Centaurium erythraea Extracts Exert Vascular Effects through Endothelium- and Fibroblast-dependent Pathways.

After a preliminary screen of extracts obtained from sequential extraction of C. erythraea aerial parts, effects of the methanolic fraction (MFCE) on changes in perfusion pressure of isolated rat mesenteric vascular bed (MVB) and in rat cardiac fibroblasts proliferation were investigated, gathering information on the mechanisms involved in endothelium-dependent effects and their dependence on a pro-proliferative stimulus. The HPLC-DAD determination of the phenolics content of MFCE revealed the presence of 22 phenolic compounds. MFCE reduced (63.3 ± 3.9%; n = 4) perfusion pressure in MVB and almost completely abrogated the Ang II-induced increase in cardiac fibroblasts proliferation. Reduction of the perfusion pressure caused by MFCE was endothelium-dependent and occurred in parallel with an increase in NO release. These effects were inhibited by muscarinic receptor antagonists, by L-NAME (a NO synthase inhibitor), and by ODQ (a soluble guanylate cyclase inhibitor). Experiments revealed that effects required the involvement of K+ channels, being inhibited by tetraethylamonium (TEA; a Ca2+ activated K+ channels inhibitor) and by glibenclamide (an ATP-sensitive K+ channels inhibitor). In conclusion, extracts from C. erythraea, particularly the compounds present in the MFCE, induce endothelium-dependent vasodilation and prevent fibroblast proliferation induced by angiotensin II, which can account for the claimed antihypertensive effects of C. erythraea in traditional medicine.

Moreover, this extract revealed to have some effect on the inhibition of 5-lipoxygenase, no cytotoxicity against RAW 264.7 macrophage cells line, and a weak potential to decrease the nitric oxide levels in this cell system. The results suggest that C. erythraea aerial parts could be valuable sources of health benefits compounds, contributing to its valorisation and its further application in functional foods.

Hydroxyl radical and hypochlorous acid scavenging activity of small centaury (Centaurium erythraea) infusion. A comparative study with green tea (Camellia sinensis)

This species is reported to contain considerable amounts of polyphenolic compounds, namely xanthones and phenolic acids as the main constituents. Although the antiradicalar activity of some pure polyphenolic compounds is already known, it remains unclear how a complex mixture obtained from plant extracts functions against reactive oxygen species. Thus, the ability of small centaury infusion to act as a scavenger of the reactive oxygen species hydroxyl radical and hypochlorous acid was studied and compared with that of green tea (Camellia sinensis L.). Hydroxyl radical was generated in the presence of Fe3+-EDTA, ascorbate and H2O2 (Fenton system) and monitored by evaluating hydroxyl radical-induced deoxyribose degradation. The reactivity towards hypochlorous acid was determined by measuring the inhibition of hypochlorous acid-induced 5-thio-2-nitrobenzoic acid oxidation to 5,5'-dithiobis(2-nitrobenzoic acid). The obtained results demonstrate that small centaury infusion exhibits interesting antioxidant properties, expressed both by its capacity to effectively scavenge hydroxyl radical and hypochlorous acid, although with a lower activity against the second than that observed for green tea. Green tea exhibited a dual effect at the hydroxyl radical scavenging assay, stimulating deoxyribose degradation at lower dosages.

Somatic Embryogenesis in Centaurium erythraea Rafn-Current Status and Perspectives: A Review.

Marković, Milica Bogdanović, Angelina SubotićAbstract:Centaurium erythraea (centaury) is a traditionally used medicinal plant, with a spectrum of secondary metabolites with confirmed healing properties. Centaury is an emerging model in plant developmental biology due to its vigorous regenerative potential and great developmental plasticity when cultured in vitro. Hereby, we review nearly two decades of research on somatic embryogenesis (SE) in centaury. During SE, somatic cells are induced by suitable culture conditions to express their totipotency, acquire embryogenic characteristics, and eventually give rise to somatic embryos. When SE is initiated from centaury root explants, the process occurs spontaneously (on hormone-free medium), directly (without the callusing phase), and the somatic embryos are of unicellular origin. SE from leaf explants has to be induced by plant growth regulators and is indirect (preceded by callusing). Histological observations and culture conditions are compared in these two systems. The changes in antioxidative enzymes were followed during SE from the leaf explants. Special focus is given to the role of arabinogalactan proteins during SE, which were analyzed using a variety of approaches. The newest and preliminary results, including centaury transcriptome, novel potential SE markers, and novel types of arabinogalactan proteins, are discussed as perspectives of centaury research.

