Metallothionein (MT) is a cysteine-rich protein that can influence the detoxification of heavy metals and scavenge oxidative stress for free radicals. One of the most expressive functional genes in humans is the MT2A gene. Objective This study aims to determine if the association of the Blood Lead Level and lipid biomarkers was influenced by MT2A polymorphisms. Methods We recruited 677 participants after informed consent was obtained. All the samples collected were analyzed for lipid biomarkers and Blood Lead Levels and were genotyped for MT2A polymorphisms by reverse transcription polymerase chain reaction. A short questionnaire collected the medical history and alcohol and cigarette consumption information. The data were used for descriptive analyses and linear regression models. Results The investigation revealed that Lead elevated concentration increased low-density lipoprotein cholesterol and decreased high-density lipoprotein cholesterol (HDL-C) by multiple linear models. The carriers of the rs10636 GC-rs28366003 AA genetic combination may be less susceptive to Lead elevated concentration on HDL-C than other types. Conclusion In conclusion, the association of the Blood Lead Level and HDL-C may be modified by the MT2A genetic combination: the rs10636 GC-rs28366003 AA genotype could play a protective role in Lead elevated concentration on HDL-C in humans.

The present study was conducted to elucidate the extent of cord Blood Lead Level decline in the Taipei area from 1985 to 2002 and to explore the factors affecting the cord Blood Lead Level after the cease of Leaded gasoline use. In the current study period of 2001–2002, 184 of 1310 newborns delivered in the Taipei Municipal Women and Children Hospital between September 2001 and August 2002 were eligible and randomly selected to participate in this study. Neither of their parents had an occupational Lead exposure history. At each delivery, a sample of 5–10 mL umbilical cord Blood was collected for Lead determination by graphite furnace atomic absorption spectrometry. The cord Blood Lead Level of the newborns in the current study period averaged 2.35±1.12 μg/dL. The objective of this study was to investigate and compare cord Blood Lead Levels (CBLLs) in newborns in Flint, Michigan, after the Flint water crisis, to a group of Detroit newborns. Study Design Mothers of 99 Flint newborns were surveyed about potential Lead exposures. These neonates were born after the recognition of population-wide Lead-in-water contamination. CBLLs were measured and maternal–fetal metrics were reviewed. CBLLs and maternal–fetal metrics were then compared with those of a retrospective cohort of 116 Detroit newborns who previously shared the same water source. Analysis involved descriptive statistics, independent t-test, and χ 2 analysis. Results CBLLs greater than or equal to 1 μg/dL (0.05 μmol/L) were more prevalent among Flint newborns (14%), as compared with Detroit newborns (2%; p = 0.001). This was a sevenfold disparity between Flint and Detroit newborns. No statistically significant differences were found in birth weight, head circumference, small for gestational age status, gestational age, or preterm status among the two groups. Conclusion The Flint water crisis potentially exposed newborns to Lead in utero, implicating maternal–fetal outcomes and future health and development. Primary prevention efforts, including identification and mitigation of Lead exposure before conception and during pregnancy, are needed. New environmental exposure detection methods and long-term neurodevelopmental follow-up will complement the findings of this study.

