    Contagious caprine pleuropneumonia (CCPP) caused by Mycoplasma capricolum subsp. capripneumoniae is a severe epidemic affecting mainly domestic Caprinae species but also affects wild Caprinae species. M. capricolum subsp. capripneumoniae belongs to the "Mycoplasma mycoides cluster." The disease features prominently in East Africa, in particular Kenya, Tanzania, and Ethiopia. CCPP also endangers wildlife and thus affects not only basic nutritional resources of large populations but also expensively built-up game resorts in affected countries. Here, we report the complete sequences of two M. capricolum subsp. capripneumoniae strains: the type strain F38 and strain ILRI181 isolated druing a recent outbreak in Kenya. Both genomes have a G+C content of 24% with sizes of 1,016,760 bp and 1,017,183 bp for strains F38 and ILRI181, respectively.

    The 'Mycoplasma mycoides cluster' comprises the ruminant pathogens Mycoplasma mycoides subsp. mycoides the causative agent of contagious bovine pleuropneumonia (CBPP), Mycoplasma capricolum subsp. capripneumoniae the agent of contagious caprine pleuropneumonia (CCPP), Mycoplasma capricolum subsp. capricolum, Mycoplasma leachii and Mycoplasma mycoides subsp. capri. CBPP and CCPP are major livestock diseases and impact the agricultural sector especially in developing countries through reduced food-supply and international trade restrictions. In addition, these diseases are a threat to disease-free countries. We used a multilocus sequence typing (MLST) approach to gain insights into the demographic history of and phylogenetic relationships among the members of the 'M. mycoides cluster'. We collected partial sequences from seven housekeeping genes representing a total of 3,816 base pairs from 118 strains within this cluster, and five strains isolated from wild Caprinae. Strikingly, the origin of the 'M. mycoides cluster' dates to about 10,000 years ago, suggesting that the establishment and spread of the cluster coincided with livestock domestication. In addition, we show that hybridization and recombination may be important factors in the evolutionary history of the cluster.

    Abstract:

    Sarcoptic mange is a cosmopolitan disease affecting the skin of domestic and wild mammalian species and humans as well. In Eurasia, sarcoptidosis (also known as sarcoptic mange or scabies) affects mountain ungulates (Caprinae) among other wild hosts, and epizootic outbreaks induce variable mortality rates. This fact, coupled with the important ecological and socio-economic values of such mammalian hosts, resulted in many research projects being focused on addressing ecological, physiological, behavioural, genetic, and pathological effects of the disease. Nevertheless, information about management of sarcoptic mange in free-ranging populations is scarce and scattered, with contradictory results and a lack of consensus on basic aspects of the disease. In this review, we summarise knowledge on the effects of sarcoptic mange in wild Caprinae, at individual, pathological and population epidemiological levels, as well as on the current tools and management strategies for its detection, diagnosis, prevention, and control. Disease spread in na�ve populations is ca. 6�km�year, and the mortality rate can be >95%. Tools for monitoring the disease include visual diagnosis, photographic traps, trained dogs, thermography, immunodiagnostics, molecular tools, radiocollars, and epidemiological modelling. Options for management include eradication, control, and prevention of the disease; biosecurity and prevention of spread to humans can be achieved by careful hygiene methods. Sarcoptic mange is a natural, biological factor controlling host population numbers and dynamics in Caprinae, so goals and strategies for its management in wild populations must be set accordingly. Specific management programmes for preventing and controlling sarcoptic mange in wild Caprinae populations must be based on reliable epidemiological data. More research is needed to provide evidence-based policies. The efficacy and safety of various management approaches remain to be tested experimentally

    Background Infectious keratoconjunctivitis (IKC) is a clinical condition affecting eyes of domestic and wild Caprinae worldwide, and Mycoplasma conjunctivae is considered the primary causative agent of IKC in sheep, goats and wild Caprinae. Domestic ruminants from high mountain habitats share grazing areas with wild mountain ungulates, such as chamois (Rupicapra spp.), Alpine ibex (Capra ibex) and European mouflon (Ovis aries musimon), and domestic sheep seem to act as M. conjunctivae reservoir. In this study, the presence of M. conjunctivae in domestic sheep and goats from the two main mountain ranges of Northern Spain, the Pyrenees and the Cantabrian Mountains, has been investigated.

    Sarcoptic mange represents the most severe disease for wild Caprinae individuals and populations in Europe, rising concerns for both conservation and management of these ungulates. To date, this disease has been investigated in different wild caprine species and under many different perspectives including diagnostics, epidemiology, impact on the host populations and genetics of both hosts and parasite, with the aim to disentangle the host-Sarcoptes scabiei relationship. Notwithstanding, uncertainty still remains and basic questions still need an answer. Among these, the effect of immunological responses on mange severity at an individual level, the main drivers in host-parasite interactions for different clinical outcomes and the role of the immune response in determining the shift from epidemic to endemic cycle. A deeper approach to pathology of this disease seems therefore advisable, all the more reason considering that immune response to S. scabiei in wild Caprinae, generally classified as a hypersensitivity, still remains poorly understood. In this paper, we started a journey into the pathological features associated to sarcoptic mange in wildlife, exploring different kinds of hypersensitivity and outcomes, with the final goal to highlight the major drivers in the different responses to this disease at an individual level and propose some key topics for future research, with a particular attention to Alps-dwelling wild caprines.