Plant regeneration in leaf culture of Centaurium erythraea Rafn. Part 3: de novo transcriptome assembly and validation of housekeeping genes for studies of in vitro morphogenesis

(common centaury, Gentianaceae) is a medicinal plant with great regeneration potential and developmental plasticity in vitro. Centaury can be regenerated from leaf explants by both somatic embryogenesis (SE) and shoot development (SD). We believe that its regeneration potential and developmental plasticity rest on high activity of certain genes, which may not be active or present in species recalcitrant to in vitro regeneration. However, there are no sequenced Gentianaceae genomes to support investigation of the molecular events during SE or SD. To this end, we have sequenced six centaury transcriptomes (embryogenic calli, globular somatic embryos, cotyledonary somatic embryos, adventitious buds, leaves and roots of in vitro grown plants) and de novo assembled centaury referent transcriptome comprising 105.726 genes. The high quality and completeness transcriptome was functionally annotated against NCBI nt, Swissprot and PFAM databases with KOG and GO enrichment. In addition, 11 housekeeping and functional genes were validated for expression stability in 27 tissue samples representing the processes of SE and SD, plants from nature and wounded tissues using GeNorm and NormFinder. The most stable genes that can be used for expression studies during SE, SD and in vitro manipulations are Ribosomal protein L2 (RPL2) and TATA binding protein 1 (TBP1) in combination with RAS (Rat Sarcoma)-related Nuclear protein (RAN) or Adenosine kinase (AK). These results comprise a complete framework for the search for genes involved in SE and SD, but may also be useful in identifying genes involved in biosynthesis of C. erythraea secondary metabolites. De novo assembled and fully annotated Centaurim erythraea transcriptome is publically available along with validated housekeeping genes with stable expression during in vitro development and mechanical injury.

Antimicrobial activity, antioxidant potential and total phenolic content of transgenic atckx1 centaury (Centaurium erythraea rafn.) plants grown in vitro

Centaury has been used for centuries in traditional medicine. Secondary metabolites such as bitter secoiridoid glucosides (gentiopicrin, swertiamarin and sweroside), xanthones (eustomin and demethyleustomin), and phenolic acids are the main constituents responsible for the therapeutic properties of centaury. Previous investigation showed that overexpression of the AtCKX genes in transgenic centaury plants did not result in a decrease of total cytokinin (CK) content, but in an altered CK profile leading to a decline of bioactive, the most important physiologically active group of CKs.The aim of this study was to investigate antibacterial and antifungal activity of transgenic centaury methanol extracts as well as pure secoiridoid and xanthone compounds on four Gram positive, four Gram negative bacteria and eight species of microfungi. All tested methanol extracts of control and transgenic AtCKX1 centaury shoots and roots showed better antibacterial activity, while pure compounds (gentiopicrin, swertiamarin, eustomin and demethyleustomin) showed better antifungal activity. The results obtained in this work suggest that centaury methanol extracts and pure compounds represent potential antimicrobials confirming the possibility of using these compounds in agronomy, veterinary, medicine or food industry.

Endogenous Phytohormones in Spontaneously Regenerated Centaurium erythraea Rafn. Plants Grown In Vitro

Spontaneous morphogenesis of the common centaury ( Centaurium erythraea Rafn.) is possible on nutrition medium without addition of any plant growth regulator depending solely on endogenous phytohormone levels. Thus, this plant species represents a very good model system for the investigation of numerous physiological processes under phytohormonal control in vitro. We analysed the total amount of endogenous cytokinins (CKs) including the contents of their individual groups in shoots and roots of C. erythraea plants grown in vitro. The total amount of endogenous CKs was 1.4 times higher in shoots than in roots. Inactive or weakly active N -glucosides found to predominate in both organs of centaury plants, whereas free bases and O -glucosides represented only a small portion of the total CK pool. Consequently, centaury roots showed higher IAA content as well as IAA/free CK base ratios compared to shoots. Centaury tissues also showed increased levels of “stress hormones”. In contrast to SA, considerably higher levels of ABA were found in centaury shoots than in roots. Our results could serve as a basis for understanding and elucidating spontaneous de novo shoot organogenesis and further plant regeneration of C. erythraea in vitro.

