mg of Benzopyrene. The buccal mucosa tissue samples were taken and stained for immunohistochemistry to be further examined under a light microscope at 400x magnification with 10 different angles. Result: There was no difference inGADD45 expression between the K1, K2, and K3 groups (p = 0.611), with a significant difference of wild p53 expression (p = 0.000).     Cancer is one of the leading causes of death throughout the world. In Indonesia in 2013 it reached 1.4% or an estimated 347,792 people and became the cause of death number seven. Squamous cell carcinoma is the most common cancer in the oral cavity, the patients survival is less than 50%. Until now complete cancer treatment has not been done while every year new cases continue to emerge,despite advances in the field of surgical therapy, radiation, chemotherapy is growing rapidly.Pomegranate (Punicagranatum L.)/PgL, is one of the plants with the active ingredient Ellagic Acid(EA), which has anti-cancer activity in vitro. There has not known the effectiveness of Pomegranateif given in whole extract or Ellagic acid isolation. Purpose of this study was to prove the effect of Pomegranate extract (Punicagranatum L./PgL) and Ellagic Acid on the expression of VEGF andoral cancer cells apoptosis of Musmusculus due to Benzopyrene induction. 24 mice (Balb / c),males, 5 months old, were divided randomly into 3 groups, namely: K0: not induced byBenzopyrene, but not given PgL and EA; K1: Benzopyrene induced, not given PgL and EA; P1:Benzopyrene induced and given EA; P2: Benzopyrene induced and given PgL. Benzopirene at adose of 0.04 mg was dissolved in 0.04 mL olivarum oil, given 3 times a week for 4 weeks. The application of PgL and EA at a dose of 75 mg/kg BW/day for 4 weeks.Examination of VEGF expression by immunohisto chemical techniques, while cell apoptosis with tunnel assay. There was significant difference in VEGF expression between all groups (p = 0.006), also in cancer cellapoptosis there were significant differences between all groups (p = 0.001). Pomegranate (Punicagranatum L.) extract was more effective to decrease VEGF expression and increase apoptosis cancer cell than Ellagic acid on Musmusculus which inducted by Benzopyrene

    Aim: The aim of this study was to analyze the effect of moderate exercise on the elevation of Bax/Bcl-2 ratio. Materials and Methods: Eighteen Mus musculus strain Swiss Webster (Balb/c) were divided into three groups (n=6). K1 and K2 had contact with water 3 times/week for 12 weeks, while the members of the K3 group swam 3 times/week for 12 weeks while carrying load weighed 3% of their body weight. After 5 weeks, they were induced with 0.04 ml oleum olivarum (K1), 0.08 mg Benzopyrene/0.04 ml oleum olivarum (K2, K3) 3 times/week for 4 weeks. Immunohistochemistry assays were used to determine the ratio of Bax/Bcl-2 expression. The results were analyzed using an independent t-test. Result: The Bax/Bcl-2 ratio increased significantly in K3 compared to K2 (p=0.00). Conclusion: Moderate exercise could increase the Bax/Bcl-2 ratio in oral squamous epithelial cells induced by Benzopyrene.
