    Abstract:

    Cystophorous Cercariae developing in rediae in Antalis entalis (L.) (Syn.: Dentalium entale ) (Mollusca, Scaphopoda) were found in Norwegian waters off Bergen. The second intermediate (crustacean) hosts are unknown. Experimental infections of copepods and various malacostracans with the immotile cystophorous Cercariae were unsuccessful. In one instance, three Cercariae developed into metaCercariae in one specimen of Antalis entalis kept in an aquarium for 11 months. The Cercaria and redia are described using light microscopy and scanning electron microscopy. Serial sections of rediae revealed a vestigial pharynx and caecum. The Cercaria is compared with other cystophorous Cercariae from scaphopods and other molluscs. The redia and Cercaria differ slightly from those of Cercaria prenanti Arvy, 1949 in Antalis entalis from France, but are very similar to Cercaria prenanti from the American Pacific. The metaCercaria is very similar to young specimens of Lecithophyllum botryophorum (Olsson, 1868) Odhner, 1905...

    Abstract Embryonated eggs of Lecithochirium rufoviride (Rudolphi, 1819) Luhe, 1901 (Hemiuridae) from the stomach of the European eel, Anguilla anguilla (L.), developed into Cercariae in agreement with the description of Cercaria vaullegeardi Pelseneer, 1906 in the prosobranch Gibbula cineraria (L.) (Trochidae). Since 1906, when C. vaullegeardi was originally described from G. cineraria no snail has apparently been found naturally infected with this species. The cystophorous Cercaria of L. rufoviride is compared with those of L. furcolabiatum (Jones, 1933) Dawes, 1947 and L. fusiforme Luhe, 1901, which develop in other species of Gibbula and whose Cercariae previously have been identified as Cercaria vaullegeardi.     Abstract:

    Objective: To investigate the prevalence of pleurophocercous Cercariae infection in thiarid snails from natural infection in 12 provinces of Northern Thailand. Methods: The snail specimens were collected and examined for pleurophocercous Cercariae infection using crushing method. The morphological characteristics of Cercariae, which were identified by a light microscope and illustration were done using a camera lucida. Results: Based on a sample of 2 075 thiarid snails of four snail species, it was found that Melanoides tuberculata, Tarebia granifera, Thiara scabra, and Adamieta hoesei served as the first intermediate hosts of heterophyid trematode with an overall prevalence of 14.78%, revealing four different morphological characteristic types. Pleurophocercous Cercaria Type I had the highest the prevalence (13.06%), followed by the pleurolophocercous Cercaria Type II (0.29%), pleurolophocercous Cercaria Type III (0.05%), and pleurophocercous Cercaria Type IV (1.4%), respectively. Conclusions: This research report will serve a preliminary step providing new and fundamental information regarding the current geographical distribution trends of opisthorchiid and/or heterphyid trematodes infection. The findings of this study will be useful to the development of effective control measures for this type of parasitic infection in Northern Thailand

    : Trematode Cercariae are commonly found in many freshwater gastropods. These Cercariae can serve to identify the occurrence of such trematodes as Centrocestus formosanus, Haplorchis taichui, Haplorchoides sp, and Stellantchasmus falcatus, which are important parasites in Chiang Mai Province, Thailand. As the species of these Cercariae cannot be identified accurately based on morphology, this study employed sequencing of a fragment of 28S ribosomal DNA and phylogenetic analysis to identify the trematode Cercariae found in freshwater gastropods in Chiang Mai Province. Eight types of trematode Cercariae were identified, namely, distome Cercaria (grouped with Philophthalmus spp clade), echinostome Cercaria (grouped with Echinostoma spp clade), furcocercous Cercaria (grouped with Posthodiplostomum sp/Alaria taxideae/Hysteromorpha triloba clade), monostome Cercaria (grouped with Catatropis indicus clade), parapleurolophocercous Cercaria (grouped with Haplorchoides sp clade), pleurolophocercous Cercaria (grouped with Centrocestusformosanus clade), transversotrema Cercaria (grouped with Transversotrema spp clade), and xiphidioCercaria (grouped with Prosthodendrium spp clade). These results provide important information that can be used for identifying these parasites in epidemiological surveys.