The Role of Arabinogalactan Proteins in Morphogenesis of Centaurium erythraea Rafn In Vitro
The Gentianaceae - Volume 2: Biotechnology and Applications, 2015Co-Authors: Milana Trifunovic, Marija Petrić, Angelina Subotić, Sladjana JevremovicAbstract:Histochemical localization using the β-Glc Yariv reagent and immunolocalization with arabinogalactan protein (AGP) reactive antibodies (LM2, JIM13, JIM15, JIM16, MAC207) were performed during morphogenic induction in root cultures of Centaurium erythraea Rafn cultured on half-strength MS medium without plant growth regulators. The observations revealed that β-Glc Yariv reagent specifically bound to AGPs in cells of the root epidermis and central cylinder. Monoclonal antibodies recognizing AGPs were localized in epidermal cells and cells of the central cylinder (LM2 , JIM16), vascular tissue (JIM15), globular somatic embryos (LM2, MAC207), and de novo-formed meristematic centers in the root cortex (JIM16). The effect of β-Glc Yariv reagent was investigated after supplementation (0–75 μM) in the culture medium. The morphogenetic potential was increased at lower concentrations (15–25 μM) of treatment with β-Glc Yariv reagent but inhibited (40 %) at the highest concentration. These results implicate that AGPs play a significant role during the development of somatic embryos and adventitious shoots in root cultures of C. erythraea.

Methoxylated xanthones in the quality control of small centaury (Centaurium erythraea) flowering tops.

Lourdes Bastos, Rosa M SeabraAbstract:In the course of a phytochemical study of the bitter tonic plant, small centaury (Centaurium erythraea), six methoxylated xanthones (1,5-hydroxy-3-methoxyxanthone, 1-hydroxy-3,5,6-trimethoxyxanthone, 1-hydroxy-3,5,6,7-tetramethoxyxanthone, 1-hydroxy-3,5,6,7,8-pentamethoxyxanthone, 1-hydroxy-3,7,8-trimethoxyxanthone and 1,8-dihydroxy-3,5,6,7-tetramethoxyxanthone) were isolated and identified by spectroscopic means (nuclear magnetic resonance, mass spectroscopy, and UV). Subsequently, a high-performance liquid chromatography/diode array detection method was developed for the determination of these and other methoxylated xanthones occurring in the chloroform extract of small centaury aerial parts. The methodology developed was applied to twelve samples, and in all of them, nine xanthones were identified and quantified. (Gentianaceae) has long been used in traditional medicine. This plant contains considerable amounts of polyphenolic compounds, namely, xanthones and phenolic acids as the main constituents. Because phenolic groups exhibit activity as radical scavengers and/or metal chelators, this study evaluated the superoxide radical scavenging properties of a lyophilized infusion obtained from C. erythraea flowering tops. Superoxide radical scavenging activity was assayed using enzymatic (xanthine/xanthine oxidase) and nonenzymatic (NADH/phenazine methosulfate) superoxide generating systems. This study provided evidence that C. erythraea exhibits interesting antioxidant properties, expressed either by the capacity to scavenge superoxide radical or to noncompetitively inhibit xanthine oxidase. The main phenolic compounds present in this extract were several esters of hydroxycinnamic acids, namely, p-coumaric, ferulic, and sinapic acids. In vitro culture type and elicitation affects secoiridoid and xanthone LC–ESI–TOF MS profile and production in Centaurium erythraea

In vitro culture based C. erythraea production would offer sustainable and economically feasible alternative to wild plant harvest provided that comparable productivity could be achieved in artificially propagated plants. To identify most productive C. erythraea system the secoiridoid glycoside and xanthone production and profiles using liquid chromatography-mass spectrometry were assessed in in vitro shoot, callus and cell suspension culture-derived material in comparison with wild C. erythraea plants. In vitro shoot culture was the most productive system for C. erythraea secoiridoid production (total yield 86 mg g^−1 dw) and cell culture was the most productive system for phenolic compound production (19 mg GAE g^−1 dw). The effect of elicitors yeast extract, methyljasmonate (MeJA) and chitosan on secondary metabolite yield indicated that elicitation with MeJA and chitosan significantly decreased yield of sweroside and eustomin, respectively. Significant quantitative differences of major secoiridoids and xanthones indicated that further methodological improvements for increasing yield of major secondary metabolites of in vitro origin need to be explored for in vitro cultivation to become a feasible alternative to C. erythraea wild plant harvest.