Neurotoxicology and Teratology, 2000Co-Authors: Lourdes Schnaas, Stephen J Rothenberg, Estela Perroni, Sandra Martinez, Carmen Hernandez, Reyna M HernandezAbstract:Abstract To determine the temporal pattern of the effect of postnatal Blood Lead Level on the General Cognitive Index (GCI) of the McCarthy Scales of Children's Abilities, we used data from 112 children of the Mexico City Prospective Lead Study with complete evaluations from 36 to 60 months of age at 6-month intervals. We measured Blood Lead Level every 6 months from 6 to 54 months. We controlled for 5-min Apgar, birth weight, birth order, sex, socioeconomic Level, maternal IQ, and maximum maternal educational Level in a repeated measures ANCOVA using child Blood Lead Level grouped by 6–18 month (geometric mean 10.1 μg/dl, range 3.5–37.0 μg/dl), 24–36 month (geometric mean 9.7 μg/dl, range 3.0–42.7 μg/dl), and 42–54 month (geometric mean 8.4 μg/dl, range 2.5–44.8 μg/dl) averages. There were significant interactions between the 6–18 month Blood Lead Level and age with GCI as the endpoint and between 24–36 month Blood Lead Level and age. The regression coefficient of Blood Lead at 6–18 months became more negative with age until 48 months, when the rate of decline moderated (linear polynomial contrast p =0.047). The regression coefficient of Blood Lead at 24–36 months with CGI became more negative as well from 36 to 48 months but then started decreasing toward zero from 48 to 60 months (quadratic polynomial contrast p =0.019). Significant between-subjects Lead effects on GCI were found for 24–36 month Blood Lead Level at 48 months ( p =0.021) and at 54 months ( p =0.073). The greatest effect (at 48 months) was a 5.8-point GCI decrease with each natural log unit increase in Blood Lead. Significant between-subjects Lead effects on GCI were found for 42–54 month Blood Lead Level at 54 months ( p =0.040) and at 60 months ( p =0.060). The effect of postnatal Blood Lead Level on GCI reaches its maximum approximately 1–3 years later, and then becomes less evident. Four to five years of age appears to be a critical period for the manifestation of the earlier postnatal Blood Lead Level effects.

There was only one previous report of a significant association of Blood Lead with Blood pressure in pregnant women. We measured Blood Lead Level and sitting Blood pressure of 1 627 women in their third trimester of pregnancy. We eliminated subjects with known causes of hypertension. Most women (98.4%) were normotensive. We controlled for body mass index, age, and stress—among other factors—and constructed multiple-regression models of Lead association with diastolic and systolic Blood pressures. Immigrants (73% of total) had significantly higher Blood Lead Levels and different Blood pressures than nonimmigrants, suggesting that analysis be stratified by “immigrant, nonimmigrant” status. However, poor minority groups that live in large urban centers are still at high risk for elevated Lead Levels. In this study, our data showed that pregnant immigrants (n = 1 428) who live in South Central Los Angeles–one of the most economically depressed regions of California–have significantly higher (p < .0001) Blood Lead Levels (geometric mean = 2.3 μg/dl [0.11 μmol/l]) than 504 pregnant nonimmigrants (geometric mean = 1.9 μg/dl [0.09 μmol/l]). Supplementation of vitamin C is beneficial to reduce the oxidative stress and to improve the antioxidant status of these workers. AIM The main aim of this study was to observe the changes in Blood Lead Levels, oxidative stress i.e. serum lipid peroxide and antioxidant status parameters such as erythrocyte superoxide dismutase and catalase and serum nitrite after the vitamin C supplementation in battery manufacturing workers. MATERIALS AND METHODS This study included 36 battery manufacturing workers from Western Maharashtra, India, having age between 20-60 years. All study group subjects were provided vitamin C tablets (500 mg/day for one month) and a Blood sample of 10 ml each was drawn by puncturing the anterior cubital vein before and after vitamin C supplementation. The biochemical parameters were estimated by using the standard methods. RESULTS Blood Lead Levels were not significantly altered, however, serum lipid peroxide (p<0.001, -15.56%) and serum nitrite (p<0.001, -21.37%) Levels showed significant decrease and antioxidant status parameters such as erythrocyte superoxide dismutase (p<0.001, 38.02%) and catalase (p<0.001, 32.36%) revealed significant increase in battery manufacturing workers after the supplementation of vitamin C. CONCLUSION One month vitamin C supplementation in battery manufacturing workers is not beneficial to decrease the Blood Lead Levels. However, it is helpful to reduce the lipid peroxidation and nitrite formation and enhances the erythrocytes superoxide dismutase and catalase activity.