    Minute intestinal fluke,  Haplorchis taichui , remain clinical importance, especially in the north-eastern and northern regions of Thailand. For obtaining an effective epidemiological control program, a sensitive, accurate, specific detection are required. Sequences of Internal transcribed Spacer subunit 2 were performed to identify Cercarial trematodes in snail intermediate hosts.The results showed that phylogram depicting phylogenetic relationships was constructed based on combined sequence data of ITS2 and showed that, 2 distinct clusters were formed by first containing with the group of middle-larger sizes trematode while the remaining was minute size with all of them were belonged to family Heterophyidae. Within the minute size cluster,  H.taichui  and  Pleurolophocercous Cercaria  were placed in the same branch which can confirm the identities of PC found in this study that they will be developed and identified to  H. taichui .     The investigation of biological diversity of intestinal trematode in snails which the report concerned is rarely and not extended in current status. Especially in minute intestine trematode, Haplorchis taichui Witenberg, 1930, family of Heterophyidae, which was found in the small intestine of mammals including human, and causes serious clinical problem worldwide. So, this study was aimed to investigate H. taichui infection in freshwater snails from Mae Taeng basin, Chiang Mai, Thailand. Total of 1,836 snails were collected during April 2008 to August 2012. Cercarial infection was examined by crushing method. Molecular identification of H. taichui were conducted by a DNA specific primer which amplified the mCOI gene. The PCR product of mCOI were sequenced and confirmed by BLAST program. Six types of Cercariae were found viz. megalurous, furcocercous, monostome, pleurolophocercous, parapleurolophocercous, and virgulate Cercaria. The parapleurolophocercous Cercaria in Melanoides tuberculata, Tarebia granifera, and Thiara scabra were larval stages of H. taichui which yield the specific fragment of 256 bp. Overall of H. taichui infection of snails was 42.71%. The mCOI sequences had 99% similarity with the 29 isolated gene references of the H. taichui in Genbank data base. The molecular method had suitability as an epidemiological tool for suitable control programs against the dissemination of trematodes.

    Freshwater snails from difference microhabitat including rice filed and stream, from Mae Taeng district, Chiang Mai province, were collected during April to June 2009. A total of 193 snail samples comprised of 5 species were found. These were 60 Tarebia granifera, 38 Melanoides tuberculata, 30 Thaira scabra, 37 Filopaludina martensi martensi, and 28 Bithynia siamensis siamensis. The cercaraie were investigated by crushing method where they were classifieds into 4 types: pleurolophocercous Cercaria, distome Cercaria, transversotrema Cercaria and monostome Cercaria. The Cercarial infection rates in above five species of snail were 48.33% (28:60), 47.36% (18:38), 13.33% (4:30), 0.00% (0:30), and 21.42% (8:28), respectively. The molecular identification using the 256 bp amplicon specific-polymerase chain reactions (specific - PCR), were performed and revealed that pleurolophocercous Cercariae was as an intestinal trematode, Haplochis taichui. The Hap-t_F and Hap-t_R specific primer, the 256 bp amplicon was generated, which was shown to have a positive result. The total Cercarial infection rates of H. taichui were 8.30% (16:193). This study would be represented some application base on epidemiological monitoring and detections in snail intermediate hosts which usefulness for preventing, management and epidemiological control programs.

    The minute intestinal trematode, Haplorchis taichui , is an important parasite species that can infect humans and other mammals. This study investigated the outbreak of H. taichui in thiarid snails in the lower part of the Chao Phraya Basin, Thailand by employing morphological and molecular-based methods. In development of a specific primer of H. taichui , the PCR reaction was conducted with no cross-reaction to their hosts and other related trematode species. The highest level of sensitivity that could be amplified was 0.50 ng/μl and this was detected with only one egg in the sample. In terms of the epidemic results, the parapleurolophocercous Cercaria infected only two species of thiarid snails ( Melanoides tuberculata and Tarebia granifera ) with an overall prevalence of 3.80% (23/605). The process of molecular identification revealed positive results indicating that eleven from twenty-three of parapleurolophocercous Cercariae specimens in the lower part of the Chao Phraya Basin were H. taichui . In conclusion, this study has developed a rapid detection method, which can discriminate H. taichui from other parapleurolophocercous Cercaria in intermediate snail hosts with a high level of sensitivity. Moreover, the high proportion of H. taichui in parapleurolophocercous Cercaria (47.83%) indicated that H. taichui was the dominant species of this Cercarial type and could infect cyprinoid fish in the lower part of the Chao Phraya Basin leading to public health problems in this area. Thus, a specific primer could be useful in the detection and surveillance of H. taichui outbreaks in their hosts. Recognition of this has resulted in the creation of important prevention programs in these infected areas in the further study.

  • Morphological Characteristics and Phylogenetic Trends of Trematode Cercariae in Freshwater Snails from Nakhon Nayok Province, Thailand.

