    Abstract:

    Plasmodium dalam eritrosit akan menginduksi respons imun, berupa produksi radikal bebas yang dapat menyebabkan kerusakan jaringan paru. Kombinasi ekstrak Anamirta Cocculus dan artemisin diharapkan dapat mencegah komplikasi akibat radikal bebas yang dihasilkan oleh sel imun maupun artemisin. Studi eksperimental dengan metode post test control group design only dilakukan di Laboratorium Parasitologi dan Biomedik Fakultas Kedokteran Universitas Brawijaya periode Juni–Oktober 2010 untuk membuktikan efek pemberian kombinasi ekstrak A. Cocculus dan artemisin terhadap jumlah sel apoptosis jaringan paru mencit galur Balb/C yang diinfeksi Plasmodium berghei. Mencit dibagi dalam 6 kelompok, kelompok 1 terdiri atas 9 ekor mencit normal, kelompok 2 mencit yang diinfeksi P. berghei tanpa terapi, kelompok 3 mencit yang diinfeksi P. berghei dan diterapi artemisin dosis 0,04 mg/gBB, serta 3 kelompok perlakuan mencit diinfeksi P. berghei dan diterapi kombinasi artemisin 0,04 mg/gBB dengan ekstrak A. Cocculus 0,01 mg/gBB; 0,1 mg/gBB; dan 1 mg/gBB. Sel apoptosis dihitung dari ekspresi caspase-3 pada pewarnaan imunohistokimia. Pemberian ekstrak A. Cocculus dosis 0,01 mg/gBB; 0,1 mg/gBB; dan 1 mg/gBB serta artemisin selama 3 hari menurunkan jumlah sel apoptosis secara signifikan dibandingkan dengan kelompok artemisin monoterapi (p=0,00; 0,026; 0,000). Hubungan lama terapi dengan ekspresi caspase-3 menunjukkan pada kelompok terapi kombinasi ekstrak A. Cocculus 0,01 mg/gBB dan artemisin 0,04 mg/gBB memiliki korelasi positif yang signifikan (p=0,013). Simpulan, terapi jangka pendek kombinasi ekstrak A. Cocculus dan artemisin mempunyai efek yang lebih baik dibandingkan dengan pemberian artemisin monoterapi dalam hal penurunan jumlah sel apoptosis jaringan paru.     Abstract:

    The purpose of this study was to evaluate the anti-inflammatory activity of the ethanolic fruit extract of Anamirta Cocculus in experimental acute and chronic inflammatory models. Ethanolic extract of fruit of Anamirta Cocculus was prepared and tested for anti-inflammatory activity in Albino Wister rats weighing 200-250 grams. The animals were randomly divided into three groups of 6 each; one group served as control and other two groups received diclofenac and ethanolic extract of Anamirta Cocculus orally 1 hr prior to experimentation. The in-vivo anti-inflammatory activity was studied using the acute (Carrageenan induced paw edema, Histamine induced paw edema) and chronic (Cotton pellet induced granuloma) animal models. Anti-inflammatory activity was expressed as Percent inhibition (pI). Statistical analysis was performed using One-way analysis of variance (ANOVA) followed by Dunnett’s Multiple Comparison Test P<0.001 was considered statistically significant. The maximum PI with diclofenac and ethanolic extract of AC in Carrageenan induced paw edema were 60.56% and 58.83%, in histamine induced paw edema 58.63% and 60.27% and in cotton pellet induced granuloma were 53.11% and 36.08% respectively. Diclofenac showed highly significant activity in all the three models. However, ethanolic extract of AC showed highly significant activity in all the three models studied. As ethanolic extract of AC showed significant anti-inflammatory activity in the models studied, it can be a promising anti-inflammatory agent.

    Abstract:

    Objective: The fruits of Anamirta Cocculus Linn are used traditionally in India for the treatment of bronchitis, chronic skin diseases, foul ulcers, dermatophytosis, phthisis, fungal infections, vertigo and vitiated conditions of vata & kapha. The purpose of the present study was to evaluate the wound healing activity of the ethanolic extract of its fruit in excision wound model. Methods: Albino Wistar rats of either sex were divided into three groups, viz, wounded control, wounded rats administered standard drug Ciprofloxacin (2 %), and wounded rats administered A.C. 200 mg/kg, respectively. In excision wound model, wound contraction was studied. Results: Increased wound contraction was observed in the extract-treated group when compared to control. Wound healing activity was statistically significant (p < 0.05) & (p < 0.001) in animals treated with 200 mg/kg of the extract. Conclusion: The ethanolic extract of A.     Abstract:

    Anamirta Cocculus (Linn.) belongs to the family Menispermaceae is commonly known as fish berry or crow killer, also known as Garalaphala in Sanskrit and kakamari in Hindi. The plant is well known for producing poisonous seeds. Anamirta Cocculus is found in Southeast Asian and Indian subcontinent. This wild medicine is being ex- ploited by human beings for several purposes including medication, hunting and fishing. In traditional Ayurvedic system of medicine in India, Anamirta Cocculus (Linn.) has been used as a (Rasayan) drug in vitiated condition of vata and kapha. This plant is also used in the treatment of bronchitis, chronic skin diseases, foul ulcers, dermato- phytosis, phthisis, inflammation, vertigo, flatulence, nocturnal epilepsy, chorea, paralysis of pharynx, leg and res- piratory center etc. It has also been suggested to possess anti inflammatory, antifungal, anthelmintic antioxidant, antimicrobial, insecticidal, germicidal, CNS actions, a nervine tonic and a best antidote to morphine poisoning. The Phytochemical screening of this plant has shown the presence of alkaloids, steroids, phenolic terpenoids, fixed oils, carbohydrates and proteins. The active constituents like picrotoxinin, picrotin, menispermine, paramenisper- mine etc., present in Anamirta Cocculus (Linn.) have been found to be responsible for the therapeutic potentials. Isolation of anticancer constituents from this plant was also reported newly and moreover, the most useful and immense medicinal values that require more exploration in all the pharmaceutical aspect.

    Seeds of the phytopiscicide Anamirta Cocculus are used recently to eradicate unwanted fishes from aquaculture ponds during pond preparation. However, the mode of action of A. Cocculus and the biochemical responses caused to exposed fishes largely remain unexplained. The present study attempts to assess the impact of 24 h LC50 (18.79 ppm) of the ethanolic extract of A. Cocculus seeds on the tissue damage biomarkers viz., alkaline phosphatase (ALP), acid phosphatase (ACP), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) of the predatory wild fish Heteropneustes fossilis. The tissues assayed were brain, gills, liver, kidney, muscle, accessory respiratory organs (ARO) and blood. The exposure has caused varying levels of significant reductions in the activities of all these enzymes in various tissues except blood. The activities of serum phosphatases and aminotransferases became significantly higher in the later stages of exposure. The observations clearly indicate the tissue-damaging effects of A. Cocculus seed extract

    Abstract The present work is aimed at primary screening of the phytochemical contents of seven solvents (petroleum ether, benzene, chloroform, acetone, methanol, ethanol and a mixture of methanol and ethyl acetate (1:3)) extracts of the seeds of Anamirta Cocculus (Linn.) and to evaluate the antibacterial activity of these extracts against five species of pathogenic bacteria. The powdered seeds of A. Cocculus were extracted with seven solvents (polar and non-polar) with Soxhlet apparatus and the extracts were subjected to preliminary phytochemical screening by standard biochemical tests. The antibacterial activity of various extracts was evaluated against five bacterial species ( Staphylococcus aureus , Proteus vulgaris , Escherichia coli , Salmonella typhi and Klebsiella pneumoniae ). The zone of inhibitions and minimum inhibitory concentration (MIC) of the various extracts were determined by running the experiments in triplicates. The extracts of the seeds of A. Cocculus contained various pharmaceutically active substances viz ., aldehydes, alkaloids, phenolic compounds, flavonoids, saponins, carbohydrates, proteins, lipids, glycosides, phytosterols, volatile oils, gums and mucilage and other minor phytochemicals. All the extracts showed significant levels of antibacterial activity. Methanol extract was the most active one with remarkable antibacterial activity on the various species tested. MICs of the extracts revealed methanol extract as the most potent one with the lowest inhibitory concentration of 3 μg/mL on E. coli . The findings of the present study indicated that the seeds of A. Cocculus possess various secondary metabolites having the potential for developing pharmaceutical drugs, especially antimicrobial ones.

