    Abstract We assessed effects of tissue collection methods (i.e., patagial microbiopsy and down feathers) and chick age at sampling on morphometrics and 21-day survival of 600 captive neonatal northern bobwhite (Colinus virginianus). We observed minimal effects on morphometrics and no difference in survival among patagial microbiopsy (x ¯ = 0.96 ± 0.03), down feathers (x ¯ = 0.92 ± 0.04), and control (x ¯ = 0.86 ± 0.05) methods. DNA analysis from patagial microbiopsy, down feather, and egg tooth samples showed greater concentrations of DNA from patagial microbiopsy (x ¯ = 10.28 ± 1.74 µg/ml) than either down feather (x ¯ = 4.10 ± 1.74 µg/ml) or egg teeth (x ¯ = 2.35 ± 1.74 µg/ml).

    Abstract:

    Ecological studies using microsatellite data often require the selection of an optimal marker set for use in parentage and relatedness inference. Commonly, this requires a candidate pool of microsatellite markers from which several are selected to ensure data are acquired efficiently and accurately. We developed 10 microsatellite loci for use with Northern Bobwhite (Colinus virginianus) and tested these loci using individuals collected from two distinct populations in GA and VA. Our new markers yielded seven alleles/locus (range: 2–16) in the Georgia population and six alleles/locus (range: 2–13) in the Virginia population. Exclusionary power of all markers in each population with both parents unknown was >0.98. These microsatellite loci should be combined with previously developed markers to select an optimal set for use in subsequent analyses of parentage and relatedness.

    Abstract:

    We describe polymerase chain reaction (PCR) primers and conditions to amplify eight dinucleotide, one trinucleotide and 14 tetranucleotide microsatellite DNA loci isolated from the northern bobwhite ( Colinus virginianus ). The PCR primers were tested on 16 individuals collected from a population located within the Red Hills region of south Georgia and north Florida. The 23 primer pairs developed in this study yielded an average of 6.5 alleles per locus (range 2–11), an average observed heterozygosity of 0.47 (range 0.06–0.94) and average polymorphic information content of 0.60 (range 0.06–0.85).

    The eyes, breast muscle, and internal organs of 128 hunter-shot northern bobwhites (Colinus virginianus) from the Rolling Plains ecoregion of Texas, U.S.A., were examined for helminths. Six species of nematodes, 1 acanthocephalan, and 1 cestode were found in 120 bobwhites, representing 18,205 helminth individuals. Aulonocephalus pennula was the most common (91% prevalence) and numerically abundant (91% of all individuals) species, followed by Oxyspirura petrowi (66% prevalence and 7% of all individuals).     Abstract:  Mesocestoides spp. have a cosmopolitan distribution with zoonotic potential. Mesocestoides tetrathyridia were found under the pericardial sac, on the surface of the crop, and in the peritoneal cavity of a hunter-harvested Northern Bobwhite (Colinus virginianus) and a Scaled Quail (Callipepla squamata) collected during the 2016–17 quail hunting season in northwest and southern Texas, US respectively. Molecular analysis indicated that the tetrathyridia from the birds likely belonged to an undescribed species and are identical to pretetrathyridium stages recently found in Scincella lateralis skinks in Oklahoma. This is the first report of Mesocestoides from North American birds.

    Abstract:

    Northern Bobwhites (Colinus virginianus) have been in decline throughout the southeastern US. Prevalence of Trichomonas gallinae in wild bobwhites is unknown, although T. gallinae caused morbidity and mortality in experimentally infected bobwhites. Many species of Columbidae (pigeons and doves) in Texas are hosts to T. gallinae. Bobwhites potentially may become exposed to this protozoan through supplemental feed or water sources contaminated by columbids infected with T. gallinae. All of 506 bobwhites collected in Oklahoma and Texas, 2011-13, were PCR negative for T. gallinae. These data suggest T. gallinae is not contributing to the population decline of bobwhites in this region.