    : The prevalence of Cercarial infection in freshwater snails and their evolutionary trends were studied in Nakhon Nayok province, Thailand. A total of 2,869 individual snails were examined for parasitic infections. The results showed that 12 snail species were found to host larval stages of trematodes with an overall prevalence of 4.7%. The infected specimens included 7 types at the Cercarial stage; Cercariae, megalurous Cercariae, echinostome Cercariae, furcocercous Cercariae, parapleurolophocercous Cercariae, virgulate Cercariae, and xiphidioCercariae. Regarding molecular identification, ITS2 sequence data of each larval trematode were analyzed, and a dendrogram was constructed using the neighbor-joining method with 10,000 replicates. The dendrogram was separated into 6 clades (order/family), including Echinostomatida/Echinostomatidae, Echinostomatida/Philophthalmidae, Opisthorchiida/Heterophyidae, Plagiorchiida/Prosthogonimidae, Plagiorchiida/Lecithodendriidae, and Strigeatida/Cyathocotylidae. These findings were used to confirm morphological characteristics and evolutionary trends of each type of Cercariae discovered in Nakhon Nayok province. Furthermore, this investigation confirmed that the ITS2 data of Cercariae could be used to study on phylogenetic relationships or to determine classification of this species at order and/or family level when possible.

    Objective: To analyze the prevalence of trematode infections in the snails intermediate host Tarebia granifera and Melanoides tuberculata collected from Ratchaburi Province by a molecular approach in order to construct a dendrogram to identify species at the Cercarial stage. Methods: Specimens were collected from nine districts located in Ratchaburi Province using the stratified sampling method. The samples were examined for Cercarial infection using the crushing method. All specimens were amplified by region of internal transcribed spacer 2 (ITS2) based on the PCR technique, and the sequence data were aligned in order to construct the dendrogram by maximum likelihood with 10 000 bootstrap replicates. Results: Five Cercarial types were observed comprising parapleurolophocercous Cercaria, xiphidioCercaria, megarulous Cercaria, furcocercous Cercaria and transversotrema Cercaria. The overall prevalence of Cercarial infection was found to be 7.92% (63 / 795). The parapleurolophocercous Cercaria was found in the highest level of prevalence followed by xiphidioCercaria, megarulous Cercaria, furcocercous Cercaria and transversotrema Cercaria, respectively. The parapleurolophocercous Cercaria samples were identified and separated into Haplorchis taichui and Haplorchis pumilio, while xiphidioCercaria were identified as Lecithodendriidae and megarulous Cercaria were identified as Philophthalmus. The dendrogram separated the Cercariae into five groups, which were comprised of parapleurolophocercous Cercaria, xiphidioCercaria, megarulous Cercaria, furcocercous Cercaria and transversotrema Cercaria using the sequence of Angiostrongylus cantonensis as an out-group. Conclusions: This is the first study to report on the prevalence of Cercarial infection in Ratchaburi Province, Thailand. The high prevalence of Cercariae was revealed by the infection in Tarebia granifera and Melanoides tuberculata. Thus, the sequence data of ITS2 can be used to investigate the dendrogram of trematodes and can help to separate the collected samples at the species level. This information can be used to create the prevention program for parasite infection from intermediate hosts in the future

    Schistosomes are parasitic worms that have a complex life cycle. The larval stage Cercaria, infectious to mammals, is described as highly susceptible to the complement system, largely due to the glycocalyx that covers the Cercarial membrane. In an attempt to have a more complete understanding of Cercaria reaction to the complement system, three different approaches were used. Live Cercariae exposed to normal human serum (NHS) as source of complement factors were assessed for (i) membrane attack complex (MAC) deposition on the parasite surface, (ii) Cercaria survival rate by Hoechst staining of parasite DNA, and (iii) transformation into schistosomula by detection of the glucose transporter protein 4 (SGTP4), a marker for new tegument formation. We found that 82–95 % of Cercariae directly exposed to NHS for 18 h were viable and retained their ability to shed the glycocalyx, suggesting minimal tegument damage. In contrast, inhibition of glycocalyx shedding using eserine caused significant MAC binding and parasite death. Culturing complement-exposed Cercariae to measure long-term survival showed that more parasites died over time, reaching a survival rate of 18–31 % by day 6 in culture. The reason for this slow death is unknown, but the surviving parasites were able to form a new tegument as shown by detection of SGTP4 on the parasite surface. Furthermore, we found that complement activation significantly damaged the acetabular gland ducts and lysed secretory vesicles released by transforming Cercariae. These findings should contribute for future in vivo studies of the effects of the complement system in skin migrating Cercariae.