    Seeds of the Indian fish berry Anamirta Cocculus (Linn.) are a potential piscicidal agent used for catching fish from the wild by native people. In the present study, laboratory evaluation of the acute toxicity of the seeds of A. Cocculus was done at various time intervals on three species of freshwater predatory fishes viz., Clarias batrachus (Linn.), Channa striatus (Bloch.) and Mystus vitattus (Bloch.). The piscicidal agent was administered as a stomach poison in two different modes viz., after deepfrying and without heating. The LC50(s) and LC99(s) at different durations were calculated in both the modes of administration on all the three species tested. For C. batrachus, the LC50(s) at 1, 2, 24, 48, 72 and 96 hours (h) of piscicidal administration in the heated mode was 203.8463, 170.5797, 137.785, 100.5709, 85.4432 and 62.7660 mg/kg body weight and in the without heated mode 167.2705, 101.2334, 68.4538, 63.9773, 51.6782 and 50.2421 mg/kg body weight respectively. For C. striatus, the LC50(s) at 1, 2, 24, 48, 72 and 96 h in the heated mode was 109.2334, 77.4538, 57.9773, 44.2705, 34.6782 and 24.2421 mg/kg body weight and in the without heated mode 90.7660, 55.4433, 34.5709, 28.7685, 20.5798 and 15.3158 mg/kg body weight respectively. For M. vitattus the LC50(s) at the corresponding durations in the heated mode was 32.2706, 22.4433, 19.7686, 15.2883, 7.2517 and 3.4484 mg/kg body weight and in the without heated mode 26.8463, 17.2705, 10.0193, 7.9773, 4.9850 and 1.9082 mg/kg body weight respectively. LC99(s) in both the modes of administration in the respective exposure periods for all the three species tested were also calculated. The results reveals that of all the species tested, C. batrachus is the most resistant one towards the toxicity of A. Cocculus followed by C. striatus and M. vitattus. The present study has also shown that these seeds may be used as a potent aquaculture management tool to eradicate unwanted wild fish from culture ponds before stocking.

Studi eksperimental dengan metode post test control group design only dilakukan di Laboratorium Parasitologi dan Biomedik Fakultas Kedokteran Universitas Brawijaya periode Juni–Oktober 2010 untuk membuktikan efek pemberian kombinasi ekstrak A. Cocculus dan artemisin terhadap jumlah sel apoptosis jaringan paru mencit galur Balb/C yang diinfeksi Plasmodium berghei. Mencit dibagi dalam 6 kelompok, kelompok 1 terdiri atas 9 ekor mencit normal, kelompok 2 mencit yang diinfeksi P. berghei tanpa terapi, kelompok 3 mencit yang diinfeksi P. berghei dan diterapi artemisin dosis 0,04 mg/gBB, serta 3 kelompok perlakuan mencit diinfeksi P. berghei dan diterapi kombinasi artemisin 0,04 mg/gBB dengan ekstrak A. Cocculus 0,01 mg/gBB; 0,1 mg/gBB; dan 1 mg/gBB. Sel apoptosis dihitung dari ekspresi caspase-3 pada pewarnaan imunohistokimia. Pemberian ekstrak A. Cocculus dosis 0,01 mg/gBB; 0,1 mg/gBB; dan 1 mg/gBB serta artemisin selama 3 hari menurunkan jumlah sel apoptosis secara signifikan dibandingkan dengan kelompok artemisin monoterapi (p=0,00; 0,026; 0,000). Hubungan lama terapi dengan ekspresi caspase-3 menunjukkan pada kelompok terapi kombinasi ekstrak A. Cocculus 0,01 mg/gBB dan artemisin 0,04 mg/gBB memiliki korelasi positif yang signifikan (p=0,013). Simpulan, terapi jangka pendek kombinasi ekstrak A. Cocculus dan artemisin mempunyai efek yang lebih baik dibandingkan dengan pemberian artemisin monoterapi dalam hal penurunan jumlah sel apoptosis jaringan paru. Mice were divided into 6 groups, the first normal Balb/C mice, the second mice infected by P. berghei without treatment, the third mice infected by P. berghei and treated with artemisin 0.04 mg/g BW, and 3 combination contains mice infected by P. berghei and treated with a combination of artemisin 0.04 mg/ gBW and A.Cocculus extract 0.01 mg/gBW, 0.1 mg/gBW, 1 mg/ gBW respectively. The apoptotic cells of lung tissue were counted from the expression of caspase-3 in immunohistochemical staining. In day 3 combination A. Cocculus extract dose 0.01 mg/gBW, 0.1 mg/gBW, 1 mg/ gBW and artemisin reduced the number of apoptotic cells significantly compared to the artemisin monotherapy (p=0.00, 0.026, 0.000). There was a positive corelation between the length of treatment and the expression of caspase-3 (p=0.013) on group that treated with combination of A. Cocculus extract 0.01 mg/gBW and artemisin 0,04 mg/gBB. In conclusion, the short treatment of combination A. Cocculus extract and artemisin has a better effect than artemisin monotherapy in decreasing number of apoptotic lung tissue cells.
