Preventive Cobalt Therapy in heterotopic ossification consequent to prosthetic hip reimplantation.

Preventive treatment may be carried out with radiation or drug Therapy. The authors report the results of 54 cases submitted to prosthetic reimplantation and treated by Cobalt Therapy, as compared to the results of 76 reimplantations (control group) that did not undergo any kind of treatment: the occurrence of high grade ossification (exceeding Brooker grade III) was 2% in cases treated, as compared to 9% in those not treated. Males were more at risk for the development of ossification.

The authors report the results obtained in 96 cases treated by Cobalt Therapy: the occurrence of high grade ossification (Brooker III and IV) is equal to 1% in cases treated, as compared to 9% in cases not treated. Male patients of advanced age and who are characterized by primary arthrosis are more at risk for ossification.

Toni, Brizio L, Armando GiuntiAbstract:The authors report 7 cases in which removal of high grade ossification which developed after hip arthroplasty was performed: all of the patients were treated postoperatively by radiation Therapy with the purpose of preventing recurrence of ossification. After surgery ossification was present, but to a moderate degree, in 6 out of 7 cases examined; movement had clearly improved. The results obtained show that removal surgery is justified if movement of the hip is considerably impaired (ankylosis) and that prevention plays an essential role and must be carried out every time that the patient risks the development of ossification.

Effects of supplementation with a combination of Cobalt and ascorbic acid on antioxidant enzymes and lipid peroxidation levels in streptozocin-diabetic rat liver
Biological Trace Element Research, 2002Co-Authors: Özlem Yildirim, Zeliha BüyükbingölAbstract:The effect of Cobalt(II) chloride (CoCl_2) and CoCl_2 with ascorbic acid (AA) on components of the antioxidant defense system and lipid oxidative damage were studied in controls and streptozotocin-induced diabetic rat livers. Three days after injection, rats received either 0.5 m M CoCl_2 or 0.5 m M CoCl_2 with a combination of 1 g/L AA in drinking water up to 6 wk. The elevated blood glucose levels in diabetic rats were about 12% restored by oral administration of CoCl_2 (0.05 m M ) and were significant reduced (46%) following AA addition (1 g/L) to CoCl_2. Cobalt Therapy effectively decreased the increased activities of catalase (CAT), superoxide dismutase (SOD), and thiobarbituric acid reactant substances (TBARS) but could not restore the increased glutathione peroxidase (GSH-Px) in the liver of diabetic rats. Our findings suggest that Cobalt Therapy may prove effective in improving the impaired antioxidant status during the early state of diabetes, and ascorbic acid supplementation at this dose potentiates the effectiveness of Cobalt action.

Antioxidant Defense System in Cobalt Treated Diabetic Rat Lung
Biotechnology & Biotechnological Equipment, 2002Co-Authors: Özlem YildirimAbstract:ABSTRACTTo study the effect of Cobalt intake on catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) activities and the levels of lipid peroxidation by measuring thiobarbituric acid reactive substance (TBARS), streptozotocin (STZ)-induced diabetic rats were treated with Cobalt chloride (CoC12)(0.5 mM in the drinking water) for up to 44 days. Diabetes was associated with a significant increase in the activity of CAT, SOD, GSH-Px and TBARS in lung. Cobalt Therapy effectively normalized the increased activities of CAT, SOD and TBARS levels but could not restore GSH-Px in the lung. The ensemble of results suggests that, the Cobalt dose which was used in this study can be considered as the regulation of oxidative and antioxidative systems.

We observed increased kidney oxygen consumption (QO2) and altered renal hemodynamics in the A/I kidney that were normalized after combined angiotensin II (ANG II) blockade. Studies suggest hypoxia inducible factor as a protective influence in A/I. We induced hypoxia-inducible factor (HIF) and HIF target proteins by two different methods, Cobalt chloride (CoCl2) and dimethyloxalyglycine (DMOG), for the first week after creation of A/I and compared the metabolic and renal hemodynamic outcomes to combined ANG II blockade. We also examined the HIF target proteins expressed by using Western blots and real-time PCR. Treatment with DMOG, CoCl2, and ANG II blockade normalized kidney oxygen consumption factored by Na reabsorption and increased both renal blood flow and glomerular filtration rate. At 1 wk, CoCl2 and DMOG increased kidney expression of HIF by Western blot. In the untreated A/I kidney, VEGF, heme oxygenase-1, and GLUT1 were all modestly increased. Both ANG II blockade and CoCl2 Therapy increased VEGF and GLUT1 but the Cobalt markedly so. ANG II blockade decreased heme oxygenase-1 expression while CoCl2 increased it. By real-time PCR, erythropoietin and GLUT1 were only increased by CoCl2 Therapy. Cell proliferation was modestly increased by ANG II blockade but markedly after Cobalt Therapy. Metabolic and hemodynamic abnormalities were corrected equally by ANG II blockade and HIF therapies. However, the molecular patterns differed significantly between ANG II blockade and Cobalt Therapy. HIF induction may prove to be protective in this model of CKD.
